Helicobacter pylori infection is associated with chronic gastritis, peptic ulcers, and gastric cancer. The flavonoid compounds baicalin and baicalein found in many medicinal plants exhibit an anti-inflammatory effect. The administration of Lactobacillus strains reducing the risk of H. pylori infection is well accepted. In this study, the therapeutic effects against H. pylori infection of baicalin, baicalein, and L. rhamnosus JB3 (LR-JB3), isolated from a dairy product, were investigated. Compared to baicalin, baicalein exhibited stronger anti-H. pylori activity and cytotoxicity on human gastric cancer epithelial AGS cells. Baicalin and baicalein both suppressed the vacA gene expression of H. pylori and interfered with the adhesion and invasion ability of H. pylori to AGS cells, as well as decreased H. pylori-induced interleukin (IL)-8 expression. In the mice infection model, high dosages of baicalin and baicalein inhibited H. pylori growth in the mice stomachs. Serum IL-1β levels and H. pylori-specific serum IgM and IgA levels in mice treated with baicalin and baicalein were decreased. Moreover, a synergistic therapeutic effect of baicalein and LR-JB3 on eradicating H. pylori infections was observed. Thus, administrating baicalin, baicalein, or LR-JB3 for an H. pylori infection could offer similar therapeutic effects to administering antibiotics while not disturbing the balance of gut microbiota. This study revealed the effects of baicalin, baicalein, and LR-JB3 on attenuating the virulence of H. pylori. The synergistic effect with baicalein and LR-JB3 provides the experimental rationale for testing the reliability, safety, and efficacy of this approach in higher animals and perhaps ultimately in humans to eradicate H. pylori infections.
Background The biting midge, Forcipomyia taiwana, is one of the most annoying blood-sucking pests in Taiwan. Current chemical control methods only target the adult, not the immature stages (egg to pupa), of F. taiwana. Discovering new or alternative tactics to enhance or replace existing methods are urgently needed to improve the effectiveness of F. taiwana control. The egg is the least understood life stage in this pest species but may offer a novel point of control as addition of NaCl to the egg environment inhibits development. Thus, the objective of this study was to use RNA profiling to better understand the developmental differences between wild-type melanized (black) and NaCl-induced un-melanized (pink), infertile F. taiwana eggs. Results After de novo assembly with Trinity, 87,415 non-redundant transcripts (Ft-nr) with an N50 of 1099 were obtained. Of these, 26,247 (30%) transcripts were predicted to have long open reading frames (ORFs, defined here as ≥300 nt) and 15,270 (17.5%) transcripts have at least one predicted functional domain. A comparison between two biological replicates each of black and pink egg samples, although limited in sample size, revealed 5898 differentially expressed genes (DEGs; 40.9% of the transcripts with long ORFs) with ≥2-fold difference. Of these, 2030 were annotated to a Gene Ontology biological process and along with gene expression patterns can be separated into 5 clusters. KEGG pathway analysis revealed that 1589 transcripts could be assigned to 18 significantly enriched pathways in 2 main categories (metabolism and environmental information processing). As expected, most (88.32%) of these DEGs were down-regulated in the pink eggs. Surprisingly, the majority of genes associated with the pigmentation GO term were up-regulated in the pink egg samples. However, the two key terminal genes of the melanin synthesis pathway, laccase2 and DCE/yellow, were significantly down-regulated, and further verified by qRT-PCR. Conclusion We have assembled and annotated the first egg transcriptome for F. taiwana, a biting midge. Our results suggest that down-regulation of the laccase2 and DCE/yellow genes might be the mechanism responsible for the NaCl-induced inhibition of melanization of F. taiwana eggs.
<p>Cosmopolitan consumers generally refer to individuals who are open and unbiased towards foreign products and identify as citizens of the world. Despite growing relevance of understanding consumer cosmopolitanism (COS) in today’s changing international landscape, both the construct itself is still not that well understood and operationalized, as well as its social and psychographic antecedent. In terms of culture studies, recent culture studies within the IB discipline continue to debate over the appropriate definitions of culture, as well as its conceptualization, operationalization, and especially measurement. This research aims to explore the impact of personal cultural orientations (PCOs), as individual-level culture value concepts, on COS, as well as the potential moderating role of product involvement (e.g., high- vs low-involvement products). Young-adult consumers in Taiwan and New Zealand were chosen to provide a contrast between a typical Western, Anglo-Saxon-based perspective and a typical Eastern, Asian, Confucian-based cultural context. Overall, this study could not fully support PCOs as having a significant impact on COS, nor product involvement as a moderator. However, COS could not be tested as a second-order reflective latent construct as originally intended in the seminal paper by Riefler et al. (2012). Compared to Riefler et al. (2012)’s paper, differences can be observed in the sampling where Riefler et al. (2012) sampled respondents aged 19 to 93 years (mean=46.6). This suggests that young adults a generational cohort hold significantly different perspectives and dispositions to other generations and the overall population. This finding aligns with recent IB literature in looking for smaller ‘containers’ of culture. Of the three PCOs tested, two displayed significant effects to COS in both country samples, but only to one dimension of the COS construct (Open-mindedness) and not the other (Diversity appreciation). Hence it is highly likely the PCOs tested in this study have significant effects on COS, if only COS could be operationalized as originally intended as discussed above. This points towards potential issues in appropriateness of the scales used for studies on young adults, as both the PCO and COS scales were developed on a wide range (age diverse) of respondents. This study also showed that within-country differences appear to be smaller than across-country differences. This is not consistent with previous cross-cultural research in the IB literature, which suggest cultural values differ significantly at the individual level due to differences in individuals’ experiences. Again, such research was done on more diverse respondent populations, not a specific demographic cohort with distinct social experiences. This finding has implications for the general assumption that within-country differences are considerably larger than across-country differences, when it comes to cultural value studies. Thus, when focusing on a specific demographic generational cohort, it seems that even when it comes to representatives from two very diverse cultural backgrounds, one grounded in a more Western and Protestant-based cultural context with more independent identity construal and the other in a more Eastern and Confucian-based cultural context with more interdependent identity construal, my evidence shows grater tendency towards a generational archetype understanding of young adults.</p>
Recent developments in sequencing technologies have greatly improved our knowledge of phylogenetic relationships and genomic architectures throughout the tree of life. Spiralia, a diverse clade within Protostomia, is essential for understanding the evolutionary history of parasitism, gene conversion, nervous systems and animal body plans. In this review, we focus on the current hypotheses of spiralian phylogeny and investigate the impact of long-read sequencing on the quality of genome assemblies. We examine chromosome-level assemblies to highlight key genomic features that have driven spiralian evolution, including karyotype, synteny and the Hox gene organization. In addition, we show how chromosome rearrangement has influenced spiralian genomic structures. Although spiralian genomes have undergone substantial changes, they exhibit both conserved and lineage-specific features. We recommend increasing sequencing efforts and expanding functional genomics research to deepen insights into spiralian biology.

KEGG pathway analysis revealed that 6,888 transcripts could be assigned to 102 significantly enriched pathways in 3 main categories (metabolism, genetic information processing, environmental information processing). As expected, most (78.24%) of these DEGs were down-regulated in un-melanized eggs. Surprisingly, the majority of genes associated with the pigmentation GO term were up-regulated in the pink egg samples; however, two key terminal genes, laccase2 and DCE/Yellow, were significantly down-regulated, and further verified by qPCR.
