The growth of ochratoxigenic fungus and the presence of ochratoxin A (OTA) in grapes and their derivatives can be caused by a wide range of physical, chemical, and biological factors. The determination of interactions between these factors and fungal species from different climatic regions is important in designing models for minimizing the risk of OTA in wine and grape juice. This study evaluated the influence of temperature, water activity (aw), and pH on the development and production of OTA in a semisynthetic grape culture medium by Aspergillus carbonarius and Aspergillus niger strains. To analyze the growth conditions and production of OTA, an experimental design was conducted using response surface methodology as a tool to assess the effects of these abiotic variables on fungal behavior. A. carbonarius showed the highest growth at temperatures from 20 to 33°C, aw between 0.95 and 0.98, and pH levels between 5 and 6.5. Similarly, for A. niger, temperatures between 24 and 37°C, aw greater than 0.95, and pH levels between 4 and 6.5 were optimal. The greatest toxin concentrations for A. carbonarius and A. niger (10 μg/g and 7.0 μg/g, respectively) were found at 15°C, aw 0.99, and pH 5.35. The lowest pH was found to contribute to greater OTA production. These results show that the evaluated fungi are able to grow and produce OTA in a wide range of temperature, aw, and pH. However, the optimal conditions for toxin production are generally different from those optimal for fungal growth. The knowledge of optimal conditions for fungal growth and production of OTA, and of the stages of cultivation in which these conditions are optimal, allows a more precise assessment of the potential risk to health from consumption of products derived from grapes.
Diversos relatos evidenciam os benefícios de procariotos fixadores de nitrogênio atmosférico no crescimento e na nutrição de muitas espécies vegetais; entretanto, não há, até o momento, nenhum trabalho visando à prospecção desses microrganismos na rizosfera da seringueira (Hevea brasiliensis). Assim, os objetivos deste trabalho foram verificar a ocorrência de bactérias diazotróficas em solos sob plantio de seringueira, assim como em suas raízes, e isolar e caracterizar essas bactérias. Para essa finalidade, coletaram-se amostras de solo e de raízes finas de seringueiras cultivadas no Campus Experimental da Universidade Federal de Lavras (Lavras, MG) para inoculação em meios de cultura semissólidos sem N na forma combinada, de modo a favorecer o crescimento de algumas espécies de bactérias diazotróficas. Foram obtidos 19 isolados nas amostras de solo, e não houve crescimento de bactérias fixadoras de nitrogênio nas culturas com amostras de raízes. A caracterização celular e das colônias desses isolados indicou que 17 deles produzem grande quantidade de exopolissacarídeo elástico, algumas vezes cartilaginoso. Eles são todos Gram-negativos, com formato celular de bastonete, imóveis e com dois glóbulos de poli-β-hidroxibutirato (PBH), um em cada extremidade do bastonete. O sequenciamento do 16S rDNA e sua análise filogenética confirmaram que isolados representativos desse grupo pertencem ao gênero Beijerinckia (B. indica e B. derxii) e que os outros dois isolados Gram-positivos pertencem ao gênero Bacillus. A presença da nitrogenase - a enzima responsável pela fixação biológica do nitrogênio atmosférico (FBN) - foi confirmada por meio da técnica de redução do acetileno. Conclui-se que, no solo sob plantio de seringueira, houve predominância de diazotróficas de vida livre pertencentes ao gênero Beijerinckia (B. indica e B. derxii), não havendo indícios de bactérias endofiticas ou rizosféricas.
ABSTRACT. Brazil has great biodiversity, which is observed in the Cerrado biome of the tropical Brazilian savanna. The objectives of this study were to isolate and identify bacteria from Psychotria hoffmannseggiana with potential cellulase and pectinase production and with antagonistic activity against Aspergillus carbonarius, an Ochratoxin A (OTA) producer. Ripe fruit were collected in the region of Passos City in the preserved Cerrado area in Minas Gerais State, Brazil. Serial dilutions were performed, and the bacteria isolated were biochemically characterized and identified by sequencing. To analyze the production of enzymes, the bacteria were cultivated in CMC and pectinase media. The better enzyme producers were optimized for production. Assays on the antagonistic activity for growth and sporulation were carried out in co-culture (bacteria and filamentous fungi). TLC was performed to verify the mycotoxin production. The predominant microbiota were Gram-negative bacteria belonging to the Enterobacteriaceae family. Some isolates showed potential for enzymatic and antagonistic activity, especially the isolate identified as Lysinibacillus fusiformis. This species was a better producer of cellulases (maximum activity: 103.1 mg glucose min.-1 mg -1 protein). In conclusion, the bacteria isolated from Psychotria hoffmannseggiana showed biotechnological potential for agro-industry and the environmental aspect.Keywords: microorganism, Psychotria hoffmannseggiana, biotechnological application.Potencial enzimático e antagonista de bactérias isoladas de fruto típico do Cerrado em Minas Gerais, Brasil RESUMO. O Brasil tem uma grande biodiversidade no bioma Cerrado típico de savanas tropicais. Os objetivos deste estudo foram isolar e identificar bactérias celulolíticas e pectinolíticas e com atividade antagonista contra contra Aspergillus. carbonarius produtores de ocratoxina a partir de Psychotria hoffmannseggiana. Frutos maduros de P. hoffmannseggiana foram coletadas na região da cidade de Passos em área preservada de Cerrado em Minas Gerais, Brasil. Realizou-se diluições decimais e as bactérias isoladas foram caracterizados bioquimicamente e identificadas por seqüenciamento. Para a produção de enzimas, as bactérias foram cultivadas em CMC e MP5 e MP7. Os isolados com melhor produção enzimática foram submetidos ao processo de otimização. A atividade antagonista foi realizada em co-cultura (bactérias e fungos filamentosos) e avaliou-se o crescimento e a esporulação. TLC foram realizados para verificar a produção de micotoxinas. A microbiota predominante foi Gram negativas pertencem à família Enterobacteriaceae. Alguns isolados apresentaram potencial para a atividade enzimática e antagonista, especialmente o isolado identificado como Lysinibacillus fusiformis.
