In vitro effect of the Melaleuca alternifolia, Lavandula angustifolia and Mentha piperita essential oils (EOs) against Ichthyophthirius multifiliis and in vivo effect of M. alternifolia for treating ichthyophthiriasis in one of the most important South American fish, Piaractus mesopotamicus (Holmberg), were evaluated. The in vitro test consisted of three EOs, each at concentrations of 57 μL L(-1) , 114 μL L (-1) , 227 μL L(-1) and 455 μL L (-1) , which were assessed once an hour for 4 h in microtitre plates (96 wells). The in vitro results demonstrated that all tested EOs showed a cytotoxic effect against I. multifiliis compared to control groups (P < 0.05). The in vivo treatment for white spot disease was performed in a bath for 2 h day(-1) for 5 days using the M. alternifolia EO (50 μL L (-1) ). In this study, 53.33% of the fish severely infected by I. multifiliis survived after the treatment with M. alternifolia (50 μL L (-1) ) and the parasitological analysis has shown an efficacy of nearly 100% in the skin and gills, while all the fish in the control group died. Furthermore, the potential positive effect of M. alternifolia EO against two emergent opportunistic bacteria in South America Edwardsiella tarda and Citrobacter freundii was discussed.

The utilization of probiotic bacteria have been widely tested and applied in intestinal microflora modulation, through competitive exclusion of the pathogenic bacteria. Aeromonas hydrophila is a gram-negative bacterium, responsible for fish outbreaks in farms around the world, and is one of the major loss causes for neotropical fish farmers. 660 pacus (67 ± 7 g) were distributed in 20 tanks (n = 33), constituting five groups (four tanks for each treatment): four groups were fed with different levels (2, 4, 8 and 16 g kg −1) of Bacillus cereus and Bacillus subtilis (1:1, 10 8 CFU g −1), and the fifth group was fed with a control diet (without probiotic). Pacus fed with probiotic showed increment in the ROS production associated to elevated neutrophil and monocyte counts and increased phagocytic activity without affecting the growth parameters. Probiotic fed fish presented higher survival rates, subjected to an i.p. challenge with 10 8 CFU mL −1 of A. hydrophila. The results demonstrated a dose response effect and the ideal level of the probiotic (Bacillus cereus and Bacillus subtilis, 1:1, 10 8 CFU g −1) in P. mesopotamicus diets was around 8 g kg −1 , as the highest survival rates and immunological responses were found in groups of fish fed with this diet. In addition, too much probiotic should be avoided, as pacus fed diet with 16 g kg −1 showed partial suppression of these responses.

SummaryThe aim of this study was to describe the effect of temperature on the fertilization, early developmental stages, and survival rate of two Neotropical catfishes Pimelodus maculatus and Pseudopimelodus mangurus. After fertilization, the eggs were incubated at 22°C, 26°C, and 30°C, which resulted in fertilization rates of 96.95 ± 1.79%, 98.74 ± 0.76%, and 98.44 ± 0.19% for P. maculatus and 96.10 ± 1.58%, 98.00 ± 0.63%, and 94.60 ± 2.09% for P. mangurus, respectively. For P. maculatus, hatching occurred after 22 h 30 min post-fertilization at 22°C, 16 h 30 min at 26°C, and 11 h 20 min at 30°C, and the hatching rates were 43.87 ± 7,46%, 57.57 ± 17.49%, and 53.63 ± 16.27%, respectively. For P. mangurus, hatching occurred after 28 h 30 min post-fertilization at 22°C and 17 h 30 min at 26°C with respective hatching rates of 45.4 ± 21.02% and 68.1 ± 12.67%. For this species, all embryos incubated at 30°C died before hatching. Additionally, for P. maculatus, the larvae from the lower (22°C) and higher temperatures (30°C) presented increased abnormality rates, as observed in the head, tail and yolk regions. The lowest abnormality rate was detected at 26°C, which was considered the optimal incubation temperature for both species. The developed protocol enables the manipulation of embryonic development, which is important for the application of reproductive biotechniques, including chimerism and chromosome-set manipulation. The data obtained here are also important for the surrogate propagation of this species as P. mangurus was recently categorized as an endangered fish species.

In this experiment, the beneficial effects of mannan-oligosaccharide (MOS) on immunology and intestinal microbiology of Nile tilapia juveniles was demonstrated. Prior to this, three levels of MOS in Nile tilapia diets (1, 8, and 15 g.kg −1) were tested, and hematological parameters, serum lysozyme, and intestinal microbiology were analyzed. The fish blood was sampled at day zero (basal sample) and after 45 days of trial, and the intestinal microbiota was evaluated at the end of the experiment. After 45 days of trial, fish fed 8 and 15 g.kg −1 of MOS presented an increase in both aerobic and lactic acid bacteria numbers in their guts. The MOS feeding also increased the counts of total leukocytes, monocytes, and lymphocytes of fish, but a decrease in neutrophils was also observed. Additionally, the serum lysozyme was higher in all fish fed MOS. The dietary MOS is able to modulate the intestinal microbiota, increasing the number of beneficial bacteria, and immunostimulates the Nile tilapia juvenile, giving rise to white blood cells and serum lysozyme.
