Many cellular processes in cancer are attributed to kinase signaling networks. V-akt murine thymoma viral oncogene homolog (AKT) plays a major role in the PI3K/AKT signaling pathways. AKT is activated by PI3K or phosphoinositidedependent kinases (PDK) as well as growth factors, inflammation, and DNA damage. Signal transduction occurs through downstream effectors such as mTOR, glycogen synthase kinase 3 beta (GSK3b), or forkhead box protein O1 (FOXO1). The abnormal overexpression or activation of AKT has been observed in many cancers, including ovarian, lung, and pancreatic cancers, and is associated with increased cancer cell proliferation and survival. Therefore, targeting AKT could provide an important approach for cancer prevention and therapy. In this review, we discuss the rationale for targeting AKT and also provide details regarding synthetic and natural AKT-targeting compounds and their associated studies.

Abstract. Knowledge of the physical state and morphology of internally mixed organic/inorganic aerosol particles is still largely uncertain. To obtain more detailed information on liquid-liquid phase separation (LLPS) and morphology of the particles, we investigated complex mixtures of atmospherically relevant dicarboxylic acids containing 5, 6, and 7 carbon atoms (C5, C6 and C7) having oxygen-to-carbon atomic ratios (O:C) of 0.80, 0.67, and 0.57, respectively, mixed with ammonium sulfate (AS). With micrometer-sized particles of C5/AS/H 2 O, C6/AS/H 2 O and C7/AS/H 2 O as model systems deposited on a hydrophobically coated substrate, laboratory experiments were conducted for various organic-to-inorganic dry mass ratios (OIR) using optical microscopy and Raman spectroscopy. When exposed to cycles of relative humidity (RH), each system showed significantly different phase transitions. While the C5/AS/H 2 O particles showed no LLPS with OIR = 2:1, 1:1 and 1:4 down to 20 % RH, the C6/AS/H 2 O and C7/AS/H 2 O particles exhibit LLPS upon drying at RH 50 to 85 % and ∼90 %, respectively, via spinodal decomposition, growth of a second phase from the particle surface or nucleation-and-growth mechanisms depending on the OIR. This suggests that LLPS commonly occurs within the range of O:C < 0.7 in tropospheric organic/inorganic aerosols. To support the comparison and interpretation of the experimentally observed phase transitions, thermodynamic equilibrium calculations were performed with the AIOMFAC model. For the C7/AS/H 2 O and C6/AS/H 2 O systems, the calculated phase diagrams agree well with the observations while for the C5/AS/H 2 O system LLPS is predicted by the model at RH below 60 % and higher AS concentration, but was not observed in the experiments.Both core-shell structures and partially engulfed structures were observed for the investigated particles, suggesting that such morphologies might also exist in tropospheric aerosols.

Atmospheric aerosol particles may undergo liquid‐liquid phase separation (LLPS) when exposed to varying relative humidity. In this study we investigated the occurrence of LLPS for mixtures consisting of up to ten organic compounds, ammonium sulfate, and water in relationship with the organic oxygen‐to‐carbon (O:C) ratio. LLPS always occurred for O:C < 0.56, never occurred for O:C > 0.80, and depended on the specific types and compositions of organic functional groups in the regime 0.56 < O:C < 0.80. In the intermediate regime, mixtures with a high share of aromatic compounds shifted the limit of occurrence of LLPS to lower O:C ratios. The number of mixture components and the spread of the O:C range did not notably influence the conditions for LLPS to occur. Since in ambient aerosols O:C range typically between 0.2 and 1.0, LLPS is expected to be a common feature of tropospheric aerosols.

Despite major progress in the understanding of properties of tropospheric aerosol particles, it remains challenging to understand their physical state and morphology. To obtain more detailed knowledge of the phases, phase transitions and morphologies of internally mixed organic/inorganic aerosol particles, we evaluated liquid-liquid phase separation (LLPS), deliquescence relative humidity (DRH) and efflorescence relative humidity (ERH) of 33 organic/ammonium sulfate (AS)/H2O systems from our own and literature data. The organic fraction consists of single compounds or mixtures with up to ten aliphatic and/or aromatic components with carboxylic acid, hydroxyl, carbonyl, ether, and ester functionalities, covering O : C ratios between 0.29 and 1.33. Thirteen out of these 33 systems did not show LLPS for any of the studied organic-to-inorganic mixing ratios, sixteen underwent LLPS showing core-shell morphology, and four showed both core-shell and partially engulfed configurations depending on the organic-to-inorganic ratio and RH. In all cases the organic fractions of the systems with partially engulfed configurations consisted of dicarboxylic acids. AS in mixed organic/AS/H2O particles deliquesced between 70 and 84% RH. AS effloresced below 58% RH or remained in a one-liquid-phase state. AS in droplets with LLPS always showed efflorescence with ERH between 30 and 50% RH, providing clear evidence that the presence of LLPS facilitates AS efflorescence. Spreading coefficients of the organic-rich phase on the AS-rich phase for systems containing polyethylene glycol 400 (PEG-400) and a mixture of dicarboxylic acids are in agreement with the optically observed morphologies of droplets deposited on the hydrophobic substrate. Analysis of high resolution elastic Mie resonance spectra allowed the detection of LLPS for single levitated droplets consisting of PEG-400/AS/ H2O, whereas LLPS was difficult to detect in (2-methylglutaric acid + 3-methylglutaric acid + 2,2-dimethylsuccinic acid)/AS/H2O. Measured Mie spectra of PEG-400/AS/H2O at 93.5% and at 80.9% RH agreed with computed Mie spectra for a homogeneous and a core-shell configuration, respectively, confirming the results obtained from droplets deposited on a hydrophobic substrate. Based on the presented evidence, we therefore consider the core-shell morphology to be the prevalent configuration of liquid-liquid-phase-separated tropospheric organic/AS/H2O particles.

