Fungal infections in immune-compromised patients are an important cause of mortality and morbidity. Amphotericin B (Amp B) is considered a powerful fungicidal drug but its clinical usage has certain limitations when administered intravenously due to its toxicity and poor solubility. In consideration of such challenges, in cutaneous leishmaniasis, the topical application of Amp B can be a safer option in many aspects. Thus, herein, biopolymer of polycaprolactone (PCL) nanoparticles (NPs) were developed with the loading of Amp B by nanoprecipitation for the treatment of topical leishmanial infections. Various parameters, such as concentration of PCL and surfactant Poloxamer 407, were varied in order to optimize the formation of nanoparticles for the loading of Amp B. The optimized formulation exhibited a mean hydrodynamic particle size of 183 nm with a spherical morphology and an encapsulation efficiency of 85%. The applications of various kinetic models reveal that drug release from nanoformulation follows Korsmeyer–Peppas kinetics and has a high diffusion exponent at a physiological pH of 7.4 as well a skin relevant pH = 5.5. The activity of the prepared nanoparticles was also demonstrated in Leishmania infected macrophages. The measured IC50 of the prepared nanoparticle formulation was observed to be significantly lower when compared to control free Amp B and AmBisome® for both L. tropica KWH23 and L. donovani amastigotes in order to demonstrate maximum parasite inhibition. The prepared topical nanoformulations are capable of providing novel options for the treatment of leishmaniasis, which can be possible after in vivo assays as well as the establishment of safety profiles.

Magnetically responsive engineered smart thin films of nanoferrites as contrast agent are employed to develop surface based magnetic resonance imaging to acquire simple yet fast molecular imaging. The work presented here can be of significant potential for future lab-on-chip point-of-care diagnostics from the whole blood pool on almost any substrates to reduce or even prevent clinical studies involve a living organism to enhance the non-invasive imaging to advance the '3Rs' of work in animals-replacement, refinement and reduction.

A new class of a series of amphiphilic polyelectrolytes functionalized with azobenzene chromophores have been synthesized and thoroughly characterized by various techniques. A facile two stage strategy is developed, and first involved the preparation of a precursor base polymer, designated as P(APEA), by the free-radical copolymerization of the monomers of acrylic acid (AA) and 2-(phenylethylamino)ethyl acrylate (PEA). In the second step, precursor PAPEA polymers are reacted and post-polymer modified with the diazonium salts to synthesize azobenzene polyelectrolyte, PAPEA-R with different substituted R-groups present on the para position of the aromatic ring of the azo chromophores. The PAPEA-R polyelectrolytes are same in their degree of polymerization and repeat unit compositions of AA and PEA monomers, and only differ from each other by the type of R-groups. The copolymers were classified on the basis of the characteristics of their R-groups into the hydrophilic-ionizeable smaller pair of SO 3 H and COOH, the hydrophobic-non-ionizeable smaller pair of H and OC 2 H 5 , and the hydrophobic-non-ionizeable larger octyl pair of C 8 H 17 and C 8 F 17 . The prepared copolymers are also characterized by NMR spectroscopy for structure, GPC for molecular weight, and UV-Visible spectroscopy for absorption determination. In DMF solvent, approximately similar absorption maxima, (λ max ) values were observed for azo chromophore-containing monomers and after incorporating these into their corresponding polymers structures. However, for the self-assembled multilayer thin films, λ max moved to lower wavelengths to exhibit a blue shift with hydrophobic Rgroup of C 8 H 17 , while this behaviour was not observed for hydrophilic R-group of COOH. The shift in λ max is found to be highly dependent on the type of substituted R-group, and attributed to aggregation of hydrophobic azo chromophores in DMF:H 2 O mixture employed for self-assembly. The presence of the ionizeable AA and light-sensitive azo-chromophore functionalized PEA monomers in the PAPEA-R polyelectrolytes impart self-assembling and photoswitchable characteristics, respectively. Through understanding and controlling the solubility and complex solution aggregation behaviour of the different substituted R-groups of azo PEL, their adsorption, thickness, morphology, wetting, molecular-control, and photoresponsiveness can be tailored to enhance the capabilities of the self-assembled multilayer film process in diversified areas of microfluidics, sensing, and controlled release.
