AnimalsMale Syrian hamsters (110-130 g) were maintained in temperature-controlled rooms and the experiments were performed in accordance with the Gifu University Animal Care and Use Committee and Japanese Department of Agriculture guidelines. However, the effect(s) of intrinsic neurones on vagally mediated contractions of oesophageal striated muscles has not been defined. The present study was designed to investigate the role of intrinsic neurones on vagally evoked contractions of oesophageal striated muscles, using hamster oesophageal strips maintained in an organ bath. Stimulation (30 ms, 20 V) of the vagus nerve trunk produced twitch contractions. Piperine inhibited vagally evoked contractions, while capsaicin and N G -nitro-L-arginine methyl ester (L-NAME) abolished the inhibitory effect of piperine. The effect of L-NAME was reversed by subsequent addition of L-arginine, but not by D-arginine. L-NAME did not have any effect on the vagally mediated contractions and presumed 3 H-ACh release. NONOate, a nitric oxide donor, and dibutyryl cyclic GMP inhibited twitch contractions. Inhibition of vagally evoked contractions by piperine and NONOate was fully reversed by ODQ, an inhibitor of guanylate cyclase. Immunohistochemical staining showed immunoreactivity for nitric oxide synthase (NOS) in nerve cell bodies and fibres in the myenteric plexus and the presence of choline acetyltransferase and NOS in the motor endplates. Only a few NOS-immunoreactive portions in the myenteric plexus showed vanilloid receptor 1 (VR1) immunoreactivity. Our results suggest that there is a local neural reflex that involves capsaicin-sensitive neurones, nitrergic myenteric neurones and vagal motor neurones.

ABSTRACT. Musculature and glands of the esophagus in various wild birds and mammals were examined histologically. Cervical and thoracic esophagi of all birds used (mallard, spot-billed duck, Ural owl and Hodgson's hawk-eagle) were comprised of smooth muscle fibers only. In contrast, esophagi of the nutria, Japanese raccoon dog, common raccoon and Japanese marten consisted largely of striated muscle fibers. In the masked palm civet, Japanese macaque and bottlenose dolphin, esophageal muscle layers consisted of both striated and smooth muscle fibers. Esophageal glands were observed except for the nutria and masked palm civet. These results show a wide variety of the structural composition in the esophagus of wild animals, particularly mammals, examined in this study. Very little is known about esophageal innervation in the hamster. In the present study, we used protein gene product 9.5 (PGP 9.5) to determine immunohistochemically the architectural features of the enteric nervous system in the hamster esophagus. The myenteric plexus consisted of a loose and irregular network of ganglia and interganglionic nerve bundles. The density of the neurons in the myenteric plexus was relatively low (479 +/- 75/cm(2), n = 5), with a preferentially higher density in the upper cervical portion than other parts of the esophagus. Regional differences in the number of PGP 9.5-positive neurons and ganglia were observed. PGP 9.5-immunoreactive fibers in the ganglia often branched, giving rise to expanding nerve endings of laminar morphology resembling intraganglionic laminar endings described in rats and cats. Fine varicose fibers originating from the secondary plexus were occasionally observed near the motor endplates, suggested a dual innervation of the striated muscle. The submucosal plexus was free from ganglionated plexus. A regional difference in the submucosal nervous network was observed. The number of motor endplates in the inner muscle layer was higher than that in the outer muscle layer.
Abstract:A two-year-old, male long-coat Chihuahua presented with slowly progressive grouped papules at only edge of the eyelids. Antimicrobial was not effective, then skin biopsy was performed. Histopathologic examination of the lesion revealed multi-focal pyogranuloma in the dermis. This present dog was diagnosed as having idiopathic stelie granulomatosis, and the lesions were improved with one week course of oral prednisolone 1.0 mg/kg SID dramatically.
