Цель исследования -изучить вирулицидную активность декаметоксина в отношении прототипного штамма семейства коронавирусов IBV (infectious bronchitis virus) в условиях in vitro и оценить возможность его использования для неспецифической профилактики коронавирусной инфекции у человека.Материал и методы. При проведении исследований были применены современные и классические вирусологические методы исследования: определение цитотоксического действия декаметоксина на монослой клеточных культур в условиях in vitro; культивирования, накопления и определения инфекционного титра вируса с цитопатическим действием на монослой клеточных культур; оценка вирулицидного действия препарата in vitro; определения ингибирующей активности препарата по степени снижения инфекционного титра вируса в культуре клеток, суспензионный метод, покраска препаратов по Романовскому-Гимзе. Как тест-вирус в работе использовали коронавирус инфекционного бронхита птиц (Infection bronchitis virus -IBV). Вирулицидное действие декаметоксина исследовали в культурах клеток фибробластов эмбрионов кур (ФЭК) и почек сирийского хомяка (BHK-21). Результаты регистрировали методами световой микроскопии.Результаты. Установлено, что инфекционный титр IBV снижается на 2-5 lg по сравнению с контролем в условиях обработки вируса физиологическим раствором декаметоксина в концентрациях 200-1000 мкг / мл. Показано, что в дозе 200 мкг / мл, средство полностью инактивирует 100-1000 инфекционных доз вируса при продолжительности экспозиции 30 минут без проявлений цитотоксического действия при серийных разведениях.Выводы. Выявленные вирулицидные свойства декометоксина в отношении прототипного штамма семьи Coronaviridae в фармакопейного разрешенных концентрациях позволяют рекомендовать его для применения в качестве дезинфицирующего средства для неспецифической профилактики коронавирусных инфекций у взрослых людей.Ключевые слова: четвертичные аммониевые соединения, декаметоксин, вирулицидное действие, цитопатическое действие, коронавирус, вирус инфекционного бронхита IBV.Укр. пульмонол. журнал. 2020, № 2, С. 27-30.
The data on the representative of decamethoxin short-term action on infectious bronchitis virus (IBV) strain H120 used as a human-safe model of SARS-CoV-2 virus are presented. The viral activity was estimated with the use of inverted microscope PrimoVert (Germany) by destructive effect on BHK21 fibroblastic cell line. In vitro results demonstrated that decamethoxin (100 μg/ml) completely inactivated IBV coronavirus strain at exposure of 30 sec and more. At the lowest decamethoxin exposure of 10 sec the antiseptic virucidal activity was 33% and 36% of control at 24 and 48 h of cultivation respectively. Molecular docking analysis indicated the significant similarity of IBV and SARS-CoV-2 main protease (Mpro) structure. Docking studies of decamethoxin interaction with IBV Mpro and SARS-CoV-2 Mpro active centers demonstrated the ligand-protein complexes formation with the estimated binding energy of -8.6, -8.4 kcal/mol and key amino acid residues ASN26, GLY141, GLU187, GLU164, THR24, THR25, ASN142, GLY143, CYS145, HIS164 and GLU166. Yakovenko Abstract The aim of the study is a preclinical evaluation of the antiviral activity of aminocaproic acid (ACA) against the prototype strain IBV (Infectious bronchitis virus) of Coronavirus family in vitro. Material and methods. During the research, modern methods were used to determine the cytotoxic effect of the evaluation on a monolayer of BHK-21 cell culture in vitro; cultivation, accumulation and determination of the infectious titer of IBV by cytopathic action on a monolayer of cell cultures; assessment of the antiviral effect of the drug — the establishment of the inhibitory concentration and the chemotherapeutic index (CTI) of ACA in various modes of drug administration: 2 hours before infection, simultaneously with infection and 2 hours after infection. Results. With the introduction of ACA 2 hours before infection, a decrease in the infectious titer of the IBV virus was not established. The antiviral activity of ACA was detected when the drug was added in 2 modes: simultaneously and 2 hours after infection. The introduction of ACА into the medium for cell cultivation at non-toxic concentrations of 7.91–15.82 mg / ml led to a decrease in the infectious titer of the virus by 1.4–2.0 lg TCD50 / 0.1 ml. The CTI of the ACA was 6 in the indicated concentrations and modes, which is an indicator of its promising potential for further studies of antiviral activity in vivo, including clinical studies. Conclusions. The direct antiviral effect of ACA against the prototype H-120 virus strain from the Coronaviridae family in vitro was revealed. The suppression of viral reproduction with an established low toxicity of the drug, a decrease in the infectious titer of IBV by 1.4–2.0 lg TCD50 / 0.1 ml and with a CTD equal to 6.0, indicate the prospects for further study of the antiviral properties of ACA in clinical trials.
