The monograph presents the most simple and widely used methods for determining the most important of plant antioxidants: vitamin C, polyphenols, carotenoids, capsaicin, and belinovich photosynthetic pigments, flavonoids, anthocyanins, alkaloids, tannins, and minerals antioxidant: selenium and iodine. Special attention is paid to methods of extraction of antioxidants, providing maximum extraction of antioxidants from plant material, and the correct selection of the most appropriate method of analysis of one or another component. Provides detailed information developed by the authors method of using thin layer chromatography to assess the carotenoid composition of tomatoes and peppers. The data presented here include results of research conducted on the basis of FICO, as well as the latest developments of foreign scientists devoted to natural antioxidants and methods of their determination. Presented in this monograph methodology was successfully tested in the laboratory and analytical Department of PNCO in 2012-2018. The unique biological properties of A. annua have stimulated the research on its cultivation in different regions of the world. In this study, the effect of the Se and nano-Si supply on the yield, biochemical characteristics and mineral content of A. annua was investigated. Growth stimulation and a significant increase in the antioxidant status were recorded under Se and nano-Si foliar application. A decrease in the number of essential oil components and significant changes in the essential oil amount and composition led to significant phenophase shifts: nano-Si significantly stimulated eucalyptol and artemisia ketone accumulation and decreased germacrene D production, whereas Se demonstrated the opposite effect. A joint Se and nano-Si supply significantly decreased the camphor content and increased artemisia ketone and artemisinin levels by 1.3–1.5 times. Se/Si supplementation affected the macro- and microelements content, causing either a redistribution of leaves/stems elements (Al, Li and Zn) or a significant decrease in Ca, Mg, K, B, Cu, Fe and Mn concentrations in leaves, with no signs in growth inhibition or a decrease in the photosynthetic pigments content. The biofortification of A. annua with Se singly or in combination with nano-Si resulted in the synthesis of products with a Se content of as much as approximately 16% of the daily adequate Se consumption level (ACL) when using 5 g day-1 as a spice, or 36% of ACL when using 50 mL of tea infusion (1:2, v/w). The results indicated a high possibility of Se and nano-Si application toward the regulation of A. annua growth, biochemical characteristics (including essential oil and artemisinin) and mineral content.
Mongolia is characterized by restricted sources of drinking water and intensive water pollution due to high rates of urbanization, mining industry development, enormous amount of livestock, and ever-growing attempts in domestic production of cereals and vegetables. Among others, Se is the least studied element in Mongolian water resources. Based on fluorimetric method of analysis, the first results on Se levels in drinking water of five aimags, Ulaanbaatar, and Erdenet were obtained. Uneven distribution of Se in Mongolia was manifested, the highest Se concentrations being typical for the southern resources (up to 18,600 μg/L) and the lowest, for the Northern ones (up to 0.022 μg/L). ICP-MS data of Al, As, B, Ca, Cd, Co, Cr, Cu, Fe, Hg, I, K, Li, Mg, Mn, Na, Ni, P, Pb, Si, Sn, Sr, V, and Zn contents indicate poly-microelementosis existence in the South of Mongolia (Dorno-Gobi aimag) where ground water is characterized by elevated levels of As and extremely high levels of Se, Li, Na, F, Cl, B, and nitrates ions, exceeding maximum permissible levels by 1.86; 4.3; 3.1; 3.1; 2.7; 3.4; and 1.8 times respectively. Toxic concentrations of Se in groundwater of Dorno-Gobi aimag contradict with the published low human serum Se and low content of the element in horseflesh that suggests the possible effect of the above pollutants on Se bioavailability. Revealed phenomenon and mosaic distribution of heavy metals in areas with high and low Se content in water resources indicate the need of direct search for Se and other pollutant transfer in food chain in various ecological loading conditions, creation of a map of Se distribution in water resources of other Mongolian regions, and large-scale evaluation of the human poly-elemental status.
Production of functional food with high levels of selenium and other antioxidants is very valuable for human protection against different forms of oxidant stress. Among leafy vegetables parsley demonstrate the highest levels of antioxidants. Biochemical analysis and fluorimetric determination of selenium revealed that foliar biofortification of 4 parsley (Petroselinum crispum) cultivars and Mitsuba (Cryptotaenia japonica) with sodium selenate (25 mg L-1) resulted in the highest biofortification level in curly parsley cultivar Krasotka (102.9) which showed the highest leaf surface area, antioxidant activity (65 mg GAE g-1 d.w.) and flavonoids content (25.9 mg quercetin equivalent g-1 d.w.), and the increase by 1.4 times in carotene content and 1,5 times in total chlorophyll content. ICP-MS method of mineral composition evaluation recorded extremely high levels of B and Si in Mustuba, which increased due to Se supplementation by 1.23 and 1.46 times respectively. In a two-year experiment with control and Se-fortified, leafy parsley, cultivar Moskvichka reached high values of seed yield and viability, and seed Se content (6170 µg kg-1 d.w.). The results of the present investigation demonstrate high prospects of parsley and Mitsuba selenium biofortification for production of functional food with elevated levels of microelement and high antioxidant activity.
