The genus Rhinella is one of the most diverse groups of bufonid toads, currently composed by 93 valid species and naturally distributed throughout different Neotropical ecoregions. Here, we analyze nine Brazilian populations of toads representing species of the Rhinella margaritifera and Rhinella marina groups. These new data include the first description of the R. hoogmoedi and R. proboscidae karyotypes, as well as other taxonomically unresolved forms. Chromosomal analysis of the populations revealed pronounced chromosomal uniformity (2n=22), including the diploid number and chromosomal morphology. Three different NOR-bearing chromosomes were identified: in the subterminal region of pair 10q in R. hoogmoedi , Rhinella sp. 1 and Rhinella sp. 2, in subterminal region of 7p in R. proboscidae and Rhinella cf. margaritifera while in R. henseli and R. icterica was detected in interstitial region of 7p. Karyotypic uniformity of the genus permits the inference of interspecific chromosome homologies and evolutionary changes in the NOR-bearing chromosome may represent an informative character in species group level. The review of the cytogenetic data of the Rhinella species together with the new karyotypes reported here contributes to the understanding of the chromosomal evolution of these toads, which karyotypes are highly conserved despite the ample distribution of many forms.
A biocompatibilidade dos materiais utilizados na ortodontia ainda é motivo de preocupação, especialmente pela dificuldade em avaliar cada material separadamente durante os tratamentos. Diante disso e da escassez de pesquisas sobre a genotoxicidade das resinas utilizadas na fase de contenção ortodôntica, o presente estudo teve como objetivo avaliar “in vivo” a genotoxicidade destes materiais. A amostra foi composta por 27 pacientes (15 mulheres e 12 homens), com idades entre 13 e 38 anos, iniciando a fase de contenção mediante o uso de placa removível superior e fio colado inferior. Os mesmos foram submetidos à coleta de células da mucosa bucal, antes das contenções e depois de um, seis e nove meses da instalação e uso contínuo das mesmas. As células coletadas foram avaliadas pelo teste de micronúcleos. Para análise estatística, utilizou-se o teste Anova e Tukey, ao nível de significância de 5%. Os resultados demonstraram não haver diferença estatística entre as médias das coletas para micronúcleos. Foi encontrado um aumento significativo de células com cromatina condensada e cariorrética entre a primeira e segunda coleta, que demonstra morte celular, mas tornou-se não significativa nas coletas de seis e nove meses, indicando uma capacidade de reparação celular. Quando aplicado o teste não paramétrico de Spearman foi observada uma fraca, mas ainda importante correlação entre capacidade de reparação celular e a idade, mostrando que a medida em que a idade aumenta, reduz a capacidade de reparação das células. Pode-se observar que não houve aumento de danos ao DNA entre as coletas realizadas, entretanto o aumento significativo de células com cromatina condensada cariorrética, sugeriu um aumento de morte celular relacionada com a exposição.
A Síndrome de Edwards é uma condição causada pela presença de trissomia do cromossomo 18. Malformações congênitas e retardo no desenvolvimento neuropsicomotor são manifestações clínicas normalmente presentes, embora não sejam exclusivas da síndrome. Esta anomalia genética resulta, normalmente, na morte prematura do indivíduo portador. Daqueles que chegam a termo, cerca de 60% vem a óbito na primeira semana. Semelhante às trissomia do 21 e do 13, a idade materna avançada está relacionada à ocorrência da trissomia, sendo observada em cerca de 63% dos casos. O objetivo do estudo foi relatar um caso de sobrevida de paciente com síndrome de Edwards, observado na APAE de Passo Fundo/RS. O projeto foi aprovado pelo CEP/UPF (59461716.4.0000.5342). Após entrevista e autorização da responsável, foi feita a coleta de sangue periférico da paciente, seguido pela cultura temporária de linfócitos em meios de cultura RPMI 1640, suplementado com soro fetal bovino e estimulado com fitohemaglutinina, para a obtenção de metáfases. As preparações cromossômicas foram submetidas ao bandamento G e 30 metáfases analisadas. Os genitores não são consanguíneos e a idade materna, no momento da gestação, era avançada. Não há relatos de doenças genéticas na família. A.R.B.D., sexo feminino, nasceu de parto cesáreo, com IG de 37 semanas, peso de 2.400g, e está com 53 meses de idade. Apresenta deficiência motora e mental grave, dificuldades visuais, estrabismo, resposta diminuidas ao som, choro fraco, dificuldades de sucção, crânio com diâmetro bifrontal diminuido, nariz e boca pequenos, palato ogival e estreito, orelhas hiperplásicas com implantação baixa, redundância de pele e hipertricose. Na análise das 30 metáfases, detectou-se trissomia livre do cromossomo “18” 47, XX,+18. A.R.B.D mostra-se como um caso raro de Síndrome de Edwards, por apresentar sobrevida prolongada. De acordo com a literatura, o sexo feminino apresenta maior longevidade em relação à esta trissomia. A idade materna avançada pode ter sido um dos fatores responsável da alteração cromossômica.
