Abiotic stress is one of the major limiting factors for plant development and productivity, which makes it important to identify microorganisms capable of increasing plant tolerance to stress. Dark septate endophytes can be symbionts of plants. In the present study, we evaluated the ability of dark septate endophytes isolates to reduce the effects of water stress in the rice varieties Nipponbare and Piauí. The experiments were performed under gnotobiotic conditions, and the water stress was induced with PEG. Four dark septate endophytes were isolated from the roots of wild rice (Oryza glumaepatula) collected from the Brazilian Amazon. Plant height as well as shoot and root fresh and dry matter were measured. Leaf protein concentrations and antioxidant enzyme activity were also estimated. The dark septate endophytes were grown in vitro in Petri dishes containing culture medium; they exhibited different levels of tolerance to salinity and water stress. The two rice varieties tested responded differently to inoculation with dark septate endophytes. Endophytes promoted rice plant growth both in the presence and in the absence of a water deficit. Decreased oxidative stress in plants in response to inoculation was observed in nearly all inoculated treatments, as indicated by the decrease in antioxidant enzyme activity. Dark septate endophytes fungi were shown to increase the tolerance of rice plants to stress caused by water deficiency.
RESUMO.A análise conjunta de variáveis qualitativas e quantitativas tem sido apontada como ferramenta útil na estimativa da divergência genética entre os acessos de uma coleção de germoplasma. O presente trabalho teve como objetivos caracterizar uma coleção de germoplasma de tomateiro do grupo cereja, com base em descritores qualitativos e quantitativos e utilizar o algoritmo de Gower na quantificação da divergência genética. Estudaram-se 40 acessos de tomateiro cereja, cultivados em manejo orgânico nas condições de Seropédica, Estado do Rio de Janeiro. Foram utilizados quatro descritores qualitativos relacionados aos frutos e nove descritores quantitativos. Os dados foram analisados de forma conjunta pelo algoritmo de Gower. Detectou-se variabilidade para coloração, formato e número de lóculos do fruto. A coloração vermelha foi observada em 25 acessos testados, registrando-se também a produção de frutos amarelos, laranjas e marrons. Para número de lóculos, foi registrada a ocorrência de frutos bi, tri, tetra e pluriloculares (com até sete lóculos). O método UPGMA foi o que obteve o maior coeficiente de correlação cofenética (0,80), observando-se a formação de sete grupos. Os grupos formados permitiram a distinção de frutos classificados como cereja e outros que não se adequaram a esta definição, por estarem acima do diâmetro equatorial proposto para esta classe.Palavras-chave: Solanum lycopersicum, análise multivariada, descritores qualitativos, descritores quantitativos, programa R.ABSTRACT. Using Gower's algorithm on the genetic divergence determination among cherry tomato accessions. The joint analysis of qualitative and quantitative variables has been considered a useful tool to estimate the genetic divergence among accessions of a gene bank. The purpose of this research was: (i) to characterize a collection of cherry tomatoes, using quantitative and qualitative descriptors, and (ii) to use Gower's algorithm to quantify genetic divergence among genotypes, employing a joint descriptors analysis. Forty accessions of organically grown cherry tomato were studied in Seropedica, Rio de Janeiro, Brazil. Four qualitative fruit descriptors and nine qualitative descriptors were used. Data were analyzed using Gower's algorithm. There was variability for color, shape and number of locules per fruit. The red color was observed in fruits of 25 accessions. Fruit with different colors such as yellow, orange and brown were also observed. Regarding locule number, the presence of fruit with two, three, four and even with seven locules was detected. The UPGMA method resulted in the highest value for cophenetic correlation (0.80) and seven clusters were formed. The formed clusters made it possible to distinguish fruit classified as cherry tomato from other type grouped in different clusters, because the fruits this category were larger than cherry type considering the equatorial diameter of the fruit.
Among the members of the genus Burkholderia, Burkholderia tropica has the ability to fix nitrogen and promote sugarcane plant growth as well as act as a biological control agent. There is little information about how this bacterium metabolizes carbohydrates as well as those carbon sources found in the sugarcane juice that accumulates in stems during plant growth. Reverse transcription quantitative PCR (RT-qPCR) can be used to evaluate changes in gene expression during bacterial growth on different carbon sources. Here we tested the expression of six reference genes, lpxC, gyrB, recA, rpoA, rpoB, and rpoD, when cells were grown with glucose, fructose, sucrose, mannitol, aconitic acid, and sugarcane juice as carbon sources. The lpxC, gyrB, and recA were selected as the most stable reference genes based on geNorm and NormFinder software analyses. Validation of these three reference genes during strain Ppe8 growth on the same carbon sources showed that genes involved in glycogen biosynthesis (glgA, glgB, glgC) and trehalose biosynthesis (treY and treZ) were highly expressed when Ppe8 was grown in aconitic acid relative to other carbon sources, while otsA expression (trehalose biosynthesis) was reduced with all carbon sources. In addition, the expression level of the ORF_6066 (gluconolactonase) gene was reduced on sugarcane juice. The results confirmed the stability of the three selected reference genes (lpxC, gyrB, and recA) during the RT-qPCR and also their robustness by evaluating the relative expression of genes involved in glycogen and trehalose biosynthesis when strain Ppe8 was grown on different carbon sources and sugarcane juice.
