In the present study, ameliorating role of hydrogen sulfide (H2S) in oilseed rape (Brassica napus L.) was studied with or without application of H2S donor sodium hydrosulfide (NaHS) (0.3 mM) in hydroponic conditions under three levels (0, 0.1 and 0.3 mM) of aluminum (Al). Results showed that addition of H2S significantly improved the plant growth, photosynthetic gas exchange, and nutrients concentration in the leaves and roots of B. napus plants under Al stress. Exogenously applied H2S significantly lowered the Al concentration in different plant parts, and reduced the production of malondialdehyde and reactive oxygen species by improving antioxidant enzyme activities in the leaves and roots under Al stress. Moreover, the present study indicated that exogenously applied H2S improved the cell structure and displayed clean mesophyll and root tip cells. The chloroplast with well-developed thylakoid membranes could be observed in the micrographs. Under the combined application of H2S and Al, a number of modifications could be observed in root tip cell, such as mitochondria, endoplasmic reticulum, and golgi bodies. Thus, it can be concluded that exogenous application of H2S under Al stress improved the plant growth, photosynthetic parameters, elements concentration, and biochemical and ultrastructural changes in leaves and roots of B. napus.

Grafting has been reported as a factor that influences fruit quality. However, a comprehensive study of the metabolic profile related to fruit quality and the underlying molecular mechanism in grafted watermelon has not been carried out. Metabolomics and transcriptome analysis were performed on both pumpkin-grafted watermelon and ungrafted watermelon at different developmental stages. In total, 56 primary metabolites were identified with either high or low abundance between ungrafted and pumpkin-grafted watermelon. The results indicated that ornithine, arginine, lysine (amino acids), glucose, sucrose, glucosamine (sugars), malic acid, fumaric acid and succinic acid (organic acids) were among the dominant metabolites influencing fruit quality. Additionally, comparative RNA sequence analysis on grafted and ungrafted watermelon yielded 729, 174, 128 and 356 differentially expressed genes at 10, 18, 26 and 34 days after pollination (DAP), respectively. Functional annotations of these genes indicated that grafting significantly altered the biological and metabolic processes related to fruit quality. Our comparative metabolomics and transcriptome analysis revealed that FBA2, FK, SuSy, SPS, IAI, AI and sugar transporter gene (SWT3b) might play a central role in the accumulation of glucose and sucrose, whereas higher malic acid content was attributed to high down regulation of ALMT13 and ALMT8 in pumpkin-grafted watermelon. Changes in the ornithine, glutamine, alanine, tyrosine, valine, asparagine, phenylalanine, arginine and tryptophan contents were consistent with the transcript level of their metabolic genes such as NAOD, GS, AGT, TaT, aDH1, OGDH, aDC, 4CL 1, PaL, CaT and two nitrate transporter genes (NRT1) in pumpkin-grafted watermelon. This study provides the basis for understanding the graft-responsive changes in the metabolic profile and regulatory mechanism related to fruit quality.

Contamination by petroleum hydrocarbons (PHs) is a great threat to environment due to the higher persistence and bio-toxicity of PHs. Therefore, removal of PHs from contaminated environment and strategies to reduce their toxic effects on living organisms are crucial for environmental safety and human health. The toxic effects of PHs from the polluted soil can be reduced by the addition of microbes and biochar. In this study, a pot trial was carried out to evaluate the effects of sugarcane bagasse (SB) biochar and Bacillus sp. MN54 addition on phytoremediation of PHs and growth of maize (Zea mays L.) in soil artificially contaminated with diesel. Maize seeds were sown in uncontaminated or contaminated (with PHs) soil, treated with biochar and Bacillus sp. MN54. The results revealed that PHs showed significant phytotoxicity to maize plants and the application of strain MN54 and biochar greatly reduced the toxic effects of PHs on plants growth and physiology by increasing the nutrients uptake in PHs contaminated soil. Interestingly, the phytotoxicity of PHs on maize plants was further reduced in the co-supplementation of strain MN54 and biochar. Plants physiological (25–48%) and agronomic (38–47%) attributes were significantly higher as compared to only PHs contaminated soil in the co-supplementation of strain MN54 and biochar. Similarly, nitrogen (41%), phosphorus (43%) and potassium (37%) concentrations were also increased in the co-supplementation of strain MN54 and biochar. Furthermore, maize plants successfully phytoremediate a considerable amount of PHs from soil particularly in the presence of strain MN54 and biochar, and this PHs removal was further enhanced in the co-supplementation of strain MN54 and biochar (i.e., 46% and 77% of initial PHs were removed in unplanted and planted treatments, respectively). The present results indicate that co-supplementation of biochar and Bacillus sp. MN54 could be effective in enhancing the degradation of PHs and improving plant growth in the hydrocarbons contaminated soil.
