The bacterium Flavobacterium columnare was recovered and identified as the aetiological agent causing freshwater columnaris infection in farmed striped catfish Pangasianodon hypophthalmus (Sauvage) fingerlings that had suffered high mortality rates within commercial hatchery ponds in Vietnam. The gross clinical signs were typical of columnaris-infected fish. Histological examination found numerous Gram-negative, filamentous bacteria present on the skin, muscle and gill tissues of affected fish. The yellow-pigmented bacteria were isolated and identified as F. columnare using primary, biochemical and PCR methods. An experimental immersionchallenge study with 2 strains was also performed. It fulfilled Koch's postulates and showed a median lethal concentration (LC 50 ) of 4.27 × 10 5 and 1.66 × 10 6 cfu ml −1 for the F. columnare strains FC-HN and FC-CT, respectively. To the best of our knowledge this is the first report of freshwater columnaris infection in P. hypophthalmus. An experimental bacterial challenge study was also completed to confirm Koch's postulates and to determine the median lethal concentration (LC 50 ) for 2 different F. columnare strains on P. hypophthalmus. MATERIALS AND METHODSA total of 83 Pangasianodon hypophthalmus (weighing 3 to 20 g) were collected during natural disease outbreaks from 5 hatchery and 7 earthen pond farms located in Vietnam. Histology samples of the skin, gill, kidney, liver and spleen were aseptically removed, placed in 10% (v/v) neutral buffered formalin and processed for routine wax sections. Four-micron thick sections were cut and stained with haematoxylin and eosin (H&E) and Giemsa (Roberts 1989). These were examined under light microscopy. Bacterial isolation was performed where samples were taken aseptically from gills, skin, liver, kidney and spleen of the affected animals directly onto peptone, yeast extract, salt and agar (PYES) (Triyanto & Wakabayashi 1999) and tryptone soy agar (TSA; Oxoid) and then incubated at 28°C. Inoculated agar plates were checked daily up to 4 d post-inoculation for the presence of yellow-pigmented colonies, and primary identification tests included Gram stain, oxidase, oxidation/fermentation (O/F) of glucose, and motility, as described in Frerichs & Millar (1993). Effect of water-soluble chitosan (WSC) on green mould disease caused by Penicillium digitatum P4 was investigated in vitro and in vivo. Results of the experiments showed that P. digitatum P4 growth was much inhibited by WSC and inhibitory effect increased as WSC concentration increased. In vitro test results indicated that complete inhibition of spore germination and mycelial growth were observed in the medium containing 1 mg/ml WSC. In the in vivo study, WSC treatments significantly reduced disease incidence and lesion diameter of green mould disease on orange fruits. After 8 days of inoculation, the lesion diameter of orange fruits coated by 0.5%, 1%, 1.5% and 2% WSC were 5.49 cm, 5.01 cm, 3.22 cm and 1.87 cm, respectively, which smaller than that of control fruits. Biochemical experiments demonstrated that the activities of the main defense-related enzymes in flavedo tissue including chitinase and β-1,3-glucanase were enhanced by both P. digitatum P4 infection treatment and challenged with P. digitatum P4 and treated with WSC treatment. However, these enzymes of samples inoculated with P. digitatum only were lower than samples inoculated with P. digitatum P4 and treated with 2% WSC concentration. These findings suggest that the in vitro and in vivo effects of WSC on controlling green mould disease may be associated with direct antifungal activities and the elicitation of biochemical defense responses in fruit.
