ABSTRACT:The aim of this study was to establish the morphological basis for the classification of major cassava cultivars (Manihot esculenta Crantz) planted in Vietnam. Twenty typical descriptors were observed in various specific growth stages. Four traits related to mature leaves and two traits related to leaf veins were detected in cassava plants after the 4-month stage. At the same time, 12 root-and stem-related descriptors were collected in cassava plants from 8 months old to harvest. We found that two traits in the apical leaves can be observed in every growth stage. A classification tree based on these traits was constructed with 6 essential morphological descriptors to recognize the 7 most common cassava cultivars in Vietnam. KM 140 variety is distinguished by purplish-red in apical leaves, while KM 98-7 can be classified with straight or linear shapes of central leaflet. The irregular orientation of the petiole is a unique descriptor of local cultivar XVP. In the case of SM 937-26, weak foliar scars are observed. Finally, KM 94, KM 419, and KM 98-5 varieties can be classified by the color of the stem epidermis. These morphological descriptors will be a foundation to classify all cassava varieties planted in Vietnam.
H5N1 (Avian influenza virus) has caused economic loss and remained a serious human health risk in many countries, including Vietnam. Plant-derived vaccines have offered the advantages over tradition subunit vaccine with low-cost production potential. Duckweed (Sprirodela polyrhiza L.) has found a valuable target plant for various application. In this study, we have attempted to re-generate and transform the gene encoding the hemagglutinin antigen of H5N1 virus in frond of S. polyrhiza, a widely great duckweed grown in Vietnam. The results have shown that the culture medium for S. polyrhiza duckweed is Hutner medium with reduced mineral concentration ½ (H/2) supplemented with 10g/l sucrose at pH 6. This medium showed appropriate for the best growth and development of duckweed. Results of PCR analysis of S. polyrhiza lines from 14 transformation experiments that has obtained 4 duckweed lines carrying HA1 gene. This prelinary results will be provide the basis for transgenic research in the dukweed species and be useful for further generate the producing enable vaccine via the transgenic plants

Sự ôxi hóa Met trên phân tử protein gây ra bởi gốc oxy hóa tự do có thể gây rối loạn quá trình trao đổi chất, dẫn truyền tín hiệu, làm suy yếu hoặc gây chết tế bào. Trong nghiên cứu này, chúng tôi đã tìm được 121 protein giàu Met (AtMRP) từ cơ sở dữ liệu genome của cây Arabidopsis thaliana, trong đó, khoảng 50% số gen là chưa được xác định chức năng. Dựa trên các nghiên cứu trước đây về biểu hiện của gen trong điều kiện bất lợi, chúng tôi đã xác định được 23 và 16 gen AtMRP có mức độ phiên mã tăng và giảm tương ứng trong điều kiện hạn, 11 và 17 gen có mức biểu hiện tăng và giảm khi xử lý mặn. Trong số các gen này, có 10 đáp ứng gen phiên mã với cả hạn và mặn. Gen AT4G33467 đáp ứng mạnh nhất và có độ biểu hiện trong điều kiện hạn cao gấp 337,5 lần so với ở điều kiện thường. Gen bị kìm hãm phiên mã mạnh nhất là AT3G55240, giảm tương ứng 60,3 và 26,9 lần khi xử lý hạn và mặn. Cây Arabidopsis RBC1 siêu biểu hiện gen AT3G55240 mẫn cảm hơn so với cây kiểu dại trong điều kiện mặn (NaCl 175 mM). Đây là những bằng chứng đầu tiên và quan trọng về vai trò của gen mã hóa MRP trong đáp ứng với điều kiện bất lợi ở thực vật, mở ra hướng nghiên cứu ứng dụng các gen này trong chọn tạo giống bằng công nghệ sinh học.Từ khóa: Arabidopsis thaliana, độ mặn, khô hạn, methionine, protein giàu methionine. MỞ ĐẦUThực vật là nguồn cung cấp protein chủ yếu cho toàn bộ sinh giới. Protein thực vật được cấu thành từ hơn 20 loại axit amin, trong đó quan trọng nhất là một số axit amin không thể thay thế. Methionine (Met) là một axit amin cần thiết, có vai trò quan trọng trong đời sống sinh giới nói chung, là tiền chất tạo ra một số hợp chất trao đổi thứ cấp thiết yếu trong tế bào. Mã hóa cho bộ ba mở đầu AUG, Met là tín hiệu bắt đầu cho quá trình sinh tổng hợp chuỗi polypeptide. Met điều hòa trao đổi chất thông qua dẫn xuất S-Adenosyl Methionine (SAM), là phân tử cung cấp nhóm methyl (-CH 3 ) cho các phản ứng diễn ra trong tế bào. SAM cũng tham gia vào chu trình sinh tổng hợp ethylene [36], vitamin B 1 và một số hợp chất liên quan đến quá trình tăng trưởng và biệt hóa tế bào, apotosis, cân bằng nội môi như polyamine, spermidine, spermine [13,19]. Tuy là một axit amin thiết yếu, nhưng hàm lượng Met ở một số cây trồng quan trọng lại rất thấp, điển hình như ở khoai tây (Solanum tuberosum) có hàm lượng Met đạt 30 mg/100 gam, trong khi lúa (Oryza sativa), lúa mỳ (Triticum spp.), ngô (Zea mays), và đậu tương (Glycine max) cũng chỉ chiếm tương ứng là 170 -220 -190 -580 mg trên mỗi 100 gam [28]. Starch metabolism is known to be an important pathway in the growth and development of plants. This study was conducted to investigate the genome-wide identification and structural analysis of genes encoding uridine diphosphate glucose pyrophosphorylase (UGPase), a key enzyme in starch synthesis in cassava, and to analyze the expression profiles of these genes based on publicly available RNA-seq data. A total of 11 members were found in the UGPase gene family (MeUGP) in cassava. Ten of the MeUGP genes were successfully mapped onto the chromosomes of the current cassava genome assembly. Based on their nucleotide sequences, the lengths of the genomic DNA sequences of the MeUGP genes ranged from 3,200 to 11,601bp, while the size of the coding sequence (CDS) varied from 831 to 3,654bp. According to the recent RNA-seq data, we found that a majority of the MeUGP genes were expressed in at least 1 tissue under normal conditions. Interestingly, MeUGP4 was greatly expressed in the shoot apical meristem, while MeUGP10 was more specific in the root apical meristem. The expression profiles of these MeUGP genes should be carried out in various conditions in further studies.
Nuclear factor-Y (NF-Y) transcription factor can be found in most of plant species and is known to play important role in plant growth, development and stress response. In this study, 17 genes encoding NF-YB subunit were identified in cassava (Manihot esculenta Crantz). These genes were located in chromosomes with an uneven ratio. Analysis of gene structure indicated that MeNF-YB genes were varied in length and exon/intron organization. Genes in the same clade shared similar exon/intron structure. Based on available microarray database, most of NF-YB genes were up-regulated in at least 1 major tissue. Interestingly, MeNF-YB12 was highly expressed in shoot apical meristem, while MeNF-YB2 was specifically accumulated in friable embryogenesis callus. Additionally, some genes, such as MeNF-YB4/8/13/5 were found to be strongly expressed in friable embryogenesis callus. Our results strongly indicated that NF-YB genes in cassava might be associated with various growth and developmental processes in meristem tissues.
