Herein we show that functional phenylalkylamine-sensitive L-type calcium channels are expressed by human NK cells and are involved in the killing of tumor targets. Blocking of these channels by phenylalkylamine drugs does not affect effector/target cell binding but inhibits the release of serine esterases responsible for cytotoxicity. Interestingly, treatment of NK cells with HIV-1 Tat, which is known to affect several calcium-mediated events in immune cells, impairs their cytotoxic activity. In addition, Tat inhibits the rise in intracellular free calcium concentration upon cross-linking of the adhesion molecule CD11a, engaged during effector/target cell interaction, and the activation molecule CD16. Exogenous Tat does not influence NK-target cell binding but prevents NK cell degranulation. We propose that the molecular structure(s) on NK cells mediating the inhibitory effects HIV-1 Tat belong to L-type calcium channels, based on three lines of evidence: 1) binding of phenylalkylamine derivatives to these channels is cross-inhibited by Tat; 2) L-type calcium channels from NK cell lysates bind to Tat linked to Sepharose columns; 3) the inhibitory effect of HIV-1 Tat on NK cell function is prevented by the agonist of L-type calcium channels, Bay K 8644. Altogether, these results suggest that exogenous Tat is deeply involved in the impairment of NK cell function during HIV-1 infection.

IntroduzioneLe neoplasie maligne della glia ed in particolare il glioblastoma multiforme, sono tumori a crescita rapida per i quali la chemioterapia e d'importanza limitata, in quanta non esiste una terapia di prima linea.Solo la radioterapia, dopo chirurgia radicale, permette di prolungare la sopravvivenza media. Oggigiorno la terapia medica e radioterapica non hanno modificato la prognosi di tale forma di tumore, per la quale l'incidenza e la mortalita sono analoghe 1 • 2 • La caratteristica di questo tumore di non metastatizzare al di fuori del sistema nervosa centrale ha portato all'introduzione di una terapia locoregionale intracranica. In particolare due inibitori delle DNA Topoisomerasi II, !'Etoposide (VP16) e il Teniposide (VM26), si sono dimostrati potenzialmente efficienti per questa terapia 3 • 4 • Recentemente ha dato risultati promettenti il Tumor Necrosis Factor (TNF) somministrato per via intraarteriosa 5 • La possibilita di una terapia combinata TNF + VP16 + TNF + VM26 potrebbe essere di notevole vantaggio nei confronti di questa neoplasia.Razionale a questa terapia e la conduzione di studi sperimentali atti a capire la biologia di tale neoplasia o a modularne le caratteristiche.In questa comunicazione abbiamo studiato la capacita del TNF di sensibilizzare una linea di glioblastoma multiforme umano T98G al Teniposide e all'Etoposide, valutando in particolare l'attivita citotossica del VP16, del VM26 e del TNF «in vitro» e la modulazione della stessa nell'associazione dei due farmaci con la citochina; la produzione cellulare di TNF e la presenza di recettori per tale fattore sulla membrana citoplasmatica. Inoltre abbiamo saggiato l'azione farmacologica del VP16 sia da solo sia in combinazione con il TNF attraverso l'analisi dei DNA single strand breaks in eluizione alcalina. RisultatiPrimo passo nel valutare l'azione antiproliferativa dei due farmaci inibitori della DNA Topoisomerasi e del TNF e stato quello di saggiare la sopravvivenza cellulare dopo un periodo di incubazione di 24 h + 20 h di esposizione a diverse concentrazioni di ciascuna sostanza a 3rC, in base ad un adattamento del metodo proposto da Ruff e Gifford 6 • Successivamente, abbiamo anche testata l'azione sinergica del VP16 e del VM26 incubati contemporaneamente al TNF a concentrazioni equitossiche per 20 h a 37°C. Da un'analisi dei risultati riportati in Tabella 1 emerge che la linea cellulare T98G risulta essere sensibile in modo significativo all'azione antitumorale del Teniposide e dell'Etoposide solo quando vengono somministrati insieme al TNF, che da solo da una sopravvivenza del 100%, alla concentrazione di 1000 U/ml. Infatti, in tali condizioni, si osserva un considerevole incremento della citotossicita non altrimenti rilevabile in assenza di tale fattore di necrosi.
