The operating conditions of polyhydroxybutyrate (PHB) production processes are among the factors that most influence yields. In this study, we evaluated PHB production synthesized by Bacillus megaterium LVN01. Batch and fed-batch cultures were used to produce PHB from residual glycerol. For this, dry cell weight (DCW) and PHB productivity were analyzed at a preliminary stage by central composite design using batch systems under different temperature, C/N ratio, and fermentation time conditions. The maximum PHB productivity occurred at 30.8 °C, 44.9 mol mol-1, and 39.9 h. The same conditions were tested for studies in fed-batch culture. Fed-batch experiments were comparable to each other, where the DCW was around 1.9 g L-1, with PHB productivities of 29.5 mg L-1 h-1 and 35.6 mg L-1 h-1 for bioreactors of 5 L and 14 L, respectively. The PHB was characterized by NMR, FTIR, DSC, TGA, and DTG analys

The E4/E6 ratio, the ∆ log K, the melanic index (MI) and the optical density of oxalate extract (ODOE) were calculated from the absorbances. The results of the E4/E6 ratio, the ∆ log K and the MI in the extracts obtained with sodium pyrophosphate and with ammonium acid oxalate showed a higher degree of humification for the organic matter extracted from the A and 2B/A horizons and one smaller for the one extracted from the 3C/A and 4Bs horizons. The ODOE showed the presence of organic complexes in the extracts obtained with sodium pyrophosphate and with sodium hydroxide and the iluvial accumulation of non-crystalline Fe oxides and hydroxides in the 4Bs horizon in the extract obtained with ammonium acid oxalate. In the horizon A, the humification indexes marked a change in the degree of evolution of the MO between 8 and 10 cm of depth, which could indicate the presence of a lithological discontinuity or of buried soil.KEYWORDS: Organic complexes; evolution; selective extractions of Al, Fe and Si; humification indexes. This study aimed to determine the nutritional value of the fatty acids in the freshwater fish Prochilodus magdalenae, Pseudoplatystoma magdaleniatum and Ageneiosus pardalis during dry and wet Neotropical seasons with the view to generate useful information for nutrition and sustainable commercial exploitation. The analysis of fatty acids was performed by gas chromatography–mass spectrometry and the nutritional value was calculated using five estimators: n-6/n-3 ratio, unsaturation index (UI), atherogenicity index (AI), thrombogenicity index (TI) and ratio of hypocholesterolemic/hypercholesterolemic (h/H) fatty acids. A different number of fatty acids (P. magdaleniatum = 50, P. magdalenae = 41, and A. pardalis = 32) was identified for each species and the average abundance percentages were different in the two seasons (p < 0.05). Prochilodus magdalenae and P. magdaleniatum showed healthy n-6/n-3 ratios (1.04 and 2.72) in the different seasons. Although the three species showed low values of UI (0.37–0.63), the remaining nutritional indexes were within the healthy range (AI: 0.04–0.70, TI: 0.66–1.07, h/H: 0.80 – 24.40). Multivariate analysis showed similar healthy nutritional values for the species, with exception of P. magdaleniatum.
