Functional motor changes and morphological alterations have been associated with intestinal inflammation. The aim of our study was to evaluate functional alterations of intestinal reflexes and of the responses to CCK in the Trichinella spiralis model of intestinal inflammation. Rats were prepared with strain gauges and electrodes in the small intestine to evaluate spontaneous motor activity, the ascending contraction of the peristaltic reflex, and the motor responses to CCK-8 infusion. Infected animals showed increased motor activity at the duodenum and jejunum but not at the ileum. Ascending contraction was increased in both duodenum and ileum. Ascending excitation after N(omega)-nitro-L-arginine was still increased as well as the residual response after atropine. Response to CCK-8 during intestinal inflammation was changed in the jejunum, in which it turned from the inhibition shown in healthy animals to excitation. NADPH-diaphorase staining did not show any changes between distribution and density of positive neurons in either healthy or infected animals. In conclusion, intestinal inflammation induces functional changes in the motor activity that could explain the abnormal motor responses observed in inflammatory disorders.

The main objective was to study the role of nitric oxide (NO) in the conversion of migrating myoelectric complexes (MMC) to the irregular electrical activity characteristic of the postprandial state. Both rats and chickens were implanted with electrodes for electromyography in the small intestine. Intravenous infusion of NG-nitro-L-arginine (L-NNA), a NO synthase inhibitor, induced an organized MMC-like pattern in fed rats. Infusion of sodium nitroprusside, a NO donor, disrupted the MMC, inducing a postprandial-like motor pattern in fasting rats. Similarly, in chickens L-NNA mimicked the fasting pattern, consisting of a shortening of phase II, enlargement of phase III, orad displacement of the origin of the MMC, and an increase in the speed of phase III propagation. An inhibition of NO synthesis seems to be involved in the induction of the fasting motor pattern, whereas an increase of NO mediates the occurrence of the fed pattern. It is suggested that NO might be the final mediator in the control of small intestine motor patterns.

SUMMARYThe aim of this study was to find out how marker characteristics could affect digestive transit time in Gallus gallus. One soluble marker, Cr-EDTA, and two insoluble markers, Cr203 and chromium-mordanted plant cells of two sizes, were used. Three-to six-week-old chickens were killed in series after the oral administration of the markers at intervals of 0, 0 5, 1, 2, 3, 5, 7, and 9 h. The amount of chromium in each digestive segment was determined by atomic absorption. There were some differences in the initial distribution of markers: whereas almost the total amount of the chromium-mordanted rice husk of the largest size was found in the crop at time 0, less than half of the Cr-EDTA was found. Marker emptying out of the crop was fast and not related to either the type or size. In contrast, the emptying rate of the gizzard depended on marker particle size. As far as the caeca were concerned, the ileocaecal junction allowed the passage of soluble Cr-EDTA whereas solid markers were impeded (Cr2O3) or not allowed to pass through at all (vegetable fibre of any size). It can be concluded that marker selection is of major importance to transit time studies in chickens, since its characteristics can determine transit time in an absolute way.

SUMMARYThirty broilers 8-10 weeks old were used to study the rate of food passage in chickens. Wheat bran and rice husks of three different sizes: more than 2 mm, between 1 and 15 mm, and less than 0 5 mm, mordanted with chromium, were used as markers. The suitability ofthese markers to study the rate of food passage in chickens and the possible influence of the size and hardness of the particle on the retention time was the objective of this study. Both T1, the time of first appearance of the marker, and Tm, the mean retention time, have been evaluated. Tm was a better parameter than T1 for studying transit time. Tm was longer with the biggest particles, especially with rice husk. The gizzard, with its grinding activity and pylorus, a selector ofparticle size, seems to be the transit regulator for solid particles in chickens. No chromium was found in the caecal contents of any case.

The structural and functional bases of pacemaking (slow waves) in porcine ileal circular muscle were studied. The myenteric plexus contained two, structurally distinct types of interstitial cells of Cajal (ICC) interconnected by gap junctions and connected by close contacts to muscle layers. At the deep muscular plexus, ICC were present, not regularly close to nerve axons or in gap junction contact with one another or outer circular muscle, which had many gap junctions. Slow waves (5.2 ± 2 mV amplitude and 4.6 ± 0.7 s duration) occurred at 9.9 ± 1.1 counts/min. Tissue length and time constants were 2.00 ± 0.3 mm and 111 ± 37 ms, respectively. Large electrical field-induced hyperpolarizations or depolarizations reduced amplitudes but not frequencies or durations of slow waves; hyperpolarizations progressively reduced inhibitory junction potentials as if the K+channel opening mediated them. In conclusion, the myenteric plexus ICC of pig ileum, which appears to pace the muscle layers, appears insensitive to voltages applied to the syncytium of circular muscle cells. Limited coupling between ICC and circular muscle or voltage-insensitive pacemaking activity may explain these findings.
