RESUMO -A lignina extraída da planta forrageira pelo reagente brometo de acetila (lignina brometo de acetila -LBrAc), para ser empregada como padrão de referência no método espectrofotométrico para a determinação quantitativa da lignina, não pode originar-se da planta intacta, devido à interferência de diversas outras substâncias, que absorvem no mesmo comprimento de onda que a lignina; aconselha-se, portanto, que a extração da lignina seja realizada a partir de um dos seguintes compostos: parede celular propriamente dita (PC), fibra em detergente neutro (FDN) ou fibra em detergente ácido (FDA). Os rendimentos das extrações das ligninas, bem como o comportamento espectrofotométrico das mesmas, foram avaliados nestes três compostos provenientes de diversas plantas forrageiras. Na média, os rendimentos das extrações de LBrAc foram relativamente uniformes entre si, entretanto, como a concentração de FDA foi a menor entre todas as preparações (devido principalmente à solubilização da hemicelulose) e como esta não apresentou maior capacidade extrativa da lignina, o rendimento de LBrAc proveniente da FDA, com base na planta inteira, foi o menor observado. A absorção da luz ultravioleta das diversas ligninas foi desigual, revelada pelos espectrogramas traçados, de tal modo a sugerir que a preparação de parede celular adotada para o isolamento da lignina deve ser a mesma para a determinação final da concentração de lignina.Palavras-chave: fibra em detergente neutro, fibra em detergente ácido, lignina brometo de acetila, método espectrofotométrico, parede celular Yield and Spectrophotometric Pattern of Lignin Extracted from Cell Wall, Neutral Detergent Fiber or Acid Detergent FiberABSTRACT -The lignin extracted from the forages by the acetyl bromide reagent (acetyl bromide lignin -AcBrL) to be used as a reference standard for the quantitative spectrophotometric determination of lignin could not originate from intact plant because of interference of other substances, which absorb ultraviolet light at the same wavelength of lignin. It is then recommended that lignin extraction be performed in one of the following materials: cell wall itself (CW), neutral detergent fiber (NDF), or acid detergent fiber (ADF). The lignin extraction yields as well as the spectrographic pattern of those were evaluated in these three compounds that came from diverse forage plants. On the average, extraction yields of AcBrL were fairly uniform among them; however as the concentration of ADF was the lowest among all preparations (mainly due to hemicellulose solubilization) and as far this preparation did not exhibit higher lignin extraction capacity, the AcBrL concentration from ADF on a whole plant basis was the lowest observed. The ultraviolet light absorption from the diverse lignin was unequal, as revealed by the graphic spectrograms, which would suggest that the cell wall preparation used for lignin isolation must be the same for the final determination of lignin concentration.
RESUMO -Três métodos analíticos para a estimativa do conteúdo de lignina na parede celular, em diferentes frações vegetais, de sete espécies forrageiras, foram comparados. Os métodos empregados foram: lignina em detergente ácido (LDA), lignina permanganato de potássio e lignina solúvel em brometo de acetila (LSBA). Para o método espectrofotométrico (LSBA), foi utilizado como padrão de referência a lignina extraída da planta através do próprio reagente brometo de acetila. Não houve concordância entre os três métodos, sendo que, no geral, o método da LSBA mostrou valores mais elevados que os outros dois métodos e o método da LDA, menores valores. Na maioria das amostras, a concentração de lignina foi superior para a parede celular oriunda de planta madura em relação àquela proveniente de planta jovem, assim como o teor de lignina foi maior para a fração colmo em relação à fração folha. As leguminosas tenderam a apresentar teores mais elevados de lignina pelos métodos gravimétricos que as gramíneas, independentemente do estádio de maturidade. Entretanto, essa tendência não foi observada para o método LSBA, particularmente no estádio maduro. Preparações de parede celular foram fermentadas in vitro e os dados correlacionados com os teores de lignina obtidos por intermédio dos três métodos. O método LSBA exibiu os menores coeficientes de correlação, para a maioria das amostras, sugerindo que este método deve ser melhor avaliado.Palavras-chave: digestibilidade, lignina brometo de acetila, lignina detergente ácido, lignina permanganato de potássio, método espectrofotométrico, parede celular Correlation Between In vitro Cell Wall Digestibility and Three Analytical Methods for Quantifying LigninABSTRACT -Three methods were compared in the estimate of lignin cell wall contents in different fractions of seven forage species. The methods for assaying lignin were: acid detergent lignin, permanganate lignin and acetyl bromide soluble lignin. The lignin extracted from the forage sample through the acetyl bromide reagent was employed as a reference standard for the spectrophotometric method. There was no agreement among the three methods; in general, the ABSL yielded higher values than the two other analytical procedures while the ADL method yielded the lowest values. For most samples, lignin concentration was higher for the mature plant cell wall in relation to the young plant; similarly, lignin concentration was higher for the stem fraction as compared to the leaf fraction. Legumes tended to show higher lignin values obtained through the gravimetric methods than grasses, regardless of maturity stage; however, this trend was not observed for the ABSL method, particularly at mature stage. The cell wall preparations were fermented "in vitro" and the data correlated with lignin concentrations obtained through the three methods. The spectrophotometric method exhibited the lowest correlation coefficients for the majority of samples, which strongly suggests that this method must be better evaluated.Key Words: acetyl bromide lignin, a...
