A number of sulfamides were obtained by reaction of (5-(dichloromethylene)-2-oxoimidazolidin-4-ylidene)sulfamoyl chloride with anilines, benzylamines, Boc-protected piperazine, methylalylamine, and amino acids methyl esters with primary and secondary amino group. The antiviral and anticancer activity of new derivatives was evaluated. The most effective compounds against Human cytomegalovirus were sulfamides based on anisidine (1b), N-Boc-piperazine (1h), and the derivatives 1n,o with fragments of nipecotic and azetidine-3-carboxylic acids, respectively. Anticancer activity was most significant for sulfamides based on p-methoxybenzylamine (compound 1d), benzylmethylamine (compound 1f), and allylmethylamine (compound 1g).

This review highlights the promising science that has arisen from the synthesis of novel azaheterocycles from isocoumarins. Specific topics include their synthesis and biological activity. Isocoumarins (1H-isochromen-1-ones) are promising synthons, in particular due to the presence of a deoxybenzoin fragment which opens up wide possibilities for synthetic transformations. The deoxybenzoin cycle is highly susceptible to the action of various nucleophilic agents; its oxygen atom participates in recyclization reactions under the action of N-nucleophiles where it is replaced by a nitrogen atom, making it possible to obtain isoquinolones via a reaction with ammonia, primary amines, hydroxylamine and benzodiazepinones via a reaction with hydrazine. We systematize literature data, including patents (about 60 publications in all), demonstrate the routes of 3- arylisocoumarins modification under the action of N-nucleophiles - ammonia and primary amines, diamines, secondary amines, (het)arylamines, hydroxylamine, and hydrazines, and discuss the practical importance of these studies for medicinal chemistry.

547.814.5 V. V. Ishchenko, and V. P. Khilya 3-(2-Pyridyl)coumarins were prepared by reaction of substituted salicylaldehydes and 2-pyridylacetonitrile. Benzylation, acylation,coumarin was studied.Compounds with a coumarin ring are widely distributed throughout the plant kingdom and much less in animals. They belong to a large group of so-called phenolic compounds, the formation of which is characteristic of all representatives of the plant kingdom. Therefore, the important role in the life-cycle of plants of these compounds, which contribute to plant protective reactions and in many instances their pigmentation, cannot be disputed.As a rule, coumarins in nature occur as glycosides (highest contents in plants of the Umbelliferae, Rutaceae, Solanaceae, and Fabaceae families). Natural coumarins have been used in medicine (anticoagulants), foods, and fragrances. Synthetic coumarins and their analogs are used as fluorescent probes and markers for biological research and as antibiotics, antiallergens, and fungicides in medicine.We studied the synthesis of coumarins with a pyridine substituent via Knoevenagel condensation of substituted salicylaldehydes with 2-pyridylacetonitrile [1-8]. The resulting 2-iminocoumarins were converted to coumarins 1-10 by hydrolysis of the imine (Scheme 1). The signals of the other coumarin protons depend on the nature and position of the substituent in the coumarin ring whereas the signals for the pyridine protons are practically the same in all compounds 1-10: (3′-H: 8.2-8.5 ppm, br.d, 3 J = 8 Hz; 4′-H: 7.8-7.9 ppm, br.t, 3 J = 8 Hz; 5′-H: 7.2-7.4 ppm, br.t, 3 J = 4.5-6 Hz; 6′-H: 8.7-8.9 ppm, br.d, 3 J = 4.5-6 Hz).Two planar conformations (I and II) are possible for 3-(2-pyridyl)coumarins. Оцінка протиракової активності похідних ізохіноліну -продуктів рециклізації 3-(2-(тієн-2-іл)тіазол-4-іл)ізокумаринуПредставлено членом-кореспондентом НАН України В.П. Хилею З метою створення нових біоактивних похідних ізохіноліну та встановлення взаємозв'язку структураактивність використано класичне, хоча і малодосліджене для 3-гетарилзаміщених похідних, перетворен ня ізокумарину в похідні ізохіноліну. Цілеспрямовано синтезований 3-(2-(тієн-2-іл)тіазол-4-іл)ізокумарин послідовно трансформовано в ізохінолін-1(2Н)-он (нагріванням при високому тиску у спиртовому розчині амоніаку), 1-хлорізохінолін (шляхом тривалого кип'ятіння ізохінолін-1(2Н)-ону у великому надлишку POCl 3 ), і 1-(4-метилпіперазин-1-іл)-та 1-(морфолін-4-іл)ізохіноліни з 2-(тієн-2-іл)тіазол-4-ільним замісником у положенні 3 (нагріванням 1-хлорізохіноліну із відповідним аміном у розчині ДМФА в присутності поташу).Дослідження протиракової активності 3-(2-(тієн-2-іл)тіазол-4-іл)ізокумарину та похідних від нього ізохінолінів проведені на 60 лініях ракових клітин у Національному інституті раку США. Показано, що 4-(1-(4-метилпіперазин-1-іл)ізохінолін-3-іл)-2-(тієн-2-іл)тіазол призупиняє ріст більшості досліджених ліній клітин і навіть є летальним для деяких із них; зокрема, для ліній COLO 205, НСС-2998 та НТ29 колоректального раку, а також для лінії меланоми M14 та лінії лейкемії К-562. Аналогічна похідна з морфоліновим замісником у середньому має низьку активність, але все ж досить ефективна проти ме ланоми MALME-3M та UACC-257, раку молочної залози MDA-MB-468 і колоректального раку COLO 205. Натомість вихідний ізокумарин, а також-1-хлорізохінолін виявляють дуже низьку цитотоксичність і здатні лише слабко уповільнювати ріст окремих ліній ракових клітин.Опрацьовані підходи до синтезу 1-функціоналізованих ізохінолінів на основі 3-гетарилізокумарину базуються на використанні простих методик та зручних реагентів і забезпечують широке різноманіття цільових продуктів, що є надійним підґрунтям для подальших біологічних досліджень даного класу речовин, які демонструють чітку та сильну залежність рівня протипухлинної активності від характеру замісників у базовій структурі.

Synthesis of a series of 2-(dichloromethyl)pyrazolo [1,5-a][1,3,5]triazines and evaluation in vitro of their anticancer activity against a panel of 60 cell lines derived from nine cancer types, namely leukemia, non-small cell lung cancer, colon cancer, CNS cancer, melanoma, ovarian cancer, renal cancer, prostate cancer, breast cancer. Methods. Organic synthesis; biological tests; spectral methods; statistical methods. Results. In vitro screening of the anticancer activity showed that 5 of 26 tested compounds can effectively inhibit the growth of certain cancer cell lines. Conclusions. New type of N-(2,2-dichloro-1-cyanoethenyl)carboxamides heterocyclization with 1H-pyrazol-5-amines led to the formation of 2-(dichloromethyl)pyrazolo [1,5-a][1,3,5]triazines. Some of these compounds inhibit growth of certain cancer cell lines.
