Two modern rose varieties, Floribunda and Hybrid Tea, were used to analyze and identify metabolic changes after foliar application with benzyladenine (BA). Volatile organic compounds (VOCs) as metabolites were detected. Two pairs of doses of BA, at 11.16 and 17.87 mg/cm2, and 7.17 and 12.26 mg/cm2 were applied to the foliage of Hybrid Tea and Floribunda, respectively. Sampling time was optimized and treatment duration was 4 weeks. After treatment, the volatiles from the treated and untreated control roses were extracted using headspace solid-phase microextraction (HS-SPME) technology by three-phase fiber 50/30 µm divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) and analyzed by gas chromatography (GC) coupled with a flame ionization detector (FID), and with mass spectrometry (GC-MS).The results showed that BA and its dose rate led to metabolic changes of treated roses in comparison with untreated controls. The number of VOCs extracted and detected from leaves, stem, rhizosphere and whole plants from the two rose varieties at doses rate of 17.87 and 12.26 mg/cm2 were 43, 65, 40 and 68 compounds for each plant material, respectively, for both rose varieties. Whilst the VOCs extracted and detected from both rose varieties for leaves, stem, rhizosphere and whole plants were 38, 61, 34 and 66 compounds for each plant material, respectively. The results demonstrate that some volatiles, such as 4-Heptyn-2-ol, Phenyl methyl ether and 3-Methyl-apopinene, increased with increasing doses of BA; these compounds are aroma chemicals with a very powerful smell. This study shows that BA treatments can have a significant effect on metabolite changes in different rose tissues. This method could be applied to other floriculture plants.
The study focused on the influence of the plant growth regulators (PGRs) benzyladenine (BA) and naphthalene acetic acid (NAA) on the production of volatile organic compounds (VOCs) from the flowers of two modern rose varieties, Hybrid Tea and Floribunda. Thirty-six plants of Hybrid Tea and Floribunda were tested. Benzyladenine and naphthalene acetic acid were applied at 0, 100 and 200 mg/L to both rose varieties. Gas chromatography, coupled with flame ionization detection and mass spectrometry, was used to analyze and identify the volatile organic compounds from the flowers. A three-phase fiber 50/30 µm divinylbenzene/carboxen/polydimethylsiloxane was used to capture VOCs, at 2, 4 and 8 weeks, and 4 weeks was selected as it had the highest peak area. In total, 81 and 76 VOCs were detected after treatment of both rose varieties with BA and NAA, respectively. In addition, 20 compounds, which had significant differences between different treatments, were identified from both rose varieties. The majority of VOCs were extracted after the application of 200 mg (BA and NAA) /L of formulation, and four important compounds, cis-muurola-4(141)5-diene, y-candinene, y-muurolene and prenyl acetate, increased significantly compared to the controls. These compounds are commercially important aroma chemicals. This study used the rapid and solvent-free SPME method to show that BA and NAA treatments can result in significant VOC production in the flowers of two rose varieties, enhancing the aromatic value of the flowers. This method has the potential to be applied to other valuable aromatic floricultural plant species.
Hydroponic cropping technique was utilized for evaluation of salinity tolerance of certain genotypes of rice under prevailing arid conditions of Iraq. The conditions envelop low level of characteristic issue, calcium carbonate conglomeration of soil and soils containing clear measures of gypsum, delicate to coordinate profile progression and low natural development. The seedling test is simple, rapid (4 to 6 weeks) and accurate. Furthermore, this test allows the screening of several seedlings under various experimental parameters. Therefore, the three genotypes of IRRI rice (IR71999-3R-3-2-2B-1-1, IR71829-3R-82-1-1and IR63731-1-3-3-2) and two varieties of rice (Anber and Jasmine) under salinity stress (1.2, 4, 8 and 10 dS/M) were investigated. The experiment was conducted in a growth chamber under simulated arid conditions in a completely randomized distribution. Salt stress caused reductions in rice plant production (yield) reached more than 30%. Dry matter weight was decreased by 24% in all varieties ofrice. Sodium (Na + ) concentration was significantly (p ≤ 0.05) higher in NaCI-treated plants. The Na + /K + and Na + /Mg 2+ ratio in IRSSTN-SS2 (IR71999-3R-3-2-2B-1-1, IR71829-3R-82-1-1) genotypes and Anber plant indicate that it is more susceptible to salinity than the rice verities. Consequently, this cultivar is more vulnerable to saltiness than the assortment Cotaxtla, which demonstrated more noteworthy resistance to this anxiety.
Rose (Rosa hybrida L.) is one of the most important specialty cut flowers produced. Characterization of physiological variability in photosynthetic efficiencies, respiration rate and chlorophyll content is one of the greatest challenges in assessing rose net primary production of flowering, quality of cut flowers and other rose uses. Two modern rose varieties Floribunda and Hybrid Tea were used to optimize different methods for analysing physiological characteristics (photosynthesis rate, respiration rate and chlorophyll content). Many parameters were optimized, five different lights intensity were used (600, 900, 1200, 1500 and 2000 μmol•m •s −1 gave the highest photosynthesis rate. Five different measuring times were used (8 am, 10 am, 12 pm, 2 pm and 4 pm) and 12 pm was shown to be the optimum time for measuring photosynthesis rate. Among two different weather conditions (sunny day and cloudy day) sunny day was selected. For respiration rate two different measuring times (1 and 2 hours) after darkness were studied and 1 hour was chosen. Among three different times (10 am, 12 pm and 2 pm) for measuring chlorophyll content, 12 pm was selected. Using these optimized variables will allow researchers to collect robust and reproducible results to be obtained from different studies, and in turn lead to improved yields of horticultural plants.
An experiment for strawberry Fragaria ananassa cv. Festival in vitro conservation of cultures for nine months was studied. Three MS medium strength (full strength MS, ¾ MS, ½ MS, and ¼ MS) and the storage period (3, 6, and 9 months) in slowing down or reducing growth. Results showed a significant effect of the medium salt strength, as the full strength treatment was higher by achieved the highest percentage of viability (survival %) and storage characteristics of cultures (N%, P%, carbohydrates%, protein %) and regeneration ability characteristics (shoots and roots number and length). Three-month storage period achieved the highest rate compared to the nine-month storage period, which recorded the lowest average for the studied characteristics. The full strength treatment for three months storage period realized the highest rate compared to treatment ¼ strength for the storage period of nine months, which recorded the lowest rate of the studied traits. The application of minimal strength for in vitro conservation allows strawberry cultures to remain viable with a slow growth rate and a significant increase in the storage life of the tissues to nine months.
