Several Passiflora species have been used widely as a folk medicine due to their sedative and anxiolytic activities. In Brazil, a number of native plants of the genus Passiflora exist, but only Passiflora edulis f. flavicarpa (PE) and Passiflora alata (PA) are of commercial value. Thus, the aim of the present study was to investigate the sedative effects of aqueous extracts obtained from the pericarp as well as from the leaves of PE and PA in mice using radiotelemetry. Aqueous extracts from PE and PA were tested for effects on locomotion over 180 min in 300 mg/kg, 600 mg/kg and 1200 mg/kg, in male C57BL/6J mice after oral administration. For validation of the telemetry system, caffeine (negative control) and midazolam (positive control) were used. All tested extracts decreased locomotor activity in a dose-dependent manner in comparison to the control group. The two lower concentrations of each extract showed the highest decrease in locomotion after 24 min, while 1200 mg/kg had a significant sedative effect already after 18 min. Interestingly, aqueous extracts of PA were more active in comparison to aqueous extracts of PE and the pericarp extracts of both plants showed more pronounced effects on locomotor activity if compared to leaf extracts. In conclusion, the present study represents an innovative, objective approach to measure sedative effects of plant extracts with minimized handling-related stress and remote data collection.
ImportanceThe incidence of diabetes in childhood has increased during the COVID-19 pandemic. Elucidating whether SARS-CoV-2 infection is associated with islet autoimmunity, which precedes type 1 diabetes onset, is relevant to disease etiology and future childhood diabetes trends.ObjectiveTo determine whether there is a temporal relationship between SARS-CoV-2 infection and the development of islet autoimmunity in early childhood.Design, Setting, and ParticipantsBetween February 2018 and March 2021, the Primary Oral Insulin Trial, a European multicenter study, enrolled 1050 infants (517 girls) aged 4 to 7 months with a more than 10% genetically defined risk of type 1 diabetes. Children were followed up through September 2022.ExposureSARS-CoV-2 infection identified by SARS-CoV-2 antibody development in follow-up visits conducted at 2- to 6-month intervals until age 2 years from April 2018 through June 2022.Main Outcomes and MeasuresThe development of multiple (≥2) islet autoantibodies in follow-up in consecutive samples or single islet antibodies and type 1 diabetes. Antibody incidence rates and risk of developing islet autoantibodies were analyzed.ResultsConsent was obtained for 885 (441 girls) children who were included in follow-up antibody measurements from age 6 months. SARS-CoV-2 antibodies developed in 170 children at a median age of 18 months (range, 6-25 months). Islet autoantibodies developed in 60 children. Six of these children tested positive for islet autoantibodies at the same time as they tested positive for SARS-CoV-2 antibodies and 6 at the visit after having tested positive for SARS-CoV-2 antibodies. The sex-, age-, and country-adjusted hazard ratio for developing islet autoantibodies when the children tested positive for SARS-CoV-2 antibodies was 3.5 (95% CI, 1.6-7.7; P = .002). The incidence rate of islet autoantibodies was 3.5 (95% CI, 2.2-5.1) per 100 person-years in children without SARS-CoV-2 antibodies and 7.8 (95% CI, 5.3-19.0) per 100 person-years in children with SARS-CoV-2 antibodies (P = .02). Islet autoantibody risk in children with SARS-CoV-2 antibodies was associated with younger age (<18 months) of SARS-CoV-2 antibody development (HR, 5.3; 95% CI, 1.5-18.3; P = .009).Conclusion and relevanceIn young children with high genetic risk of type 1 diabetes, SARS-CoV-2 infection was temporally associated with the development of islet autoantibodies.
The current study was designed to compare the consumer acceptance of the three Rvi6 scab-resistant apple cultivars ‘Rustica’, ‘Ariane’ and ‘Ladina’ with the two standard disease-susceptible cultivars ‘Braeburn’ and ‘Mariella’. A consumer test with 189 participants, using written questionnaires, was carried out alongside a sensory descriptive analysis with a trained panel. For appearance acceptance ‘Ariane’ reached the highest score. Three distinct groups of consumers with different visual preferences were found using hierarchical cluster analysis. In terms of eating quality ‘Mariella’ was liked the most. Yet again, three groups were found, with specific preferences regarding textural and taste properties. Participants also responded to questions concerning apple consumption and demographics. Significant effects of these parameters on cultivar likings were found. The study showed that there are scab resistant apples which are well accepted and therefore could be equal or superior alternatives to disease susceptible cultivars from a consumer’s point of view.
Introduction: This study examined the emotional impact that parents experience when confronted with an increased genetic risk of type 1 diabetes (T1D) in their child. Population-based screening of neonates for genetic risk of chronic disease carries the risk of increased emotional burden for parents.Methods: Information was collected using a well-being questionnaire for parents of infants identified as having an increased risk for T1D in a multinational research study. Parents were asked to complete this questionnaire after they were told their child had an increased risk for T1D (Freder1k-study) and at several time points during an intervention study (POInT-study), where oral insulin was administered daily.Results: Data were collected from 2595 parents of 1371 children across five countries. Panic-related anxiety symptoms were reported by only 4.9% after hearing about their child having an increased risk. Symptoms of depression were limited to 19.4%

Diese Web-Applikation kann durch ihre einfache, praktische und kostenlose Anwendung –zugänglich unter http://www.frudistor.de von jeglichem digitalen Endgerät aus (PC, Tablet bzw. Smartphone)– Produzenten, Lagertechniker, Handelsorganisationen und Konsumenten dabei begleiten, die verschiedenen Schäden, die während der Lagerung von Früchten entstehen können, zu identifizieren und zu erkennen, um ihnen vorbeugen zu können und sie zu vermeiden. Dies wird mit Hilfe eines ausgeklügelten, einfachen Filtersystems realisiert. Die Software enthält mehr als 40 technische Datenblätter (derzeit in Italienisch, Deutsch, Englisch und Niederländisch) und Bilder, die es ermöglichen, den Ursprung von Schäden schnell zu erkennen und somit eine Vermeidung zu gewährleisten.
The storage life of ‘Alexander Lucas’ pear is limited by the occurrence of internal storage disorders during controlled atmosphere (CA). 1-Methylcyclopropene (1-MCP) can be used to delay pear ripening, however, its interaction with CA conditions is not well understood. ‘Alexander Lucas’ fruit were treated with 1-MCP at 300 nL L-1 for 24 h in air at-0.5 or 1.0 °C before being stored. Fruit were kept at-0.5 or 1.0 °C in normal air or CA (2.0 kPa O2 plus <0.7 kPa CO2). After six months of storage, superficial scald did not develop in fruit. The highest incidence of flesh browning (72.2 %) was found in air-stored fruit at-0.5 °C without 1-MCP treatment. Storage in air at 1.0 °C plus 300 nL L-1 1-MCP produced 88.8 % of healthy fruit. In contrast, 1-MCP increased the incidence of flesh and core browning under CA conditions. 1-MCP and CA maintained greener skin color and higher titratable acidity. No significant differences were found for fruit firmness, total soluble solids and ascorbic acid content between the treatments. In conclusion, the quality of ‘Alexander Lucas’ pear was best maintained during six months storage under normal air at 1.0 °C combined with 300 nL L-1 1-MCP treatment.
