El cacao (Theobroma cacao) enfrenta durante cada uno de sus ciclos productivos patógenos que disminuyen considerablemente a su producción. Cada año fitopatógenos como Moniliophthora roreri y Phytophthora palmivora afectan directamente a las mazorcas y pueden llegar a generar graves pérdidas. Esta investigación se desarrolló con la finalidad de identificar especies vegetales locales cuyos extractos contengan compuestos que tienen la capacidad de reducir la germinación de las esporas de los patógenos, así como también el desarrollo de micelio de los mismos. Extractos de las especies Bledo (Amaranthus bledo), Coquito (Cyperus odoratus), Lechosa (Euphorbia hirta), Rosa de muerto (Tagetes minuta), Teatina (Scoparia dulcis) y Verdolaga (Portulaca oleracea) se utilizaron para enmendar medios de cultivos en concentraciones de 10, 20 y 30%, sobre los cuales se inocularon suspensiones de esporas y discos de micelio de los patógenos. Transcurridos los periodos de incubación se evaluaron los porcentajes de germinación, porcentaje de inhibición de desarrollo del micelio, actividad fungicida de los extractos y su efecto sobre las estructuras de las hifas de los patógenos. Los resultados mostraron que todos los extractos en alguna medida influyeron en alguna medida en el desarrollo de los patógenos en todas las variables estudiadas, sin embargo, Amaranthus bledo y Euphorbia hirta fueron aquellas especies cuyo efecto fue notablemente superior a las otras especies en estudio, lo que indicaría que son especies con potencial para controlar los patógenos a nivel de campo.
RESUMEN.-Para comparar la técnica de PCR anidado frente a la prueba serológica Rosa de Bengala (RB) en la detección de Brucella spp. se utilizó 40 muestras de sangre, de cuatro hatos de bovinos con alta prevalencia de brucelosis en la región central de Ecuador. El 35% de los animales dio positivo para el análisis con RB, mientras que con PCR simple fue del 45% con una correlación del 30%, lo que sugiere que la PCR frente a la RB es 22% más sensible. El análisis de correlación de los animales positivos obtenidos entre PCR y PCR anidado fue del 100% y, la sensibilidad del PCR anidado demostró el 37,9% superior a la PCR. Entre RB y PCR anidado, la correlación de sensibilidad fue del 51,7% superior para la segunda. La detección de los animales positivos mediante PCR y PCR anidado se mostró más específica y sensible que RB, por lo tanto en este trabajo se considera al PCR anidado como una herramienta molecular útil en el diagnóstico de Brucella spp. y su uso en la prevención de la enfermedad.PALABRAS CLAVE: Brucelosis, correlación, diagnóstico, enfermedad, prevalencia.ABSTRACT .-Forty blood samples from 4 herds with high prevalence of brucellosis in the central region of Ecuador were used in order to compare the nested PCR with the serological Rose Bengal (RB) tests for the detection of Brucella spp. Thirty five % of the animals were detected positive with the RB analysis and 45% with simple PCR , with a correlation of 30%, suggesting that PCR was 22% more sensitive than the RB serological method. Resumen La tuberculosis y la brucelosis siguen siendo causas importantes de morbilidad y mortalidad en muchos países, para la detección de ambas enfermedades se requieren de herramientas eficientes y sensibles para efectivizar el diagnóstico. Este trabajo fue dirigido a evaluar y comparar la técnica de Dúplex PCR frente a la PCR anidada, para la detección de Brucella spp. (BR) y Mycobacterium spp. (TB). Se utilizó un total de 100 muestras de tejidos a partir de nódulos linfáticos traqueo bronquiales, pulmón de bovinos y, aislados bacterianos de TB y BR como controles positivos. Diez combinaciones de iniciadores dirigidos a flanquear un segmento de la secuencia 16S ARNr (BR) y el gen antígeno MPB70 (TB) fueron evaluados, el mejor resultado para la Dúplex PCR se logró con los iniciadores Bru-2F/Bru-2R para BR y para TB, Tub-1F/Tub-N-R. Los productos de amplificación fueron de 225 y 230-pb respectivamente. A fin de potencializar los resultados de la técnica propuesta, todas las muestras fueron inicialmente analizadas y comparadas entre la PCR simple y la PCR anidada (Kappa, k = 0,85) y, la concordancia entre los resultados obtenidos con la PCR anidada y la Dúplex PCR (k = 0,88), para las dos bacterias fue muy buena.
RESUMEN.-En este estudio, los análisis con marcadores RAPD (Random Amplification of Polymorphic DNA) fueron usados para estimar la diversidad genética y la relación de 20 accesiones de Theobroma cacao L. de la variedad Nacional en la finca (La Buseta) ubicada en la Parroquia Tenguel provincia del Guayas. Las plantas fueron seleccionadas con criterios de productividad, resistencia, susceptibilidad a enfermedades causadas por hongos como Ceratocystis fimbriata, Moniliophtora roreri y Crinipelis perniciosa. Se utilizaron 14 primers, de los cuales nueve amplificaron, generando 67 bandas de las cuales 59 (88%) fueron polimórficas. Los resultados indican la formación de dos grupos de accesiones A y B, en el grupo A se incluyen dos accesiones y, en el grupo B se encuentran 18 de los cuales incluyen 2 subgrupos B1 y B2. E el subgrupo B1 se incluye la accesión (L-22-H-40), y en el subgrupo B2 están incluidas las accesiones que poseen características de productividad. El nivel de diversidad génica más alto se obtuvo con los primers OPC 04 (0,80), OPC 07 (0,82) y el nivel más bajo fue OPC 01 (0,37).
