In the imidazo[1,2]diazine series, only the imidazo[1,2‐a]‐ and [1,2‐c] pyrimidine systems were found to undergo a Dimroth‐type rearrangement under basic aqueous conditions. The electronic and steric factors influencing the rearrangement rates of methyl substituted imidazo‐[1,2‐a] pyrimidines are discussed.
Aus dem Imidazo(l ,2‐a)pyrimidinen (I) erhält man beim Erhitzen mit Alkali über eine in ihrem Ablauf ausführlich untersuchte Umlagerung die Isomeren (II), wobei sich z.Tl. Gleichgewichte zwischen den beiden Isomeren (I) und (II) einstellen.
Aminoimidazole, wie z.B. (I) geben beim Erhitzen mit den β‐Dicarbonylverbindungen (II) [(IIa) und (IIb) reagieren als Bis‐dimethylacetale und (IIc) als Dimethylacetal] in Äthanol oder Essigsäure die Zwischenstufen (III), die durch Säurekatalyse zu den Imidazopyrimidinen (IV) (25‐95% Ausbeute) cyclisiert werden können; die Reaktion von (I) mit (IIc) (als Dimethylacetal) in Methanol/HCl führt über (V) zu dem Derivat (VI).
Das durch katalytische Hydrierung des entsprechenden Nitroderivats in neutraler wäßriger Lösung erhältliche Aminoimidazol (I) reagiert in situ in Gegenwart von Essigsäure mit Tetrarnethoxypropan (II) zu der Titelverbindung (III) (Ausbeute bezogen auf das Nitroimidazol) als Hauptprodukt; daneben entsteht das Isomere (IV).
