Heat shock protein 70 (Hsp70; also known as HSP70A1A) is one of the most induced proteins in cancer cells; however, its role in cancer has not yet been fully elucidated. In the present study, we proposed a hypothetical model in which the silencing of Hsp70 enhanced the metastatic properties of the HeLa, A549 and MCF7 cancer cell lines. We consider that the inability of cells to form cadherin-catenin complexes in the absence of Hsp70 stimulates their detachment from neighboring cells, which is the first step of anoikis and metastasis. Under these conditions, an epithelial-to-mesenchymal transition (EMT) pathway is activated that causes cancer cells to acquire a mesenchymal phenotype, which is known to possess a higher ability for migration. Therefore, we herein provide evidence of the dual role of Hsp70 which, according to international literature, first establishes a cancerous environment and then, as suggested by our team, regulates the steps of the metastatic process, including EMT and migration. Finally, the trigger for the anti-metastatic properties that are acquired by cancer cells in the absence of Hsp70 appears to be the destruction of the Hsp70-dependent heterocomplexes of E-cadherin/catenins, which function like an anchor between neighboring cells.
Η HSP70 αποτελεί έναν από τους δημοφιλέστερους μοριακούς συνοδούς. Διαδραματίζει σημαντικό ρόλο την επιβίωση του κυττάρου δεδομένου ότι εμπλέκεται άμεσα ή έμμεσα σε πολλά βασικά κυτταρικά μονοπάτια. Στη βιβλιογραφία έχει σχετιστεί με κυτταρικούς μηχανισμούς που εμπλέκονται σε ασθένειες, όπως είναι ο καρκίνος και διάφορα καρδιαγγειακά νοσήματα, όπου εκφράζεται σε πολύ υψηλά ποσοστά.Στην παρούσα εργασία ο κύριος στόχος ήταν η διερεύνηση του ρόλου της HSP70 στην Anoikis απόπτωση καθώς και στην επαγωγή μεσεγχυματικής μετάπτωσης σε ενδοθηλιακά και καρκινικά κύτταρα.Τα αρχικά μας πειράματα πραγματοποιήθηκαν στα ενδοθηλιακά κύτταρα HMEC1, όπου μελετήθηκε η απόκριση των κυττάρων στο θερμικό σοκ και καθορίστηκε η καταλληλότερη τεχνική διαμόλυνσης των κυττάρων με εξωγενές DNA. Εν συνεχεία κατασκευάστηκε το πλασμίδιο pcDNA3-siRNA-HSP70, το οποίο φέρει ειδικά επιλεγμένο ένθεμα για την έκφραση siRNA, για τη γονιδιακή αποσιώπηση της HSP70. Πραγματοποιήθηκε απόπειρα δημιουργίας σταθερής κυτταρικής σειράς κυττάρων HMEC1 με γονιδιακή αποσιώπηση της HSP70, τα κύτταρα όμως δεν επιβίωσαν. Σε καλλιέργεια μαζικών κλώνων μελετήθηκαν οι φαινοτυπικές μεταβολές που συνοδεύουν τα κύτταρα HMEC1 με αποσιωπημένη την HSP70. Παρατηρήθηκε ότι τα κύτταρα απέκτησαν έναν πιο μεσεγχυματικό φαινότυπο (κύτταρα ατρακτοειδή, απουσία διακυτταρικών συνδέσεων, πολύστοιβη ανάπτυξη), γεγονός που επιβεβαιώθηκε μοριακά, με ανοσοαποτύπωση κατά Western και ανοσοφθορισμό, από την απώλεια της ενδοθηλιακής πρωτεΐνης VE-καντερίνη και την έκφραση της μεσεγχυματικής πρωτεΐνης βιμεντίνη.Στο επόμενο μέρος της εργασίας μελετήθηκε ο ρόλος της HSP70 σε βασικές για την εξέλιξη των όγκων διαδικασίες, όπου παρατηρήθηκαν τα παρακάτω: Η απουσία της HSP70 δεν επηρεάζει τον ρυθμό ανάπτυξης των κυττάρων παρά μόνο σε συνθήκες πλήρους ταπητίου. Η απουσία της HSP70 αυξάνει τη μεταναστευτική ικανότητα των κυττάρων. Σε συνθήκες στρες όπως είναι η αυξημένη θερμοκρασία, η παρουσία χημειοθεραπευτικών ουσιών και η απουσία προσκόλλησης των κυττάρων (πρόκληση Anoikis απόπτωσης) η HSP70 προσδίδει ανθεκτικότητα στα κύτταρα με τον αντι-αποπτωτικό της ρόλο. Τα χημειοθεραπευτικά δοξορουβικίνη και cis-πλατίνη αυξάνουν τη μετανάστευση των κυττάρων, ενδεχομένως επάγοντας την ΕΜΤ.Εν συνεχεία προτείναμε ένα μοντέλο για την εμπλοκή των πρωτεϊνών β-κατενίνη και p53 στην Anoikis απόπτωση των κυττάρων σύμφωνα με το οποίο απουσία προσκόλλησης με την ECM και διακυτταρικών συνδέσεων η β-κατενίνη απελευθερώνεται από τα σύμπλοκα των διακυτταρικών συνδέσεων, στο κυτταρόπλασμα όπου συσσωρεύεται και ενεργοποιεί την p53. Η φωσφορυλιωμένη ενεργή p53 μεταβαίνει στον πυρήνα όπου, σύμφωνα με την βιβλιογραφία, επάγει τη μεταγραφή γονιδίων που σχετίζονται με την απόπτωση και στη συνέχεια η ενεργοποίηση αυτών των αποπτωτικών μονοπατιών καταλήγει στην ενεργοποίηση των κασπασών και την πρωτεόλυση της β-κατενίνης από αυτές.Τέλος επιβεβαιώθηκε η ετερογένεια των καρκινικών κυττάρων όσον αφορά την έκφραση επιθηλιακών και μεσεγχυματικών δεικτών και αποδείχθηκε ότι η απουσία της HSP70 οδηγεί στην απόκτηση ενός μεσεγχυματικού φαινοτύπου από τα κύτταρα (ΕΜΤ), με τρόπο ανεξάρτητο του WNT μονοπατιού.
