Climate change, and in particular the increase in temperature we are currently observing, can affect herbivorous insects. Aphids, as poikilothermic organisms, are directly exposed to temperature increases that influence their metabolism. Heat stress causes disturbances between the generations and the neutralization of reactive oxygen species (ROS). The aim of this work is focused on explaining how the aphid, using the example of Aphis pomi, responds to abiotic stress caused by temperature increase. The experiment was carried out under controlled conditions at three temperatures: 20, 25, and 28 °C. In the first stage, changes in the activity of enzymatic markers (superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), β-glucosidase, polyphenol oxidase (PPO), and peroxidase (POD)) were determined in aphid tissues, at each temperature. In the second stage, microcalorimetry monitored changes in heat emitted by aphids, at each temperature. Our results showed that A. pomi defense responses varied depending on temperature and were highest at 28 °C. The flexible activity of enzymes and increase in the metabolic rate played the role of adaptive mechanisms and ran more effectively at higher temperatures. The A. pomi thus protected itself against ROS excessive induction and the aphids were able to respond quickly to environmental stress.
Despite many studies of the aging process, questions about key factors ensuring longevity have not yet found clear answers. Temperature seems to be one of the most important factors regulating lifespan. However, the genetic background may also play a key role in determining longevity. The aim of this study was to investigate the relationship between the temperature, genetic background (fruit fly origin), and metabolic rate on lifespan. Experiments were performed with the use of the wild type Drosophila melanogaster fruit flies originating from Australia, Canada, and Benin and the reference OregonR strain. The metabolic rate of D. melanogaster was measured at 20 °C, 25 °C, and 28 °C in an isothermal calorimeter. We found a strong negative relationship between the total heat flow and longevity. A high metabolic rate leads to increased aging in males and females in all strains. Furthermore, our results showed that temperature has a significant effect on fecundity and body weight. We also showed the usefulness of the isothermal calorimetry method to study the effect of environmental stress conditions on the metabolic activity of insects. This may be particularly important for the forecasting of impact of global warming on metabolic activity and lifespan of various insects.
Temperature, being the main factor that has an influence on insects, causes changes in their development, reproduction, winter survival, life cycles, migration timing, and population dynamics. The effects of stress caused by a temperature increase on insects may depend on many factors, such as the frequency, amplitude, duration of the stress, sex, or the developmental stage of the insect. The aim of the study was to determine the differences in the enzymatic activity of nymphs and adult aphids Aphis pomi, Macrosiphum rosae and Cinara cupressi, and changes in their response to a temperature increase from 20 to 28 °C. The activity of enzymatic markers (superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), β-glucosidase, polyphenol oxidase (PPO) and peroxidase (POD)) in aphid tissues was analysed for three constant temperatures. The results of our research showed that the enzymatic activity of aphids (measured as the activity of antioxidant, detoxifying and oxidoreductive enzymes) was mainly determined by the type of morph. We observed a strong positive correlation between the activity of the detoxifying and oxidoreductive enzymes and aphids’ development, and a negative correlation between the activity of the antioxidant enzymes and aphids’ development. Moreover, the study showed that an increase in temperature caused changes in enzyme activity (especially SOD, CAT and β-glucosidase), which was highest at 28 °C, in both nymphs and adults. Additionally, a strong positive correlation between metabolic activity (heat flow measured by microcalorimeter) and longevity was observed, which confirmed the relationship between these characteristics of aphids. The antioxidant enzyme system is more efficient in aphid nymphs, and during aphid development the activity of antioxidant enzymes decreases. The antioxidant enzyme system in aphids appears to deliver effective protection for nymphs and adults under stressful conditions, such as high temperatures.
Given the deficit of works describing the impact of silver nanoparticles (AgNPs) on the growth of crop plants, these studies were devoted to the determination of the influence of the surface properties of charge-stabilized AgNPs on induced phytotoxic effects. Three types of AgNPs of similar size distribution, but of diverse surface properties, were synthesized using trisodium citrate, sodium hexametaphosphate and tannic acid. The suspensions and solution of silver nitrate were applied in a hydroponic culture in order to assess their influence on the germination of Triticum aestivum seeds. Studies of the damage of the cell membrane and the activity of antioxidant enzymes were carried out on the shoots and roots of the seedlings. The differences in the plant condition were assessed while analyzing photosystem II. The results showed that the AgNPs stabilized with sodium hexametaphosphate exhibited a higher toxicity toward the tested plants than silver ions with the same concentration. It was found that the AgNPs obtained using tannic acid exerted an adverse effect on the crop plants. The citrate-stabilized AgNPs were the least toxic to the seedlings. The obtained results demonstrated that the surface properties of AgNPs strongly influence the phytotoxicity toward T. aestivum seedlings.