Ngành công nghiệp dầu khí đang trải qua cuộc khủng hoảng giá dầu lần thứ 3 trong vòng 12 năm. Hoàn toàn khác so với các cuộc khủng hoảng giá dầu trước đây, cuộc khủng hoảng “kép” năm 2020 diễn ra khi nguồn cung vượt quá nhu cầu và đại dịch Covid-19 bùng phát. Mức độ ảnh hưởng và thời điểm kết thúc của cuộc khủng hoảng này khó dự báo, tuy nhiên có thể thấy rằng ngay cả khi có thay đổi cơ bản, ngành công nghiệp dầu khí sẽ khó có thể quay lại thời kỳ phát triển mạnh mẽ như trước đây. Mặc dù đang trải qua cuộc khủng hoảng lớn về giá, dầu thô vẫn đang là nhiên liệu hàng đầu thế giới. Ngành công nghiệp dầu khí cần phải có các bước chuyển đổi mạnh mẽ để điều hành hoạt động an toàn và có lợi nhuận trong điều kiện khó khăn nhất. Bài báo đánh giá lịch sử biến động giá dầu thô gần đây, phân tích ảnh hưởng của khủng hoảng giá dầu thô và đại dịch Covid-19 đến sản xuất kinh doanh năm 2020 của Tập đoàn Dầu khí Quốc gia Việt Nam, từ đó đề xuất các giải pháp để Tập đoàn phát triển bền vững.
Chủng Penicillium digitatum P4 phân lập từ các mẫu cam có vết bệnh mốc xanh điển hình được sử dụng để nghiên cứu khả năng kháng nấm của chế phẩm chitosan ở điều kiện in vitro và in vivo. Kết quả các thí nghiệm đều cho thấy P. digitatum P4 rất nhạy cảm với chitosan, hiệu lực ức chế tăng khi nồng độ chitosan tăng. Ở các thí nghiệm in vitro, ảnh hưởng của chitosan lên P.digitatum P4 được đánh giá thông qua đường kính tản nấm và sinh khối khô. Giá trị EC50 (the 50% effective concentration value, giá trị nồng độ có khả năng ức chế ít nhất 50% sự phát triển của nấm) đạt được 52,67% và 50,18% trên hai môi trường tương ứng là PDA và PDB khi xử lý với chitosan nồng độ 0,05%. Ngoài khả năng ức chế sự sinh trưởng của nấm mốc xanh, chitosan còn làm biến đổi hình thái của chúng khi nuôi cấy trên môi trường PDA. Mẫu cam sau khi lây bệnh nhân tạo bằng P.digitatum P4 với nồng độ 105 bào tử/ml được xử lý chitosan với các nồng độ 0,5%, 1%, 1,5% và 2% cho thấy tỷ lệ bệnh và mức độ tiến triển bệnh thấp hơn so với mẫu đối chứng (xử lý với nước cất). Nồng độ ức chế EC50 nằm trong khoảng nồng độ 1,5% đến 2,0% với hiệu lực ức chế lần lượt là 44,38% và 65,21%.
Sử dụng chitosan ở các nồng độ khác nhau từ 0% (đối chứng), 0,5%, 1%, 1,5%, và 2% đã được khảo sát nhằm duy trì chất lượng và kéo dài thời gian bảo quản cà chua trong điều kiện phòng. Nồng độ chitosan sử dụng ở mức 0,5% không cho thấy được tác dụng rõ rệt về mặt duy trì chất lượng và kéo dài thời gian bảo quản cà chua so với mẫu đối chứng. Trong khi đó, nồng độ chitosan ở mức 2% lại khá cao, có tác dụng thúc đẩy quá trình chín diễn ra nhanh và ức chế sự hô hấp của quả. Quả cà chua được tạo màng với nồng độ chitosan 2% đã chín hoàn toàn sau 10 ngày bảo quản và sự hao hụt dinh dưỡng cũng diễn ra nhanh chóng trong thời gian sau đó và quả có dấu hiệu hư hỏng chỉ sau 20 ngày bảo quản. Ngược lại, nồng độ chitosan ở 1% và 1,5% lại cho tác động tích cực đến duy trì chất lượng và kéo dài thời gian bảo quản cà chua. Quả đạt độ chín hoàn toàn và các chỉ tiêu dinh dưỡng khảo sát đạt giá trị tốt nhất sau 15 ngày bảo quản và chất lượng cà chua được duy trì ở mức chấp nhận được cho tiêu dùng đến ngày thứ 25, quả chỉ có dấu hiệu mềm và hư hỏng ở ngày thứ 30 sau bảo quản.
