Метою роботи було перевірити здатність ендотеліального моноцитактивуючого фактора ІІ (EMAP II) впливати на вільнорадикальний стан серця і судин, на відновлення спряженого стану конститутивних ізоформ NO-синтаз (сNOS)
ВСТУПВ частині І ми показали, що одним із ме-ханізмів розвитку порушень редокс-ста-тусу клітин серця і судинної стінки, що зумовлюють дисфункцію цих органів за гіпертензії, може бути індукція неспряженого стану конститутивних NO-синтаз (еNOS і/ чи nNOS), яка супроводжується знижен-ням конститутивного синтезу оксиду азоту (NO) і зростанням утворення супероксид-аніона ( • О 2 -) і його токсичних похідних -пероксинітриту (ONOO -) та гідроксильного аніон-радикала ( • OH). В умовах окисного стресу відбувається глутатіонування SH-груп цистеїну в молекулі ендотеліальної NOS (еNOS) окисненим глутатіоном, що і спричи-няє її неспряження [1][2][3]. Дані останніх дос-лід жень свідчать, що неспряження еNOS мо же бу ти також результатом впливу активованих при запаленні моноцитів [4]. Саме здатність акти вувати ендотеліальні клітини і моноцити спо ну кала авторів, які вперше описали ен-дотеліальний моноцитактивуючий фак тор ІІ (EMAP II), назвати його таким чином [5]. Крім того, було показано, що він відіг-рає важливу роль у розвитку запалення, апоптозу та ангіогенезу [6], а також здатен збільшувати проникність кровотканинного бар'єра [7]. Ці властивості EMAP II спону-кали дослідників до вивчення його як мож-ли вого протипухлинного агента [8] чи зас то сування його блокади для активації ан-гіо генезу та покращення функції серця при експе риментальному інфаркті міокарда [9]. Diabetes dramatically increases the risk of cardiovascular complications. The endothelial dysfunction and diastolic heart dysfunction are associated with a decreasing level of hydrogen sulfide (H2S) and inhibition of the activity of endothelial NO-synthase in diabetes. The aim of work is to investigate the effect of modulation of hydrogen sulfide synthesis on heart functions and vasorelaxation in diabetes. The DL-propargylglycine and L-cysteine were administered intraperitoneally. H2S content in the heart tissue, markers of oxidative stress, iNOS and cNOS activities, endothelium-dependent vasorelaxation of the aortic rings, and heart function were studied. We demonstrate that our combination increased H2S syntheses by 13 times and cNOS activity by 5 times in the heart tissue of diabetic rats. Increasing NO and H2S production caused improving and restoration of endothelium-dependent relaxation of aorta, effective arterial elastance, and diastolic heart function in diabetic rats. The endothelium-dependent relaxation increased by 2.4 times; effective arterial elastance decreased by 47%. The end-diastolic myocardial stiffness decreased by 2.2 times. Thus, modulation of hydrogen sulfide synthesis leads to increased activity cNOS by 5 times in the cardiovascular system. Increasing NO and H2S production restored endothelium-dependent relaxation of aorta and improved heart function in diabetes.
ВСТУПАдаптація до фізичних навантажень (трену-вання) має різноманітний вплив на організм, підвищуючи функціональні можливості його систем, насамперед тих, що забезпечують киснетранспортні процеси. Тому фізичне тренування часто використовується з кар-діопротекторною метою [1,2]. Дослідження останніх років показали, що основну роль у впливі тренування на серцево-судинну сис-тему відіграє активація ендотелію [3][4][5] та ендотеліальної NO-синтази (eNOS) за допо-могою збільшення фосфорилювання залишку серину у положенні 1177 та зменшення фос-форилювання залишку треоніну у положенні 495 [6,7]. Тому, можливо, позитивний вплив фізичних тренувань на серцево-судинну систему виявляється сильніше при патоло-гічних процесах у цій системі, пов'язаних із пригніченням eNOS, а саме при гіпертензії, атеросклерозі, серцевій недостатності тощо [8,9].
