Cascade reactions appeared as ac utting-edge strategy to streamline the assembly of complex structural scaffolds from naturally available precursors in an atom-, as well as time,l abor-a nd cost-efficient way. We hereinr eport astrategy to control cationic cyclization cascades by exploiting the ability of anchoring dynamic substrates in the active site of terpene cyclases via designed hydrogen bonding.Thereby, it is possible to induce "directed" cyclizations in contrast to established "non-stop" cyclizations (99:1) and predestinate cascade termination at otherwise catalytically barely accessible intermediates.A saresult, we are able to provide efficient access to naturally widely occurring apocarotenoids,v alueadded flavors and fragrances in gram-scale by replacing multistage synthetic routes to as ingle step with unprecedented selectivity (> 99.5 %ee) and high yields (up to 89 %).
Designed hydrogen bonds …… in the active site of terpene cyclases enable the control of cationic cyclization cascades by molecular anchoring of a substrates polar functional group. As a result, it is possible to replace multi-step synthetic routes by one enzymatic step and manufacture valuable apocarotenoids in a highly product-and stereoselective manner on gram scale, as reported by Bernhard Hauer and co-workers in their Research Article on page 13251.

In dieser Arbeit berichten wir über eine Strategie zur Kontrolle von kationischen Cyclisierungskaskaden, indem wir die Fähigkeit zur Verankerung von dynamischen Substraten im aktiven Zentrum von Terpencyclasen über designte Wasserstoffbrückenbindungen ausnutzen. Dadurch ist es mçglich,"gerichtete" Cyclisierungen im Gegensatz zu bisher etablierten "Non-Stop"-Cyclisierungen (99:1) zu induzieren und die Termination der Kaskade an ansonsten katalytischk aum zugänglichen Intermediaten zu prädestinieren. EinleitungVond er Natur abgeleitet, haben sich organokatalytische Kaskadenreaktionen als elegante Methode herauskristallisiert, um die klassische Step-by-Step-Synthesechemie zu ersetzen.