Biochemical and mineral peculiarities of plants inhabiting desert and semi-desert areas may provide important information about the mechanism of their adaptability and reveal the prospects of their utilization. Rheum tataricum L., known for its high tolerance to drought, salinity, and nutritional deficiency, is the least studied species of wild rhubarb. Using biochemical and ICP-MS analysis, the antioxidant status and mineral composition of R. tataricum were determined. Extremely high levels of antioxidant activity (148–155 mg GAE g−1 d.w.), polyphenols (24.6–25.1 mg GAE g−1 d.w.) and carotenoids (1.94 mg-eq β-carotene g−1 d.w.) were revealed in roots, proline in leaves (71.1 ± 6.2 mg kg−1 d.w.) and malic acid in stems (3.40 ± 0.50 mg g−1 d.w.). Compared to garden rhubarb, R. tataricum demonstrated significant root–leaves translocation of Li, Se, Si, and Mo, known to participate in plant antioxidant defense. Under high levels of Ca, Na, Mg, Fe, Cr and Si in soil, R. tataricum demonstrated the ability to significantly increase the accumulation of these elements in roots, showing a hyperaccumulation ability for Sr. The first broad picture of R. tataricum biochemical and mineral characteristics in semi-desert habitat and its nutritional value indicate the prospects of R. tataricum utilization in plant breeding, medicine, and nutrition.
Selenium (Se) biofortification of aromatic plants is a promising strategy to produce valuable functional food with high biological activity and enhanced essential oil yield. The experiment carried out in 2021 and 2022 on A. annua treated with sodium selenate or nano-Se sprayed on foliar apparatus demonstrated a significant increase in photosynthetic pigments, pectin, waxes, macro- and microelements and a decrease in malonic dialdehyde (MDA) accumulation. Contrary to literature reports, neither selenate nor nano-Se showed a beneficial effect on essential oil accumulation; the oil yield did not differ between the selenate treated and control plants but was halved by the nano-Se application. Extremely high variations in the number of essential oil components, as well as in the eucalyptol, artemisia ketone, camphor and germacrene D ratio in the 2021 and 2022 experiments were recorded. The analysis of the 2016–2022 data for oil yield and composition in the control plants revealed a direct correlation between the number of components and of solar flares, and a negative correlation between oil yield and the percentage of spotless days. Both control plants and plants fortified with selenium showed higher levels of germacrene D and lower levels of artemisia ketone in 2022, characterized by more remarkable solar activity compared to 2021. Nano-Se supply resulted in the highest percentage of germacrene D accumulation. The results of the present research highlight the importance of the solar activity effect on the essential oil yield and quality of aromatic plants.
Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus are the primary bacteria that cause clinical infections, such as urinary and intestinal infections, pneumonia, endocarditis, and sepsis. Bacterial resistance is an innate natural occurrence in microorganisms, resulting from mutations or the lateral exchange of genetic material. This serves as evidence for the association between drug consumption and pathogen resistance. Evidence has demonstrated that the association between conventional antibiotics and natural products is a promising pharmacological strategy to overcome resistance mechanisms. Considering the large body of research demonstrating the significant antimicrobial activities of Schinus terebinthifolius Raddi, the present study aimed to evaluate the chemical composition and antibiotic-enhancing effects of Schinus terebinthifolius Raddi essential oil (STEO) against the standard and multidrug-resistant strains of Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. The STEO was extracted by hydrodistillation using a Clevenger-type vacuum rotary evaporator. The Minimum Inhibitory Concentration (MIC) of the STEO was assessed by the microdilution method to evaluate the antibacterial activity. The antibiotic-enhancing activity of the essential oil was assessed by determining the MIC of antibiotics in the presence of a sub-inhibitory concentration (MIC/8) of the natural product. The GC-MS analysis revealed alpha-pinene (24.3%), gamma-muurolene (16.6%), and myrcene (13.7%) as major constituents of the STEO. The STEO potentiated the enhanced antibacterial activity of norfloxacin and gentamicin against all the strains and increased the action of penicillin against the Gram-negative strains. Therefore, it is concluded that although the STEO does not exhibit clinically effective antibacterial activity, its association with conventional antibiotics results in enhanced antibiotic activity.
