Relatively little is known about the genetic control of agronomic traits in common wheat (Triticum aestivum L.) compared with traits that follow Mendelian segregation patterns. A doubled-haploid population was generated from the cross RL4452x'AC Domain' to study the inheritance of the agronomic traits: plant height, time to maturity, lodging, grain yield, test weight, and 1000-grain weight. This cross includes the genetics of 2 western Canadian wheat marketing classes. Composite interval mapping was conducted with a microsatellite linkage map, incorporating 369 loci, and phenotypic data from multiple Manitoba environments. The plant height quantitative trait loci (QTLs), QHt.crc-4B and QHt.crc-4D, mapped to the expected locations of Rht-B1 and Rht-D1. These QTLs were responsible for most of the variation in plant height and were associated with other agronomic traits. An additional 25 agronomic QTLs were detected in the RL4452x'AC Domain' population beyond those associated with QHt.crc-4B and QHt.crc-4D. 'AC Domain' contributed 4 alleles for early maturity, including a major time to maturity QTL on 7D. RL4452 contributed 2 major alleles for increased grain yield at QYld.crc-2B and QYld.crc-4A, which are potential targets for marker-assisted selection. A key test weight QTL was detected on 3B and prominent 1000-grain weight QTLs were identified on 3D and 4A.
The physiological cholesterol-lowering benefits of β-glucan have been well documented, however, whether modulation of gut microbiota by β-glucan is associated with these physiological effects remains unknown. The objectives of this study were therefore to determine the impact of β-glucan on the composition of gut microbiota in mildly hypercholesterolemic individuals and to identify if the altered microbiota are associated with bioactivity of β-glucan in improving risk factors of cardiovascular disease (CVD). Using a randomized, controlled crossover study design, individuals received for 5-week either a treatment breakfast containing 3 g high molecular weight (HMW), 3 g low molecular weight (LMW), 5 g LMW barley β-glucan, or wheat and rice. The American Heart Association (AHA) diet served as the background diet for all treatment groups. Phases were separated by 4-week washout periods. Fecal samples were collected at the end of each intervention phase and subjected to Illumina sequencing of 16S rRNA genes. Results revealed that at the phylum level, supplementation of 3 g/d HMW β-glucan increased Bacteroidetes and decreased Firmicutes abundances compared to control (P < 0.001). At the genus level, consumption of 3 g/d HMW β-glucan increased Bacteroides (P < 0.003), tended to increase Prevotella (P < 0.1) but decreased Dorea (P < 0.1), whereas diets containing 5 g LMW β-glucan and 3 g LMW β-glucan failed to alter the gut microbiota composition. Bacteroides, Prevotella, and Dorea composition correlated (P < 0.05) with shifts of CVD risk factors, including body mass index, waist circumference, blood pressure, as well as triglyceride levels. Our data suggest that consumption of HMW β-glucan favorably alters the composition of gut microbiota and this altered microbiota profile associates with a reduction of CVD risk markers. Together, our study suggests that β-glucan induced shifts in gut microbiota in a MW-dependent manner and that might be one of the underlying mechanisms responsible for the physiological benefits of β-glucan.
Wheat grain quality is a complex group of traits of tremendous importance to wheat producers, end-users and breeders. Quantitative trait locus (QTL) analysis studied the genetics of milling, mixograph, farinograph, baking, starch and noodle colour traits in the spring wheat population RL4452/'AC Domain'. Forty-seven traits were measured on the population and 99 QTLs were detected over 18 chromosomes for 41 quality traits. Forty-four of these QTLs mapped to three major QTL clusters on chromosomes 1B, 4D, and 7D. Fourteen QTLs mapped near Glu-B1, 20 QTLs mapped near a major plant height QTL on chromosome 4D, and 10 QTLs mapped near a major time to maturity QTL on chromosome 7D. Large QTLs were detected for grain and flour protein content, farinograph absorption, mixograph parameters, and dietary fibre on chromosome 2BS. QTLs for yellow alkaline noodle colour parameter L* mapped to chromosomes 5B and 5D, while the largest QTL for the b* parameter mapped to 7AL.
Durum breeders use a range of techniques in the development of new cultivars. An important selection criterion is the rheological properties of semolina dough and durum wheat breeders use this criterion in the development of new cultivars using a range of techniques. Because of the need to process large numbers of genotypes encountered in breeding programs, methods that are inexpensive, rapid, require small amounts of sample and that correlate with semolina quality are desirable. Using breeding material, this study investigated the relationship between the glutenin subunit composition and two traditional tests of gluten strength, gluten index (GI) and mixograph. Two sample sets of durum wheat breeding lines and cultivars, one grown in Canada (n = 229) and the other grown in Australia (n = 139) were analysed for GI, mixograph and both high molecular weight (HMW) and low molecular weight (LMW) glutenin subunits by SDS-PAGE. Nine different HMW and 14 different LMW allelic combinations were found. In the Canadian set, the most frequent LMW alleles were aaa, bba, caa and cfa while in the Australian set, caa was predominant. For the HMW subunits, the most common allelic groups were Glu-A1c/Glu-B1d (null, 6 + 8) and Glu-A1c/Glu-B1b (null, 7 + 8) with fewer numbers of Glu-A1c/GluB1e (null, 20) in both sample sets. LMW subunits were more important contributors to gluten strength than HMW subunits with the rank for higher GI according to the LMW allele (Canadian set) being caa = aaa > bba and aaa > cfa while HMW subunits 6 + 8 = 7 + 8 > 20. Similarly, using the mixograph, strength ranking for the LMW alleles was aaa > cfa = bba and HMW subunit 20 gave poorer rheological properties. For some samples with a good LMW allelic group a low GI was observed and vice versa. Further characterisation of the protein composition in these samples showed the GI results could be explained by polymeric/monomeric (P/M), glutenin/gliadin (Glu/Gli) and HMW/LMW ratios or the proportion of unextractable polymeric protein.
. 2005. Strongfield durum wheat. Can. J. Plant Sci. 85: 651-654. Strongfield durum wheat (Triticum turgidum L. var durum) is adapted to the durum production area of the southern Canadian prairies. It combines high yield, high grain protein concentration, and low grain cadmium concentration. Strongfield has shorter, stronger straw than Kyle, and has similar maturity and disease resistance to other currently registered durum cultivars.
Background/Objectives: To more precisely quantify the effect of barley b-glucan on blood lipid concentrations in humans and to examine the factors that could affect its efficacy. Subjects/Methods: Eleven eligible randomized clinical trials published from 1989 to 2008 were identified from nine databases. Weighted mean effect sizes were calculated for net differences in lipid profile using a random effect model (RevMan 4.2). Results: Overall, barley and b-glucan isolated from barley lowered total and low-density lipoprotein (LDL) cholesterol concentrations by 0.30 mmol/l (95% confidence interval (CI): À0.39 to À0.21, Po0.00001) and 0.27 mmol/l (95% CI: À0.34 to À0.20, Po0.00001), respectively, compared with control. The pattern of cholesterol-lowering action of barley in this analysis could not be viewed as a dose-dependent response. There were no significant subgroup differences by type of intervention and food matrix. Conclusions: Increased consumption of barely products should be considered as a dietary approach to reduce LDL cholesterol concentrations.
