RESUMONeste trabalho, objetivou-se avaliar a performance do teste multivariado de normalidade de Shapiro-Wilk implementado no R comparando o seu desempenho com os testes de assimetria e curtose de Mardia (1970Mardia ( , 1974Mardia ( , 1975 utilizando simulação Monte Carlo. Foram mensuradas e comparadas as taxas de erro tipo I e poderes dos testes. Vários dos diferentes métodos de estimação e testes de hipóteses existentes foram formulados sob a suposição de que a amostra aleatória tenha sido extraída de uma população normal. Da mesma forma que ocorre no caso univariado, têm-se inferências multivariadas sobre parâmetros que são vetores ou matrizes, pois a estatística multivariada é lida com observações simultâneas de várias variáveis. Os testes de hipóteses e os métodos de estimação são, em geral, baseados na suposição de normalidade multivariada da amostra aleatória.A necessidade de testar a hipótese de normalidade multivariada fica evidenciada quando o pesquisador pretende avaliar se as condições pressupostas para a validade da inferência que irá realizar foram atendidas. A existência de um teste adequado, com propriedades ótimas sempre foi questionada. Um destes testes de normalidade é baseado nos desvios de assimetria e curtose (MARDIA, 1970(MARDIA, , 1974(MARDIA, , 1975. Alguns problemas podem ser destacados desta abordagem. A princípio, a não rejeição da hipótese de distribuição simétrica e mesocúrtica não garante que a distribuição seja realmente normal. Existem contra-exemplos de distribuições não-normais simétricas e mesocúrticas. A segunda dificuldade é a propriedade assintótica das estatísticas dos testes. As distribuições destas estatísticas são apenas assintoticamente válidas, o que dificulta a aplicação dos testes em conjuntos de dados de tamanhos relativamente pequenos.Um teste multivariado para normalidade foi proposto por Royston (1983) generalizando o teste univariado de Shapiro-Wilk para o caso multivariado. A justificativa apresentada para a proposição deste teste refere-se ao fato de que muitos testes multivariados possuem distribuições matematicamente intratáveis sob a hipótese nula para a estatística. The diversity analysis and screening of major genes using genomewide association studies (GWAS) for quantitative traits have been exploited in several grass crops, but in maize it has been neglected. This study was conducted to evaluate the genetic diversity of a collection of lines and the potential of genomic analysis as an initial screening tool. For this purpose, a collection of 470 maize inbred lines were genotyped with 23,153 Dart‐seq markers. The genomewide analysis of ear weight was used aiming to perform an initial screening in our germplasm. The diversity analysis allowed the lines to be separated into distinct groups, and most of the lines were in agreement with previous pedigree information. The GWAS revealed ten regions potentially associated with the ear weight trait, one of which was related to drought stress. The results of a multiple marker analysis confirmed four of these regions, presenting a predictive accuracy of 0.6. Based on the molecular data and the results of GWAS, it is possible to infer the potential use of this genetic background for breeding.
With the growth in the transgenic market, fast and economically viable methodologies are necessary for undertaking transgene detection tests, both for identification of contamination in seeds and in grain. Seeds from commercial conventional GNZ 2004, and transgenic VT-Pro (MON89034), Roundup Ready (NK603) and Herculex (TC1507) maize cultivars were used. In order to simulate different levels of contamination, the transgenic seeds were mixed with conventional seeds at levels of 0.2%, 0.4%, 1.0% and 1.6% for VT-Pro, and 0.2%, 0.5%, 0.8% and 1.2% for Roundup Ready and Herculex. The lateral flow membrane strip test was performed in the whole seed, endosperm and embryo. For evaluation of the specificity of the technique in detection of the TC1507 event by means of the conventional PCR technique, seeds of the commercial maize hybrid GNZ 2004 were used as negative control, and the maize hybrid 2B655Hx as positive control. In order to simulate different levels of contamination, transgenic seeds were mixed with conventional seeds at the levels of 10%, 5%, 1%, 0.5% and 0.1%. Seeds from each sample were crushed, and then DNA extraction was performed by the CTAB 2% method. Using the immunochromatographic strip, it was possible to evaluate the expression of proteins related to the VT-Pro, Roundup Ready and Herculex events when whole seeds were used at the 0.2% level of contamination, whereas by the conventional PCR technique, it was possible to detect the TC1507 event in samples with 1% contamination.Index terms: Fast testing, contamination, specificity, transgenesis. RESUMOCom o crescimento do mercado de transgênicos, fazem-se necessárias metodologias rápidas e economicamente viáveis para detecção de transgenes, tanto para a identificação de contaminação em sementes quanto em grãos. Foram utilizadas sementes de cultivares comerciais de milho GNZ 2004 convencional, VT-Pro (MON89034), Round up Ready -RR (NK603) e Herculex (TC1507). Afim de simular diferentes níveis de contaminação, sementes transgênicas foram misturadas às sementes convencionais, nos níveis de 0,2%, 0,4%, 1,0% e 1,6%, para VT-Pro e 0,2%, 0,5%, 0,8% e 1,2%, para RR e Herculex. O teste de tiras foi realizado em semente inteira, endosperma e embrião. Para a avaliação da especificidade da técnica na detecção do evento TC1507, por meio da técnica de PCR convencional, foram utilizadas, como controle negativo, sementes do híbrido comercial de milho GNZ 2004 e, como controle positivo, o híbrido de milho 2B655Hx. Com a finalidade de simular diferentes níveis de contaminação, sementes transgênicas (TC1507) foram misturadas às sementes convencionais, nos níveis de 10%, 5%, 1%, 0,5% e 0,1%. Sementes de cada amostra foram trituradas e foi realizada a extração do DNA pelo método CTAB 2%. Utilizando-se a tira imunocromatográfica, foi possível avaliar a expressão de proteínas referente aos eventos VT-Pro, RR e Herculex, quando da utilização de sementes inteiras com níveis de contaminação de 0,2%. ABSTRACT. The main objective of a maize breeding program is to generate hybrid combinations that are more productive than those pre-existing in the market. However, the number of parents, and consequently the number of crosses, increases so rapidly that the phenotypic evaluation of all the possible combinations becomes economically and technically infeasible. In this context, predicting the performance of the most promising genotypes may increase the genetic gains with increased selection intensity and reduced breeding cycles. Thus, the present study aimed to use the total effects of associated markers method to predict genomic breeding values (GBVs) via cross-validation and by using different imbalance levels (10, 30, 50, and 70%). A set of 51 genotyped strains was used with 79 microsatellite markers and 273 hybrids that were generated by a partial diallel. A total of 186 and 272 hybrids were analyzed in the experiments within the southern and central regions of Brazil, respectively. The GBVs were, thus, predicted for each location in both the regions, and for training in one region and validation in another region. The correlation between the predicted and observed GBVs ranged from 0.48 to 0.91, depending on the imbalance level and the region analyzed. Overall, the results obtained in the present study were promising, particularly considering that a small number of markers were used and that the training and predictions occurred in the very distinct regions of southern and central Brazil.
