Abstract:Targeting apoptotic machinery is growing as an approach for targeted anticancer therapy. Evading apoptosis is one of the main cancer hallmarks. Several approaches were adopted for targeting apoptotic pathways. Targeting
IAPs (inhibitors of apoptosis proteins) is the focus of this review. Smac (second mitochondria-derived activator of caspases) is endogenous inhibitor of IAPs, thus it is one of the major proapoptotic endogenous proteins. SeveralSmac mimetics were synthesized and biologically evaluated as anticancer agents. Herein, the history of development of Smac mimetics along with the recent development in this field is briefly discussed.
