Our previous study demonstrated that angiogenesis increased during the recovery of heat-denatured endothelial cells. However, the mechanism is still unclear. This study aimed to investigate the relation of autophagy and angiogenesis during the recovery of heat-denatured endothelial cells. A rat deep partial-thickness burn model and heat-denatured human umbilical vein endothelial cells (HUVECs) model (52 °C for 35 s) were used. Autophagy increased significantly in the dermis and HUVECs in a time-dependent manner after heat denaturation and recovery for 2–5 days. Rapamycin-mediated autophagy enhanced the pro-angiogenic effect, evidenced by increased proliferation and migration of HUVECs, and formation of tube-like structures. Autophagy inhibition by 3-Methyladenine (3-MA) abolished the angiogenesis in heat-denatured HUVECs after recovery for 3–5 days. Moreover, heat denaturation augmented the phosphorylation of AMP-activated protein kinase (AMPK) but reduced the phosphorylation of Akt and mTOR in HUVECs. Furthermore, autophagy inhibition by antioxidant NAC, compound C or AMPK siRNA impaired cell proliferation, migration and tube formation heat-denatured HUVECs. At last, the in vivo experiments also showed that inhibition of autophagy by bafilomycin A1 could suppress angiogenesis and recovery of heat-denatured dermis.Taken together, we firstly revealed that autophagy promotes angiogenesis via AMPK/Akt/mTOR signaling during the recovery of heat-denatured endothelial cells and may provide a potential therapeutic target for the recovery of heat-denatured dermis.

We have previously shown that heat shock protein 70 (HSP70) markedly inhibits H 2 O 2 -induced apoptosis in mouse C2C12 myogenic cells by reducing the release of Smac. However, the molecular mechanism by which HSP70 interferes with Smac release during oxidative stress-induced apoptosis is not understood. In the current study, we showed that HSP70 increased the stability of Bcl-2 during oxidative stress. An antisense phosphorothioate oligonucleotide against Bcl-2 caused selective inhibition of Bcl-2 protein expression induced by HSP70 and significantly attenuated HSP70-mediated cell protection against H 2 O 2 -induced release of Smac and apoptosis. Taken together, our results indicate that there are important relationships among HSP70, Bcl-2, release of Smac, and induction of apoptosis by oxidative stress.
Although heat shock protein 70 (Hsp70) has been shown to markedly inhibit H2O2‐induced apoptosis in C2C12 cells, and nucleolin/C23 has also been implicated in apoptosis, the relationship of these two molecules is still largely unknown. The aim of the current study was to investigate the potential involvement of nucleolin/C23 in the antiapoptotic mechanism of Hsp70. We found that primary cultures of neonatal rat cardiomyocytes underwent apoptosis upon H2O2 treatment, and in these cells nucleolin/C23 protein was highly unstable and had a half‐life of less than 4 h. However, transfection with Hsp70 greatly stabilized nucleolin/C23 and also protected the cells from H2O2‐induced apoptosis. When nucleolin/C23 was knocked down with an antisense oligomer, H2O2‐induced apoptosis became more severe, even in Hsp70‐overexpressed cells, demonstrating an essential role of nucleolin/C23 in the antiapoptotic effects of Hsp70. Similar results were obtained by both nuclear morphology observation and caspase‐3 activity assay. Therefore, these data provide evidence that nucleolin/C23 is an essential downstream effecter of Hsp70 in the protection of cardiomyocytes against oxidative stress‐induced apoptosis.
Abstract. To control severe air pollution in China, comprehensive pollution control strategies have been implemented throughout the country in recent years. To evaluate the effectiveness of these strategies, the influence of meteorological conditions on levels of air pollution needs to be determined. Using the intensive air pollution control strategies implemented during the Asia-Pacific Economic Cooperation Forum in 2014 (APEC 2014) and the 2015 China Victory Day Parade (Victory Parade 2015) as examples, we estimated the role of meteorological conditions and pollution control strategies in reducing air pollution levels in Beijing. Atmospheric particulate matter of aerodynamic diameter ≤ 2.5 µm (PM 2.5 ) samples were collected and gaseous pollutants (SO 2 , NO, NO x , and O 3 ) were measured online at a site in Peking University (PKU). To determine the influence of meteorological conditions on the levels of air pollution, we first compared the air pollutant concentrations during days with stable meteorological conditions. However, there were few days with stable meteorological conditions during the Victory Parade. As such, we were unable to estimate the level of emission reduction efforts during this period. Finally, a generalized linear regression model (GLM) based only on meteorological parameters was built to predict air pollutant concentrations, which could explain more than 70 % of the variation in air pollutant concentration levels, after incorporating the nonlinear relationships between certain meteorological parameters and the concentrations of air pollutants. Evaluation of the GLM performance revealed that the GLM, even based only on meteorological parameters, could be satisfactory to estimate the contribution of meteorological conditions in reducing air pollution and, hence, the contribution of control strategies in reducing air pollution. Using the GLM, we found that the meteorological conditions and pollution control strategies contributed 30 and 28 % to the reduction of the PM 2.5 concentration during APEC and 38 and 25 % during the Victory Parade, respectively, based on the assumption that the concentrations of air pollutants are only determined by meteorological conditions and emission intensities. We also estimated the contribution of meteorological conditions and control strategies in reducing the concentrations of gaseous pollutants and PM 2.5 components with the GLMs, revealing the effective control of anthropogenic emissions.
