Nach den Vorschriften der Pharmakopoen werden Thyreoidpriiparate nach zwei unterschiedlichen Methoden untersucht. I n einigen Anneibuchern wird die Bestimmung des organisch gebundenen Gesamtjods vorgeschrieben (z. B. Hung. V, USP XVI, Ross. VIII), in anderen die Bestimmung des Thyroxingehalts auf Jod-Basis, wobei man sich auf die Untersnchungen Kendalls stutzt, nach denen in der Schilddriise das Thyroxin der wirksame Bestandteil ist (z. B. Helv. V, BPS 8, Franc. 49, P I). Unserer Ansicht nach ist die erste Methode die richtigere, da in neueren Untersuchungen nachgewiesen werden konnte, d a l auch die in der Schilddriise vorhandenen Derivate rnit niedrigerem Jodgehalt, hauptsiichlich Trijodthyronin, wirksam sind.Die gebriiuchlichen Vntersuchungsverfahren beruhen im wesentlichen auf dem sog. ,,festen" Schmelzverfahren, bei dem sich Alkalijodid bildet, das.in Jodat iibergefuhrt wird. Auch die Bestimmung des Thyroxinjods wird iihnlich durchgefuhrt, vorher jedoch das Thyroxin von den saureloslichen Verbindungen mit Hilfe der von KaZin ausgearbeiteten Methodel) getrennt.Zum Schmelzverfahren wird entweder festes Kalinmhydroxyd oder (meist) Natriumcarbonatevtl. unter Zusatz von anderen Hilfsmitteln, die den Verlanf der Reaktion begunstigenverwendet. Im Zuge unserer Arbeiten haben wir festgestellt, daB die erhaltenen Werte stark streuen und niedriger ausfallen als theoretisch zu erwarten war; besonders auffallend war dies bei der Kri'linschen Methode. So bemuhten wir uns, ein neues Verfaliren auszuarheiten.Zunachst stellten wir fest, daB die in den einzelnen Arzneibuchern vorgeschriebenen Schmelzverfahren mehrere Fehlerquellen in sich bergen. So ist es auch beim vomichtigsten Arbeiten unvermeidlich, daB der im Nickeltiegel mit KOH geschmolzene Stoff schiiumt und spritzt. Diese Fehlermoglichkeit liiDt sich mit der Anwendung von Natriumcarbonat und eines Deckels zwar vermindern, jedoch gibt die vorgeschriebene Temperatur von etwa 700" infolge der Fliichtigkeit von Jodverbindungen zu Bedenken Anlal.Ein zweiter Fehler kann dadurch verursacht werden, d a l die Oxydation der Alkalijodide zn Jodat in saurem Medium durchgefiihrt wird, wobei sich das gebildete H J zum Teil verfliichtigen kann, noch bevor die Umsetzung zu Jodat stattgefunden hat.Bei der Bestimmung des Thyroxinjods nach Kalin wird der Stoff zuerst im alkalischen Medium unter RuckfluB gekocht. Wird mit der Gasflamme, mit einem elektrischen Heizkorper oder mit irgendeinem anderen auf mehr als 100" erhitzten Wiirmeiibertragungsmedium erhitzt, so besteht die Gefahr, da13 der im wiilrigen Medium nur teilweise losliche Stoff uberhitzt wird und evtl. anbrennt. Auch kann e+~ unter den gegebenen Bedingungen vorkommen, d a l das Jodthyreoglobulin zwar aufgeschlossen wird, die Spaltung der iibrigen Komponenten (Di-und Monojcdtyrosin) jedoch nnr teilweise erfolgt. *) Vortrag auf dem V.