Light-emitting diodes (LEDs) and high-pressure sodium lamps (HPS) are among the most commonly used light sources for plant cultivation. The objective of this study was to evaluate the effect of two controlled-environment production systems differing in light sources on growth, photosynthetic activity, and secondary metabolism of common buckwheat. We hypothesized that LED light with the majority of red and blue waves would increase physiological and biochemical parameters compared to sunlight supplemented with HPS lamps. The experiment was performed in a phytotronic chamber (LEDs) and in a greenhouse (solar radiation supplemented with HPS lamps as a control). The effects were analyzed at the flowering phase with biometric measurements, leaf chlorophyll index, the kinetics of chlorophyll a fluorescence, content of soluble carbohydrates and phenolics in the leaves. Applied LED light decreased the biomass but stimulated the production of phenolics compared to control plants. In control plants, a positive correlation between flavonoid content and energy dissipation from photosystem II (DIo/CSm) was found, while in plants under LEDs total pool of phenolic content correlated with this parameter and the quantum yield of electron transport (φ Ro and ψ Ro) was lower than that of the control, probably affecting buckwheat biomass.
The spectrum of light significantly influences the growth of plants cultivated in closed systems. Five lettuce cultivars with different leaf colours were grown under white light (W, 170 μmol m−2 s−1) and under white light with the addition of red (W + R) or blue light (W + B) (230 μmol m−2 s−1). The plants were grown until they reached the seedling phase (30 days). Each cultivar reacted differently to the light spectrum applied. The red-leaved cultivar exhibited the strongest plasticity in response to the spectrum. The blue light stimulated the growth of the leaf surface in all the plants. The red light negatively influenced the length of leaves in the cultivars, but it positively affected their number in red and dark-green lettuce. It also increased the relative chlorophyll content and fresh weight gain in the cultivars containing anthocyanins. When the cultivars were grown under white light, they had longer leaves and higher value of the leaf shape index. The light-green cultivars had a greater fresh weight. Both the addition of blue and red light significantly increased the relative chlorophyll content in the dark-green cultivar. The spectrum enhanced with blue light had positive influence on most of the parameters under analysis in butter lettuce cultivars. These cultivars were also characterised by the highest absorbance of blue light.
Ageing is accompanied by dramatic changes in chromatin structure organization and genome function. Two essential components of chromatin, the linker histone Hho1p and actin-related protein 4 (Arp4p), have been shown to physically interact in Saccharomyces cerevisiae cells, thus maintaining chromatin dynamics and function, as well as genome stability and cellular morphology. Disrupting this interaction has been proven to influence the stability of the yeast genome and the way cells respond to stress during chronological ageing. It has also been proven that the abrogated interaction between these two chromatin proteins elicited premature ageing phenotypes. Alterations in chromatin compaction have also been associated with replicative ageing, though the main players are not well recognized. Based on this knowledge, here, we examine how the interaction between Hho1p and Arp4p impacts the ageing of mitotically active yeast cells. For this purpose, two sets of strains were used—haploids (WT(n), arp4, hho1Δ and arp4 hho1Δ) and their heterozygous diploid counterparts (WT(2n), ARP4/arp4, HHO1/hho1Δ and ARP4 HHO1/arp4 hho1Δ)—for the performance of extensive morphological and physiological analyses during replicative ageing. These analyses included a comparative examination of the yeast cells’ chromatin structure, proliferative and reproductive potential, and resilience to stress, as well as polysome profiles and chemical composition. The results demonstrated that the haploid chromatin mutants arp4 and arp4 hho1Δ demonstrated a significant reduction in replicative and total lifespan. These findings lead to the conclusion that the importance of a healthy interaction between Arp4p and Hho1p in replicative ageing is significant. This is proof of the concomitant importance of Hho1p and Arp4p in chronological and replicative ageing.
