The four weakly-inducible organoids were treated with a range of concentration of GSK126 between 0-10 µM for 72 hours, prior to determining the cell viability using CellTiter-Glo 3D Cell Viability Assay. Results were scaled relative to untreated control cells (given relative viability of 1.0) and all conditions were based on a minimum of five replicates. Mean values with standard deviation displayed. Small increases in viability noted in 3 out of 4 lines with low doses up to 2 µM, consistent with prior literature.

Characteristics of 2D cell lines assessed. Class II inducibility (flow cytometry) and CIITApIV methylation status as assessed by Satoh et al. Demographics, where available, from ATCC and Cellosaurus.org. Additional data on the site of tumour origin, mutation (KRAS and BRAF), mismatch repair (microsatellite instability MSI or microsatellite stable MSS) and CpG methylator phenotype (CIMP) status adapted from Ahmed et al. scite is a Brooklyn-based organization that helps researchers better discover and understand research articles through Smart Citations–citations that display the context of the citation and describe whether the article provides supporting or contrasting evidence. scite is used by students and researchers from around the world and is funded in part by the National Science Foundation and the National Institute on Drug Abuse of the National Institutes of Health.
