Several studies demonstrated the utility of plasma-based cell-free circulating tumor DNA (ccfDNA) in determination of mutations in non-small cell lung cancer (NSCLC). We aimed to report our results of next generation sequencing (NGS) using liquid biopsy in patients with NSCLC. Patients with advanced stage NSCLC were enrolled and their genomic profiling results were recorded. Next generation sequencing targeted panel includes 19 hot-spot genes. The plasma was separated from the peripheral blood sample and ccfDNAs were isolated for NGS. We performed genomic profiling in 100 patients (20 females and 80 males) with a median age of 59.3 (range 26-79). A second liquid biopsy was performed in eight patients who developed progressive disease after the first treatment. The study population had adenocarcinoma (AC) (n = 73), squamous cell carcinoma (SCC) (n = 14), or NSCLC-NOS (not otherwise specified) (n = 13). In the SCC group, three of 14 patients had variants on EGFR and MET genes. In the AC and NSCLC-NOS groups, 39 out of 86 patients (45.3%) had variants. The most common one was in the EGFR gene (n = 27, 31.4%) including seven mutations related to drug resistance and two were polymorphisms. Three patients had both driver and resistance mutations (EGFR T790M, n = 2; KRAS exon 2 G12S and MET exon 14 E1012K, n = 1). Fifteen patients (17.4%) had an activating EGFR mutation and eight patients (9.3%) had variants in the KRAS gene. We reported our results regarding genomic profiling related to treatment using liquid biopsy in patients with NSCLC. Advantages of this method are the non invasiveness and reproducibility.
Kemoterapi sırasında hepatit enfeksiyonu reaktivasyonu lenfoma hastalarında önemli bir sorundur. Merkezimizde tedavi edilen lenfoma hastalarında hepatit B virüsü (HBV) ve hepatit C virüsü (HCV) sıklığını araştırmayı amaçladık. Materyal ve Metot: Merkezimizde 2011-2018 yılları arasında tedavi edilen non-hodgkin lenfoma (NHL) ve hodgkin lenfoma (HL) hastalarının dosyaları retrospektif olarak incelendi. Hastaların HBsAg, Anti-HBs, Anti-HBc ve Anti-HCV parametreleri değerlendirildi. Bulgular: 240 NHL hastasının 21' inde (% 8.7) HBsAg antijeni pozitifti. 9 hastada (% 3.7) Anti-HCV pozitifliği tespit edildi. 79 hastada (%33) HBsAg (-), Anti-HBs (+) ve Anti-HBc (+) saptandı ve geçirilmiş hepatit B enfeksiyonu olarak değerlendirildi. 110 HL hastasının 10' unda (%9.1) HBsAg, 3'ünde (%2.7) Anti-HCV pozitif saptandı. 29 hastada ise (%26.4) HBsAg(-), Anti-HBs(+), Anti-HBc (+) idi ve geçirilmiş hepatit B enfeksiyonu lehine değerlendirildi. HBsAg ve Anti-HCV pozitiflik oranları açısından NHL ve HL hasta grupları karşılaştırıldığında istatiksel anlamlı fark yoktu (p değerleri sırasıyla 0.9 ve 0.63 idi). Sonuç: Lenfoma hastalarında ülkemiz verilerine göre HBV ve HCV pozitifliğinin yüksek olduğu belirlenmiştir. Bölgemizdeki Hepatit B ve C pozitifliği önemli bir sorun teşkil etmektedir. Aşılama ve korunma yöntemleri hakkında gerekli bilgiler sağlanmalıdır.
Meme kanseri kadınlarda en sık görülen kanserdir ve bir numaralı kansere bağlı ölüm nedenidir. Bu çalışmada merkezimizde takip edilen metastatik meme kanseri (MMK) tanılı hastaların klinik özelliklerini ve tedavi sonuçlarını incelemek amaçlanmıştır. Gereç ve Yöntemler: Çukurova Üniversitesi Tıbbi Onkoloji Bilim Dalı'nda 2011-2017 yıllarında takip edilen 65 MMK'li hasta çalışmaya dahil edildi. Hastaların klinik ve tedavi verileri retrospektif olarak incelendi. Bulgular: Hastaların çoğunda sadece kemik metastazı vardı. Sonradan metastatik evreye geçen 45 hastanın %66,6'sında yeniden biyopsi yapılmıştı; bu hastaların %20'sinde tümör biyolojisi değişmişti ve değişiklikler istatistiksel olarak anlamlıydı (p=0,0056). Metastatik evrede progresyonsuz sağkalım 10 ay, ortalama sağkalım 34 ay olarak hesaplandı. Tartışma ve Sonuç: Hastaların sağkalım verileri literatürle uyumluydu. Yeni kullanıma giren ajanlar, hedefleyici tedavilerdeki gelişmeler ve tümör biyolojisindeki olası değişimler göz önünde bulundurularak, relapslarda yeniden biyopsi yapılması kritik önemdedir.
