A microplate instrument has been developed enabling spectral differentiation of luminescent signals as is needed in Bioluminescent Resonance Energy Transfer (BRET). So far modified fluorescence readers have been used but suffered from poor sensitivity. The new instrument uses a novel type optical system combining extremely low crosstalk with high light collection efficiency. Computer controlled filter wheels can automatically position filters in the optical path. Reagent injectors allow for the measurement of assays with fast signal kinetics. A specially selected Bialkali PMT with enhanced sensitivity in the green spectral range is used. The instrument has shown an increased sensitivity of more than one order of magnitude compared to typical instruments used so far for BRET applications.
