Lyme-borreliosis is a zoonotic, infectious disease that has a complex chain of transmission of the pathogen Borrelia burgdorferi sensu lato and includes the relationship between ixodid ticks, vertebrate hosts, humans and companion animals in the environment. The article shows general trends in the prevalence of canine Lyme-borreliosis in Ukraine depending on environmental, climatic and physiographic factors. The results of a comparative cartographic analysis of the prevalence of Lyme borreliosis among domestic dogs in Ukraine are presented by systematizing, mathematical and statistical processing of the data obtained by surveying veterinarians engaged in clinical veterinary practice. The paper includes generalized data on the clinical manifestations, methods of diagnosis and treatment of Lyme borreliosis in dogs. We determined the dependence of the prevalence of Lyme borreliosis in dogs on the types of physical and geographical territories – natural zones of Ukraine. Each of the natural zones differs in types of relief, climatic conditions, soil types, composition of fauna and flora, which affect the epizootic chain of Lyme disease. There is a clear correlation between the incidence of Lyme borreliosis in dogs and the types of natural areas. The highest incidence of Lyme borreliosis in dogs was observed in the forest-steppe zone and the zone of broad-leaved forests. A sharp decrease in the incidence of dogs was recorded in areas of mixed forests, the Ukrainian Carpathians and in the South of Ukraine in the steppe zone. The study also confirmed that the prevalence of Lyme disease among domestic dogs was influenced by the climatic factors, in particular: gross moisture of territories, average annual air temperature and soil temperature. In Ukraine, veterinarians in the vast majority of cases use serological diagnostic methods: immunochromatographic analysis, immunoenzymatic assay, and western blot, which are insufficient, since the presence of antibodies to the Lyme borreliosis pathogen is only a confirmation of the animal's contact with the antigen and may not indicate the presence of the disease in the clinical form. The generalized data on the use of antimicrobial drugs in the treatment of Lyme borreliosis in dogs indicate the predominant use of tetracycline antibiotics and cephalosporins. The majority of veterinarians reported symptoms of Lyme arthritis, somewhat fewer reported Lyme nephritis, neuroborreliosis, Lyme carditis and in rare cases, veterinarians observed erythema at the site of tick bite. About half of the veterinarians in Ukraine observed an increase in the incidence of Lyme disease in dogs, indicating a probable deterioration of the epizootic and epidemiological situation regarding Lyme borreliosis, especially in areas with favourable conditions for the circulation of Lyme borreliosis pathogens in natural and urban ecotopes. The results substantiate the need for the further study of the circulation of Borrelia burgdorferi sensu lato and their ability to cause disease in humans and animals, as well as the need to implement the principles of the One Health concept for the control and management of Lyme borreliosis.
Lyme borreliosis (LB) is the most common tick-borne disease in the temperate climate of the Northern Hemisphere. LB is caused by spirochetes, which are grouped in the complex Borrelia burgdorferi sensu lato, vectors – Ixodidae mites, dozens of species of small mammals and birds actas reservoir hosts, and the role of reservoirs of domestic animals, such as dogs and cats, is not excluded. Although the ecology and epidemiology of the disease have been extensively studied in Europe and North America, there is considerable uncertainty regarding the study of Lyme borreliosis in veterinary medicine. Search, selection andanalysis of scientific data on the research topic were carried out according to acceptable rules for systematic reviews of the literature. The scientometric database Web of Science Core Collection, the database of scientific articles Pub Med and the database Scientific Periodicals of Ukraine were used. Seventy-four scientific articles were used for the article, which contained the necessary set of data and met the set goal. The article highlights the main issues of the etiology of the disease, which describes the characteristics of the pathogen, its properties, genotypic composition of Borrelia, which cause Lyme disease, the spread of pathogenic genotypes of Borrelia in Europe and Ukraine. The connection of Lyme borreliosis spread with ecological factors, climatic changes and anthropogenic impact on biocenoses and biotopes, the role of vectors and reservoir hosts in the spread of LB is described. Data on the prevalence and degree of infection of Ixodes mites with Borrelia, as well as the spread of LB among