Chromosomal double-strand breaks (DSBs) in mammalian cells are repaired by either homology-directed repair (HDR), using a homologous sequence as a repair template, or nonhomologous end-joining (NHEJ), which often involves sequence alterations at the DSB site. To characterize the interrelationship of these two pathways, we analyzed HDR of a DSB in cells deficient for NHEJ components. We find that the HDR frequency is enhanced in Ku70 Double-strand breaks (DSBs) are potentially catastrophic lesions that if not repaired will lead to loss of genetic information and mutagenesis or cell death. In mammalian cells, two major pathways exist to repair DSBshomologous recombination and nonhomologous endjoining (NHEJ;Liang et al. 1998). NHEJ, the rejoining of DNA ends with the use of little or no sequence homology, involves the processing of ends such that nucleotides are often deleted or inserted at the break site prior to ligation (Jeggo 1998). Such modifications are likely central to the ability of mammalian cells to rejoin DNA ends with a variety of structures. Homology-directed repair (HDR) of a DSB, in contrast, requires significant lengths of sequence homology so that a DNA end from one molecule can invade a homologous sequence and prime repair synthesis (Pâques and Haber 1999). Processing of DNA ends also occurs with HDR; however, repair is typically precise, because a homologous sequence templates the repair event.Several processes exist in which repair of a DSB is restricted to either NHEJ or HDR. For example, DSBs introduced by the RAG proteins to generate antigen receptor diversity during V(D)J rearrangement are repaired by the NHEJ pathway (Jeggo 1998), whereas those introduced during meiosis by the Spo11 protein are repaired by the HDR pathway (Keeney 2001). The restriction in type of DSB repair raises the question as to how pathway choice is regulated. Several studies point to cell cycle phase as one factor that modulates repair pathway choice. In chicken cells, HDR has been found to play a dominant role in repairing radiation-induced DSBs in late S/G 2 phase, whereas NHEJ is preferentially used during G 1 /early S phase (Takata et al. 1998). Consistent with this in mammalian cells, the preferred homologous template for HDR-the sister chromatid (Johnson and Jasin 2001)-is present only during the S/G 2 phase of the cell cycle. Despite a cell cycle preference, HDR and NHEJ can nevertheless be coupled for the repair of a single DSB in mammalian cells (Richardson and Jasin 2000), indicating that the two repair pathways are not completely restricted to different cell cycle phases and that other factors influence pathway choice.Based on in vitro studies of DNA end-binding proteins such as RAD52, it has been suggested that end-binding proteins may direct entry into alternative DSB repair pathways (Van Dyck et al. 1999). The PML gene of acute promyelocytic leukemia (APL) encodes a cell-growth and tumor suppressor. PML localizes to discrete nuclear bodies (NBs) that are disrupted in APL cells. The Bloom syndrome gene BLM encodes a RecQ DNA helicase, whose absence from the cell results in genomic instability epitomized by high levels of sister-chromatid exchange (SCE) and cancer predisposition. We show here that BLM colocalizes with PML to the NB. In cells from persons with Bloom syndrome the localization of PML is unperturbed, whereas in APL cells carrying the PML-RARa oncoprotein, both PML and BLM are delocalized from the NB into microspeckled nuclear regions. Treatment with retinoic acid (RA) induces the relocalization of both proteins to the NB. In primary PML7/7 cells, BLM fails to accumulate in the NB. Strikingly, in PML7/7 cells the frequency of SCEs is increased relative to PML+/+ cells. These data demonstrate that BLM is a constituent of the NB and that PML is required for its accumulation in these nuclear domains and for the normal function of BLM. Thus, our ®ndings suggest a role for BLM in APL pathogenesis and implicate the PML NB in the maintenance of genomic stability.

The Bloom syndrome gene, BLM, encodes a RecQ DNA helicase that when absent from the cell results in genomic instability and cancer predisposition. We show here that BLM is a substrate for small ubiquitin-like modifier (SUMO) modification, with lysines at K317, K331, K334 and K347 being preferred sites of modification. Unlike normal BLM, a double mutant BLM protein with lysine to arginine substitutions at residues 317 and 331 was not modified by SUMO, and it failed to localize efficiently to the PML nuclear bodies. Rather, double mutant BLM protein induced the formation of DNA damage-induced foci (DDI) that contained BRCA1 protein and phosphorylated histone H2AX. Double mutant BLM only partially complemented the genomic instability phenotypes of Bloom syndrome cells as assessed by sister-chromatid exchange and micronuclei formation assays. These results constitute evidence that BLM is a DNA damage sensor that signals the formation of DDI, and they establish SUMO modification as a negative regulator of BLM's signaling function.

Disruption of the gene encoding RAD51, the protein that catalyzes strand exchange during homologous recombination, leads to the accumulation of chromosome breaks and lethality in vertebrate cells. As RAD51 is implicated in BRCA1-and BRCA2-mediated tumor suppression as well as cellular viability, we have begun a functional analysis of a defined RAD51 mutation in mammalian cells. By using a dominant negative approach, we generated a mouse embryonic stem cell line that expresses an ATP hydrolysis-defective RAD51 protein, hRAD51-K133R, at comparable levels to the endogenous wild-type RAD51 protein, whose expression is retained in these cells. We found that these cells have increased sensitivity to the DNA-damaging agents mitomycin C and ionizing radiation and also exhibit a decreased rate of spontaneous sister-chromatid exchange. By using a reporter for the repair of a single chromosomal double-strand break, we also found that expression of the hRAD51-K133R protein specifically inhibits homology-directed double-strand break repair. Furthermore, expression of a BRC repeat from BRCA2, a peptide inhibitor of an early step necessary for strand exchange, exacerbates the inhibition of homology-directed repair in the hRAD51-K133R expressing cell line. Thus, ATP hydrolysis by RAD51 has a key role in various types of DNA repair in mammalian cells.The accurate repair of DNA damage, such as a chromosomal double-strand break (DSB), 1 is essential to prevent chromosomal alterations and the loss of genetic information.
