Novel heterocyclic dichloronaphthoquinone derivatives have been synthesized by chlorine atom substitution in 2,3-dichloro-1,4-naphthoquinone to pyrazole or pyrimidine fragments. The structures of these compounds have been confirmed by FT-IR, ESI-MS, 1 H-NMR, 13 C-NMR and elementary analysis. Synthesized compounds were evaluated for their anticonvulsant action in a pentylenetetrazole (PTZ)-convulsion model and antidepressant activity in the forced swimming test (FST). All naphthoquinone derivatives at a dose 100 mg/kg indicated anticonvulsant effect in PTZ-induced test at 3 h and 24 h after oral administration. In addition, these compounds possessed prolonged antidepressant properties significantly reducing the duration of immobility time when compared to the reference drug amitriptyline.
Провести дослідження антимікробної та фунгіцидної активностей амінопіразольних похідних 2,3-дихлоро-1,4-нафтохінону та спрогнозувати їхню гостру токсичність. Матеріали та методи дослідження. Протимікробну активність гетероциклічних амінопохідних нафтохінону 3a-d вивчали шляхом дифузії речовини в агар на твердому поживному середовищі та методом серійних розведень. Гостру токсичність для гризунів визначали методом моделювання QSAR, реалізованим в програмному забезпеченні GUSAR. Результати. У роботі досліджено антимікробну та фунгіцидну активності нових гетероциклічних амінопохідних нафтохінону, а також проведено визначення їх in silico гострої токсичності для щурів за чотирма типами введення субстанції. Висновки. Дослідження амінопіразольних похідних нафтохінону дозволило виявити сполуки, які проявляють високу антимікробну активність по відношенню до тест-культури Candida tenuis, а саме: 2-хлоро-3-((1-метил-1Н-піразол-4-іл)аміно)нафтален-1,4-діон 3a та 2-хлоро-3-((1-метил-1Н-піразол-3іл)аміно)нафтален-1,4-діон 3b. Встановлено, що усі синтезовані сполуки проявляють вибіркову бактеріо-і фунгістатичну активності. Expanding the range of herbal drugs is among the priorities of the modern pharmacy. The analysis presented in this article is drawn from a broader qualitative study examining the composition of Anemone nemorosa L. chloroform fraction. The study revealed 38 compounds, and 32 of them were identified. The investigated lipophilic extract has an antimicrobial activity both in terms of gram-positive and gram-negative microorganisms. The maximal antimicrobial action of the investigated extract was shown to Escherichia coli. The significant content of biologically active substances in the lipophilic volatile fraction of Anemone nemorosa indicates the prospects for further study.
Composite preparations based on heterocyclic amine-containing naphthoquinone derivatives and surfactant rhamnolipids were obtained. The intensity of the processes of lipid peroxidation (LPO), oxidative modification of proteins (OMB) and radical-absorbing activity against 1,1-diphenyl-2-picrylhydrazyl (DFPG) was studied. Compounds exhibiting high antioxidant activity against LPO and OMB processes were identified, namely: 2-[(6-(4-fluorophenyl-5-oxo-2,5-dihydro-1,2,4-triazin-3-yl) phenyl)amino]naphthalene-1,4-dione 1d and its composite preparation with rhamnolipid 1d + RL. It was found that all synthesized compounds show antioxidant activity in LPO processes.
Composite preparations based on rhamnolipid biosurfactants and N-containing heterocyclic 1,4-naphthoquinone derivatives were obtained. The formation of the compositions was confirmed by UV spectroscopy, dynamic light scattering and changes in the surface tension of the compositions in comparison with the original solutions. Physicochemical and biological properties of composite drugs have been studied. The anticonvulsant effect was assessed using the pentylenetetrazole (PTZ) model. Doses of PTZ for induction of clonic-tonic seizures (DCTC) and tonic extension (DTE) were calculated relative to control. The effect of rhamnolipid on increasing the permeability of cell membranes for compounds 1a-e was evaluated indirectly by studying anticonvulsant activity. Compound (2-chloro-3-((3-(p-tolyl)-1H-pyrazol-5-yl)amino) naphthalene-1,4-dione) (1b) in combination with rhamnolipid was found to have a higher anticonvulsant activity DCTC and DTE averaged 239% and 244%, respectively, which may indicate an improvement of the penetration of the compound when in complex with rhamnolipid into the cell and as a result, an increased anticonvulsant activity of the composite drug.
