The antimicrobial activity and probiotic properties of Bacillus subtilis strain KKU213, isolated from local soil, were investigated. The cell-free supernatant (CFS) of a KKU213 culture containing crude bacteriocins exhibited inhibitory effects on Gram-positive bacteria, including Bacillus cereus, Listeria monocytogenes, Micrococcus luteus, and Staphylococcus aureus. The antibacterial activity of the CFS precipitated with 40% ammonium sulfate (AS) remained even after treatment at 60 and 100 °C, at pH 4 and 10 and with proteolytic enzymes, detergents and heavy metals. When analyzed by SDS-PAGE and overlaid with the indicator strains B. cereus and S. aureus, the 40% AS precipitate exhibited inhibitory activity on proteins smaller than 10 kDa. However, proteins larger than 25 kDa and smaller than 10 kDa were still observed on a native protein gel. Purified subtilosin A was prepared by Amberlite XAD-16 bead extraction and HPLC and analyzed by Nano-LC-QTOF-MS. Its molecular mass was found to be 3.4 kDa, and it retained its antibacterial activity. These results are consistent with the detection of the anti-listerial subtilosin A gene of the sbo/alb cluster in the KKU213 strain, which is 100% identical to that of B. subtilis subsp. subtilis 168. In addition to stable and cyclic subtilosin A, a mixture of many extracellular antibacterial peptides was also detected in the KKU213 culture. The KKU213 strain produced extracellular amylase, cellulase, lipase and protease, is highly acid-resistant (pH 2) when cultured in inulin and promotes health and reduces infection of intestinally colonized broiler chickens. Therefore, we propose that bacteriocin-producing B. subtilis KKU213 could be used as a potential probiotic strain or protective culture.
BackgroundOxidative stress, an excess of reactive oxygen species (ROS), causes lipid peroxidation resulting in cell and tissue damages. It may be associated with the development and progression of cancers in dogs. Malondialdehyde (MDA), the end product of lipid peroxidation, is commonly used as a marker of oxidative stress. The objective of this study was to assess oxidative stress in cancer-bearing dogs by measuring serum MDA levels. All client-owned dogs underwent physical examination at the Veterinary Teaching Hospital, Faculty of Veterinary Medicine, Khon Kaen University to determine the health status with the owner’s consent. Blood samples of cancer-bearing dogs (N = 80) and clinically normal dogs (N = 101) were obtained and subjected for determination of MDA levels. In addition, complete blood count, creatinine, and alanine aminotransferase were measured.ResultsSerum MDA was significantly higher in cancer-bearing dogs than in clinically normal dogs (mean ± SD, 4.68 ± 1.32 μmol/L vs 2.95 ± 0.61 μmol/L, respectively; p < 0.001). Packed cell volume (mean ± SD, 36.18 ± 7.65% vs 44.84 ± 5.54%), hemoglobin (mean ± SD, 11.93 ± 2.88 g% vs 15.17 ± 2.00 g%) and red blood cells (median (IQA), 6.05 (2.15) vs 8.09 (1.34)) were all significantly lower in cancer-bearing dogs than in clinically normal dogs. However, the reverse was true for white blood cells (median (IQA), 18.20 (11.95) vs 14.90 (5.10)). Neither creatinine nor alanine aminotransferase levels were significantly different between groups.ConclusionsThis study supports the conclusion that oxidative stress is associated with many types of cancers in dogs, as serum MDA levels were significantly higher in cancer-bearing dogs compared to clinically normal dogs.
Aim: The present study aimed to investigate the effects of different risk factors on stillbirth of piglets born from oxytocin-assisted parturitions. Materials and Methods: Data were collected from a total of 1121 piglets born from 74 Landrace x Yorkshire crossbred sows from a herd. Logistic regression models were used to determine the associations between stillbirth and different risk factors including parity (1, 2, 3-5, and 6-10), gestation length (GL) (112-113, 114-116, and 117-119 days), litter size, birth order (BO), sex, birth interval (BI), cumulative farrowing duration, birth weight (BW), crown rump length, BW deviation, body mass index, ponderal index (PI), and the use of oxytocin during expulsive stage of farrowing. Results: The incidence of stillbirth at litter level and stillbirth rate was 59.5% (44/74) and 8.1% (89/1094), respectively. The final multivariate logistic regression selected BO, BI, PI, GL, and parity as the five most significant risk factors for stillbirth. Increased BO and BI, GL <114 and >116 days, parity 6-10, and low PI increased the stillbirth rate in piglets. Conclusion: Several factors previously determined as risks for stillbirth in exogenous oxytocin-free parturitions also existed in exogenous oxytocin-assisted parturitions. One dose of oxytocin at fairly high BO did not increase stillbirth, whereas two doses of oxytocin were potentially associated with increased values.
Seasonal changes in bone mineral density are associated with changes in serum vitamin D concentrations in alpacas. Changes in bone mineral density associated with a decline in serum vitamin D concentration may predispose some alpacas to developing fractures minimal trauma.
Background and Aim: Stillbirth causes considerable loss to the pig farming industry. Methods aimed at reducing stillbirth should base on the understanding of risk factors for intrapartum stillbirth because it accounts for 75% of all stillbirths. Unfortunately, few studies have differentiated between intrapartum and prepartum stillbirths leading to inadequate information about risk factors for sole intrapartum stillbirth. This study investigated risk factors for piglet's intrapartum stillbirth. Materials and Methods: Data of 1527 piglets born from 103 sows in one herd were recorded. Generalized linear mixed models were used to determine the relationship between investigated risk factors and intrapartum stillbirth at the piglet level. The potential risk factors were parity, gestation length (GL), litter size (LS), birth order (BO), birth interval (BI), cumulative farrowing duration (CFD), gender, crown-rump length, birth weight (BW), body mass index, ponderal index (PI), and BW deviation. Results: About 60% (60.2%, 62/103) litters had stillborn piglet(s), and the intrapartum stillbirth rate was 5.8% (89/1527). BW deviation (≤0.1 and >0.6 kg), LS >13, GL (<114 and >117 days), PI ≤54, and BO >10 were the most significant factors associated with increased intrapartum stillbirth. No effect of parity, sex, BI, and CFD on intrapartum stillbirth was detected. Conclusion: These data stressed the importance of piglets' size and shape in the prediction of intrapartum stillbirth. Furthermore, large LS, high BO, short, and long GL were associated with increased intrapartum stillbirth. The results of this study suggest that procedures aimed at increasing litter homogeneity, optimizing piglets' size and shape, avoiding short and long gestation, and increasing supervision rate, especially at the second half, of the farrowing may reduce piglet's intrapartum stillbirth.
