Резюме. Первичное обследование групп населения с повышенным риском заражения инфекционными заболеваниями (ИЗ) предполагает выполнение ряда анализов с использованием моноспецифических диагностических систем и представляет собой длительную, трудоемкую и дорогостоящую процедуру. Ранее нами разработан мультиплексный тест, позволяющий оперативно и недорого проводить комплексное первичное тестирование. Создан набор, позволяющий одновременно выявлять в препаратах крови антитела к 6 возбудителям ИЗ: вирусу иммунодефицита человека, вирусам гепатитов B и C, цитомегаловирусу, Treponema pallidum и Toxoplasma gondii. Набор основан на мультиплексном дот-иммуноанализе на плоских белковых матрицах (иммуночипах) с использованием конъюгатов коллоидного золота и серебряного проявления. Он позволяет выполнять комплексный анализ при комнатной температуре в течение 70 мин и не требует высокой квалификации оператора. В настоящей статье представлены результаты лабораторных испытаний экспериментального образца мультиплексного набора в сравнении с наборами для иммуноферментного анализа (ИФА) пяти российских производителей с использованием массива из 240 образцов препаратов крови, включающих 200 клинических проб сывороток (плазмы) крови от больных ИЗ и 40 образцов плазмы доноров. Показано, что мультиплексный тест обладает чувствительностью и специфичностью анализа не менее 95%, а оптические сигналы дот-анализа хорошо коррелируют с показателями оптической плотности, полученными с использованием коммерческих наборов для ИФА, и позволяют проводить полуколичественную оценку содержания специфических антител в образце. При этом мультиплексный дот-анализ более оперативен и экономичен, по сравнению с ИФА и может выполняться во внелабораторных условиях. Набор для мультиплексного дотиммуноанализа антител может обеспечить комплексный подход к диагностике ИЗ, значительно облегчить проведение первичного тестирования, сделать его более оперативным и доступным для пациентов.
