Three new polyether‐tethered dinickel–salphen complexes (2 a–c) have been synthesized and fully characterized by NMR spectroscopy, mass spectrometry, and elemental analyses. The binding affinity and selectivity of these complexes and of the parent mono‐nickel complex (1) towards dimeric quadruplex DNA have been determined by UV/Vis titrations, fluorescence spectroscopy, CD spectroscopy, and electrophoresis. These studies have shown that the dinickel–salphen complex with the longest polyether linker (2 c) has higher binding affinity and selectivity towards dimeric quadruplexes (over monomeric quadruplexes) than the dinickel–salphen complexes with the shorter polyether linkers (2 a and 2 b). Complex 2 c also has higher selectivity towards human telomeric dimeric quadruplexes with one TTA linker than the monometallic complex 1. Based on the spectroscopic data, a possible binding mode between complex 2 c and the dimeric G‐quadruplex DNA under study is proposed.
Soft actuators have received intensive attention in the fields of soft robots, sensors, intelligent control, artificial intelligence, and visual intelligence. By combination of tensile and torsional deformations, different types of motions can be realized, such as bending, rolling, and jumping. Soft robotics need soft actuators, such as artificial muscles to lift or move objects to perform some work. Additionally, actuation integrated with functions of sensing, signal transmission, and control is also needed in the development of advanced intelligent systems, which further stimulates the requirement of multifunctional actuators. Here different types of soft actuators that can perform tensile and torsional actuations are summarized, including twisted fiber artificial muscles, shape memory polymers, hydrogels, liquid crystal polymers, electrochemical actuators with conducting polymers, and some natural materials. Examples are also included regarding the bending or rolling deformations of the actuators for lifting objects. Then, recent interesting reports about multifunctional soft actuators combined with sensing and signal transmission performances, are summarized. Last, a summary of different ways to realize tensile and torsional actuations, different materials, and designs for lifting or moving objects, as well as construction of multifunctional actuators with actuation and sensing functions is provided.
In chronic infectious diseases caused by gram-negative bacteria, such as osteomyelitis, septic arthritis, and periodontitis, osteoclastic activity is enhanced with elevated inflammation, which disturbs the bone homeostasis and results in osteolysis. Lipopolysaccharide (LPS), as a bacteria product, plays an important role in this process. Recent evidence shows that an antimalarial drug artesunate attenuates LPS-induced osteolysis independent of RANKL. In this study we evaluated the effects of artesunate on LPS-induced osteoclastogenesis in vitro and femur osteolysis in vivo, and explored the mechanisms underlying the effects of artesunate on LPS-induced osteoclast differentiation independent of RANKL. In preosteoclastic RAW264.7 cells, we found that artesunate (1.56−12.5 μM) dose dependently inhibited LPS-induced osteoclast formation accompanied by suppressing LPS-stimulated osteoclast-related gene expression (Fra-2, TRAP, Cathepsin K, β3-integrin, DC-STAMP, and Atp6v0d2). We showed that artesunate (3.125−12.5 µM) inhibited LPS-stimulated nuclear factor of activated T cells c1 (NFATc1) but not NF-κB transcriptional activity; artesunate (6.25, 12.5 μM) significantly inhibited LPS-stimulated NFATc1 protein expression. Furthermore, artesunate treatment markedly suppressed LPS-induced Ca
2+
influx, and decreased the expression of PP2B-Aα (calcineurin) and pPLCγ1 in the cells. In addition, artesunate treatment significantly decreased the expression of upstream signals TLR4 and TRAF6 during LPS-induced osteoclastogenesis. Administration of artesunate (10 mg/kg, ip) for 8 days effectively inhibited serum TNF-α levels and ameliorated LPS (5 mg/kg, ip)-induced inflammatory bone loss in vivo. Taken together, artesunate attenuates LPS-induced inflammatory osteoclastogenesis by inhibiting the expression of TLR4/TRAF6 and the downstream PLCγ1-Ca
2+
-NFATc1 signaling pathway. Artesunate is a valuable choice to treat bone loss induced by gram-negative bacteria infection or inflammation in RANKL-independent pathway.
Zika virus (ZIKV) causes congenital neurologic birth defects, notably microcephaly, and has been associated with other serious complications in adults. The virus has been detected in human breast milk and possible transmissions via breastfeeding have been reported. Breast milk is rich in nutrients and bio-active substances that might directly affect viral infectivity. Thus, here, we analyzed the effect of human breast milk on ZIKV infection. We observed that fresh human breast milk had no effect on ZIKV, but found that upon storage, milk effectively suppressed infection. The antiviral activity is present in the fat-containing cream fraction of milk and results in the destruction of the structural integrity of viral particles, thereby abrogating infectivity. The release of the factor is time dependent but varies with donors and incubation temperatures. The viral titer of milk that was spiked with ZIKV decreased considerably upon storage at 37 °C for 8 h, was lost entirely after 2 days of 4 °C storage, but was not affected at −20 °C. This suggests that cold storage of milk inactivates ZIKV and that the antiviral factor in milk may also be generated upon breastfeeding and limit this transmission route of ZIKV.
A microfluidic manipulation system that can sense a liquid and control its flow is highly desirable. However, conventional sensors and motors have difficulty fitting the limited space in microfluidic devices; moreover, fast sensing and actuation are required because of the fast liquid flow in the hollow fibre. In this study, fast torsional and tensile actuators were developed using hollow fibres employing spiral nonlinear stress, which can sense the fluid temperature and sort the fluid into the desired vessels. The fluid-driven actuation exhibited a highly increased response speed (27 times as fast as that of air-driven actuation) and increased power density (90 times that of an air-driven solid fibre actuator). A 0.5 K fluid temperature fluctuation produced a 20° rotation of the hollow fibre. These high performances originated from increments in both heat transfer and the average bias angle, which was understood through theoretical analysis. This work provides a new design strategy for intelligent microfluidics and inspiration for soft robots and smart devices for biological, optical, or magnetic applications.
