Kim Sampaio de Lacerda Mileski – UnB, pesquisador colaborador kim_mileski@hotmail.comA obesidade é apontada como fator disruptivo da normalidade de níveis hormonais e físicos, estando relacionada a enfermidades com elevado grau de morbidade e mortalidade, como hipertensão, diabetes e acidentes vasculares. A velocidade de absorção e armazenamento de gordura no Tecido Adiposo (TA) estão associados diretamente a questões endócrinas, genéticas, sedentarismo, gasto energético e tipo de dieta do indivíduo. O gene UCP1 é alvo dos estudos que tem como temática a obesidade e a termogênese, por ter um papel importante no funcionamento do Tecido Adiposo Branco (WAT) e do Tecido Adiposo Marrom (BAT), assim como a enzima AMPK no tecido muscular. O hipogonadismo trata-se de um descontrole fisiológico, em alguns casos decorrente do grau de obesidade, caracterizado pela incapacidade de produzir testosterona, espermatozoides circulantes e consequentemente infertilidade, além de dificuldade de manutenção do tecido muscular. O objetivo do presente estudo foi avaliar a síntese de testosterona de camundongos (n=25) machos de linhagem C57BL/6 submetidos à Dieta Hiperlipídica (DHL) e diferentes protocolos de atividade física. Para realização da pesquisa os animais foram divididos em cinco grupos para avaliação física pré e pós indução de obesidade por DHL. Os grupos realizaram diferentes protocolos experimentais segregados em cinco grupos: (CC) grupo controle, (IF) grupo de inatividade física, (CH) grupo tratado com DHL sem exercício, (MO) grupo tratado com DHL submetido a exercício moderado contínuo, (IN) grupo tratado com DHL submetido a exercício intenso intervalado. Ao término dos experimentos os animais foram novamente avaliados fisicamente e tiveram seus dados, sangue e tecidos de interesse coletados. O material genético foi submetido à reação em cadeia da polimerase em tempo real (qRT-PCR), sendo realizada a amplificação do gene UCP1 por pares de oligonucleotídeos iniciadores específicos. A dosagem de testosterona foi realizada por meio ensaio imunoenzimático competitivo (ELISA kit). Os grupos submetidos a DHL obtiveram um ganho mais acentuado de peso em relação ao CC no momento pré protocolos de atividade física. Após os testes de os grupos MO e IN ganharam menos peso que o CC. Os indivíduos do protocolo de atividade moderada melhoraram a distância percorrida no teste de esteira em relação aos CH e CC. Os grupos CH e MO apresentaram massa dos testículos menores que CC, enquanto o grupo de inatividade física foi semelhante à do grupo controle. A massa do BAT do grupo IN foi maior que o CC, apontando maior eficiência da termogênese neste protocolo. O período de protocolo experimental com camundongos com obesidade induzida por DHL minimizou o ganho de peso quandocomparado com o CC, como também o ganho de peso nos animais magros. Tais dados estabelecem uma relação direta de práticas de atividade física com prevenção de obesidade e afecções metabólicas. Outros estudos envolvendo a termogênese e o papel do BAT são importantes para compreender melhor relação entre tal tecido e a perda da massa de tecido testicular, como também, o aumento da termogênese em protocolos de atividade de alta intensidade
A puberdade é a fase que marca a transição da infância para a fase adulta, sendo evidenciada por mudanças fisiológicas, psicológicas e sociais que apontam o desenvolvimento de caracteres sexuais e o amadurecimento da capacidade reprodutiva. Dentro das reações endócrinas, a reativação do eixo Hipotálamo-Hipófise-Gonadal (HHG) é responsável pelo desenvolvimento dos caracteres sexuais e por estimular a síntese de hormônio folículo estimulante (FSH) e hormônio luteinizante (LH). A ativação prematura do eixo HHG resulta em indivíduos com puberdade precoce central (PPC), caracterizada pela evolução puberal prematura. Como consequência dessa afecção têm-se o aumento das gônadas, antecipação da menarca, desenvolvimento mamário e aumento do número e espessura dos pelos. Um paciente é diagnosticado com PPC quando os sinais e sintomas manifestam-se em meninos menores de 9 anos e meninas menores de 8 anos de idade. Estudos demonstram que mutações nos genes KISS1, KISS1R, LIN28B, GPR54 e MKRN3 estão associadas à puberdade precoce. Mutações no gene MKRN3 são descritas em estudos como responsáveis pela PPC idiopática. O objetivo do presente estudo foi investigar a ocorrência de mutações no gene MKRN3 em 30 pacientes previamente diagnosticados com PPC residentes no Distrito Federal e entorno. Para realização da pesquisa foram coletadas amostras de sangue periférico dos pacientes, por punção intravenosa, e extração do DNA por meio do kit da QiAgen. O material genético obtido foi submetido à reação em cadeia da polimerase (PCR), sendo realizada a amplificação do gene MKRN3 por pares de oligonucleotídeos iniciadores específicos. A amplificação dos produtos de PCR foi confirmada em gel de agarose a 1% e os produtos foram submetidos ao sequenciamento automático de Sanger. A análise do sequenciamento foi realizada pelo software Sequencher® e os resultados encontrados foram comparados com os principais bancos de dados genômicos. Foram identificados dois pacientes com mutações autossômicas dominantes de penetrância incompleta no gene MKRN3, sendo duas irmãs com mutação missense c.982C_T/p.Arg.328.Cys. A primeira paciente foi incluída no estudo com 7 anos e 11 meses de idade apresentando menarca e pubarca aos 7 anos 8 meses, telarca aos 7 anos, idade óssea de 11 anos, e P2M3 na escala de Tanner. A segunda paciente, com 7 anos e 2 meses de idade apresentando telarca e axilarca aos 6 anos, e P2M2 na escala de Tanner. Os resultados do presente estudo foram fundamentais por auxiliarem no desenvolvimento de pesquisas que estabelecem a relação fisiológica entre os achados genéticos e os fenótipos de afecções, nos quais é possível a ampliação das informações descritas na literatura sobre puberdade precoce. Os dados obtidos são importantes por possibilitarem o acompanhamento genético dos pacientes e a identificação de novos casos familiares, como apontados em estudos que evidenciam a ocorrência de PPC familiar. A investigação genética de outros membros das famílias com pacientes diagnosticados é de suma importância para detectar a patogênese das mutações e potencializar o sucesso do tratamento. Mesmo com o avanço dos estudos, a investigação das causas da puberdade precoce ainda necessita de maior aprofundamento sendo uma área de destaque para pesquisas científicas
