RESUMOO objetivo deste trabalho foi avaliar a variabilidade de três vírus (Rupestris stem pitting-associated virus -RSPaV, Grapevine leafroll-associated virus 2 -GLRaV-2 e Grapevine fanleaf virus -GFLV) que infectam videiras no Brasil, através da caracterização molecular do gene da proteína capsidial (CP). Foram amplificados fragmentos que compreendem os genes completos da CP de nove isolados de RSPaV (780 pb), seis de GLRaV-2 (597 pb) e três de GFLV (1515 pb) por RT-PCR, utilizando-se oligonucleotídeos específicos para cada espécie viral. Os DNAs amplificados foram clonados e sequenciados. Os isolados de RSPaV foram reunidos em quatro grupos pela análise filogenética das sequências obtidas com valores de identidade de nucleotídeos (nt) que variaram de 81 a 99%. Para GLRaV-2, dois grupos foram definidos a partir das sequências de nt, com valores de identidade que variaram de 88 a 99% e, para GFLV, foram definidos dois grupos, com valores de identidade de nt que variaram de 89 a 98%. Os diferentes isolados das três espécies virais estudadas também foram detectados utilizando-se hibridização com sondas não-radioativas, marcadas com digoxigenina, possibilitando identificar de forma inequívoca as amostras infectadas, independentemente dos isolados virais empregados para síntese das sondas. -The objective of this work was to produce and characterize specifi c antisera against Brazilian isolates of Grapevine leafroll-associated virus 2 (GLRaV-2) and Grapevine virus B (GVB), developed from expressed coat proteins (CPs) in Escherichia coli, and to test their possible use for the detection of these two viruses in diseased grapevines. The coat protein (CP) genes were RT-PCR-amplifi ed, cloned and sequenced. The CP genes were subsequently subcloned, and the recombinant plasmids were used to transform E. coli cells and express the coat proteins. The recombinant coat proteins were purifi ed, and their identities were confi rmed by SDS-PAGE and Western blot and used for rabbit immunizations. Antisera raised against these proteins were able to recognize the corresponding recombinant proteins in Western blots and to detect GLRaV-2 and GVB in infected grapevine tissues, by indirect ELISA, discriminating healthy and infected grapevines with absorbances (A 405 ) of 0.08/1.15 and 0.12/1.30, respectively. Expressing CP genes can yield high amount of viral protein with high antigenicity, and GLRaV-2 and GVB antisera obtained in this study can allow reliable virus disease diagnosis.Index terms: Vitis, GLRaV-2, GVB, indirect ELISA, recombinant protein, Western blot. Produção de anti-soros policlonais a partir de proteínas capsidiais recombinantes de Grapevine leafroll-associated virus 2 e Grapevine virus BResumo -O objetivo deste trabalho foi produzir e caracterizar anti-soros específi cos contra isolados brasileiros do Vírus do enrolamento-da-folha da videira 2 (GLRaV-2) e do Vírus B da videira (GVB), desenvolvidos a partir das proteínas capsidiais expressas em Escherichia coli, e testar seu possível uso para a detecção destes dois vírus em videiras infectadas. Os genes da proteína capsidial (CP) foram amplifi cados via RT-PCR, clonados e seqüenciados. Foram, subseqüentemente, subclonados, e os plasmídeos recombinantes foram empregados na transformação das células de E. coli e na expressão das proteínas capsidiais. As proteínas capsidiais recombinantes foram purifi cadas, e suas identidades foram confi rmadas em SDS-PAGE e "Western blot" e utilizadas para imunizar coelhos. Os anti-soros produzidos contra essas proteínas foram capazes de reconhecer as proteínas recombinantes correspondentes em "Western blot", de detectar GLRaV-2 e GVB em tecidos infectados de videiras pelo ELISA indireto, e de discriminar videiras sadias e infectadas, com absorbâncias (A 405 ) de 0,08/1,15 e 0,12/1,30, respectivamente. A expressão dos genes CP pode produzir grandes quantidades de proteínas virais, com elevada antigenicidade, e os anti-soros de GLRaV-2 e GVB obtidos neste trabalho possibilitam a diagnose confi ável desses vírus.Termos para indexação: Vitis, GLRaV-2, GVB, ELISA indireto, proteína recombinante, Western blot.

O Apple stem pitting virus (ASPV) foi detectado por RT-PCR em amostras de cultivares de pereiras européias (Pyrus communis L.) cvs. Starkrimson e Abate Fetel, e asiáticas (P. pyrifolia var. culta) cvs. Kousui e Housui coletadas no início do outono de 2003 em pomar da Estação Experimental da Embrapa Uva e Vinho, Vacaria, RS. Utilizando várias combinações de oligonucleotídeos, foram amplificados fragmentos de DNA de 269 e 1554 pb, este último contendo o gene completo (1131 nt) da proteína capsidial do ASPV. Outro fragmento amplificado de 291 pb compreende parte do gene da polimerase viral. Estes fragmentos constituem-se em um excelente instrumento de diagnóstico do ASPV em pereiras. A comparação das seqüências de nucleotídeos do gene da proteína capsidial do ASPV com seqüências do banco de dados GenBank, revelou identidades de 89% com seqüências de um isolado alemão de macieira e de 85 a 88% com isolados poloneses, de pereiras. A indicadora herbácea Nicotiana occidentalis cv. 37B, inoculada mecanicamente com extrato foliar da cv. Housui, apresentou lesões locais necróticas, necrose foliar marginal e das nervuras. O ASPV também foi detectado por dot-ELISA nas cvs. Abate Fetel e Kousui, na cv. Starkrimson por imunoblot, e em Pyronia veitchii (Trabut) Guill. por enxertia de borbulhas da cv. Abate Fetel infetada.

Plant virus identification and characterization can be accomplished by several methods involving their morphological, physical, biological, cytological, serological and molecular properties. The use of molecular techniques is increasing worldwide, and some have been developed for identification and characterization of plant viruses. Reverse transcription polymerase chain reaction (RT-PCR) has been shown to be a suitable method for research with RNA plant viruses. In this study, a new approach of RT-PCR involving previous virus immunoprecipitation (IP) was used for RNA amplification of five virus species of the genera Comovirus, Cucumovirus, Potyvirus and Sobemovirus from infected plant tissues. IP-RT-PCR was practical, sensitive and minimized problems with total RNA extractions from infected tissues. The technique provides partial virus particle purification by its specific immunoprecipitation, and it should be especially useful for RNA amplification of viruses that occur in low or variable concentrations in plant tissues or when the tissues contain various forms of RT-PCR amplification inhibitors.