the population of Europe and Ukraine are presented. The seroprevalence of each of the animal species (dogs, cats, horses, ruminants), clinical manifestations of LB, if documented and indicators of LB prevalence among animals in Europe and Ukraine, as well as currently known treatments, prevention and diagnosis of LB animals are considered. Determination of the infection rate of ixodid ticks with tick-borne borreliosis pathogens and determination of belonging to the pathogenic genotype by PCR is an important component for monitoring, risk assessment and control of the epizootic situation of Lyme borreliosis in different territories. The results of testing and optimization of the internal laboratory protocol of the classical polymerase chain reaction for the identification of Lyme disease pathogens are presented. Eight samples of extracted DNA from ixodid ticks collected from vegetation in the forest park tract "Golendernya", Bila Tserkva, Kyiv region, were examined by classical PCR. Samples were formed from pools of ten tick specimens: seven pools - ticks of the genus I. ricinus and one pool - ticks of the genus D. reticulatus. For detection of borrelia DNA, primer sets were used to detect DNA of Borrelia burgdorferi sensu lato complex; Borrelia burgdorferi and pathogenic borrelia: Borrelia burgdorferi sensu stricto, Borrelia garinii and Borrelia afzelii. The protocol for nucleic acid extraction from ticks was modified using the commercial IndiSpin Pathogen Kit. Optimization of amplification temperature conditions was carried out by the annealing temperature gradient method for each primer pair. Based on the results of the study, internal laboratory protocols for classical PCR using specific oligonucleotide primers were developed. It was found that in each of the pools of I. ricinus and D. reticulatus there were infected tick specimens with the Borrelia burgdorferi sensu lato complex and Borrelia afzelii genus, and also identified the Borrelia burgdorferi sensu stricto genus in one of the pools of I. ricinus and D. reticulatus, DNA of the Borrelia garinii genus was not detected. The developed internal laboratory protocols of classical PCR will be further used to study the infection of ixodid ticks with tick-borne borreliosis pathogens: Borrelia burgdorferi sensu lato, Borrelia burgdorferi sensu stricto and Borrelia afzelii. Ixodes ticks play an important role in the spread of various infectious diseases. Representatives of the Ixodes family carry a large number of pathogens. Zoonotic infections, such as tick-borne borreliosis, tick-borne encephalitis, ehrlichiosis, anaplasmosis, etc., are especially dangerous. Areas of distribution of Ixodes ticks are quite diverse: forests, meadows, shrubs, city parks and various types of green spaces in cities. The study of the quantitative and species composition of ticks in natural foci of infectious diseases is an important component of assessing the epizootiological biopotential and forecasting the risks of infectious diseases in certain areas. In the studies, we studied and compared the distribution of Ixodes ticks based on the population density index of Ixodes ricinus and Dermacentor reticulatus in two different habitats: anthropogenically transformed semi-natural forest environment and meadow-type with pastures. The first – the forest area is located in the forest park tract "Golendernya" in Bila Tserkva, Kyiv region. The second – a natural meadow area is located in the Cherkasy region, Lysyansk United Territorial Community (OTG), village Zhabianka. Collection of Ixodes was performed by the usual method on the flag. The population density index is expressed in relation to the number of ticks per 1 m2 of the surveyed area with a correction factor of 1.2. A total of 12,000 m2 of total habitat area was surveyed, 572 specimens of Ixodes ticks were collected, including 393 Ixodes ricinus and Dermacentor reticulatus 179, respectively. 356 Ixodes ticks were collected in the Golendernia tract, of which 339 were Ixodes ricinus and only 17 were Dermacentor reticulatus. Accordingly, in Golendernia tract the population density index of the surveyed area of 6 thousand m2 Ixodes ricinus is 0.07 specimens/m2 (95.9%) and Dermacentor reticulatus – 0.003 specimens/m2 (4.1%). 216 specimens of Ixodes ticks were collected in the natural meadow biotope, of which 162 – Dermacentor reticulatus and 54 – Ixodes ricinus. The population density index of the surveyed area of 6 thousand m2 in the meadow biotope with mites of the species Dermacentor reticulatus is 0.03 specimens/m2 (75.0%), and with mites of the species Ixodes ricinus – 0.01 specimens/m2. (25.0%). Population density indices between Ixodes ricinus and Dermacentor reticulatus have a statistically significant difference (p <0.01) which is due to the types of habitats.