Abstract. Oxidation of isoprene is an important source of secondary organic material (SOM) in atmospheric particles, especially in areas such as the Amazon Basin. Information on the viscosities, diffusion rates, and mixing times within isoprene-derived SOM is needed for accurate predictions of air quality, visibility, and climate. Currently, however, this information is not available. Using a bead-mobility technique and a poke-flow technique combined with fluid simulations, the relative humidity (RH)-dependent viscosities of SOM produced from isoprene photo-oxidation were quantified for 20-60 µm particles at 295 ± 1 K. From 84.5 to 0 % RH, the viscosities for isoprene-derived SOM varied from ∼ 2 × 10 −1 to ∼ 3 × 10 5 Pa s, implying that isoprenederived SOM ranges from a liquid to a semisolid over this RH range. These viscosities correspond to diffusion coefficients of ∼ 2 × 10 −8 to ∼ 2 × 10 −14 cm 2 s −1 for large organic molecules that follow the Stokes-Einstein relation. Based on the diffusion coefficients, the mixing time of large organic molecules within 200 nm isoprene-derived SOM particles ranges from approximately 0.1 h to less than 1 s. To illustrate the atmospheric implications of this study's results, the Amazon Basin is used as a case study for an isoprenedominant forest. Considering the RH and temperature range observed in the Amazon Basin and with some assumptions about the dominant chemical compositions of SOM particles in the region, it is likely that SOM particles in this area are liquid and reach equilibrium with large gas-phase organic molecules on short time scales, less than or equal to approximately 0.1 h.

Hygroscopic growth and cloud condensation nuclei activation are key processes for accurately modeling the climate impacts of organic particulate matter. Nevertheless, the microphysical mechanisms of these processes remain unresolved. Here we report complex thermodynamic behaviors, including humidity-dependent hygroscopicity, diameter-dependent cloud condensation nuclei activity, and liquid–liquid phase separation in the laboratory for biogenically derived secondary organic material representative of similar atmospheric organic particulate matter. These behaviors can be explained by the non-ideal mixing of water with hydrophobic and hydrophilic organic components. The non-ideality-driven liquid–liquid phase separation further enhances water uptake and induces lowered surface tension at high relative humidity, which result in a lower barrier to cloud condensation nuclei activation. By comparison, secondary organic material representing anthropogenic sources does not exhibit complex thermodynamic behavior. The combined results highlight the importance of detailed thermodynamic representations of the hygroscopicity and cloud condensation nuclei activity in models of the Earth’s climate system.

Abstract. To improve predictions of air quality, visibility, and climate change, knowledge of the viscosities and diffusion rates within organic particulate matter consisting of secondary organic material (SOM) is required. Most qualitative and quantitative measurements of viscosity and diffusion rates within organic particulate matter have focused on SOM particles generated from biogenic volatile organic compounds (VOCs) such as α-pinene and isoprene. In this study, we quantify the relative humidity (RH)-dependent viscosities at 295 ± 1 K of SOM produced by photo-oxidation of toluene, an anthropogenic VOC. The viscosities of toluene-derived SOM were 2 × 10 −1 to ∼ 6 × 10 6 Pa s from 30 to 90 % RH, and greater than ∼ 2 × 10 8 Pa s (similar to or greater than the viscosity of tar pitch) for RH ≤ 17 %. These viscosities correspond to Stokes-Einstein-equivalent diffusion coefficients for large organic molecules of ∼ 2 × 10 −15 cm 2 s −1 for 30 % RH, and lower than ∼ 3 × 10 −17 cm 2 s −1 for RH ≤ 17 %. Based on these estimated diffusion coefficients, the mixing time of large organic molecules within 200 nm toluene-derived SOM particles is 0.1-5 h for 30 % RH, and higher than ∼ 100 h for RH ≤ 17 %. As a starting point for understanding the mixing times of large organic molecules in organic particulate matter over cities, we applied the mixing times determined for toluene-derived SOM particles to the world's top 15 most populous megacities. If the organic particulate matter in these megacities is similar to the toluene-derived SOM in this study, in Istanbul, Tokyo, Shanghai, and São Paulo, mixing times in organic particulate matter during certain periods of the year may be very short, and the particles may be wellmixed. On the other hand, the mixing times of large organic molecules in organic particulate matter in Beijing, Mexico City, Cairo, and Karachi may be long and the particles may not be well-mixed in the afternoon (15:00-17:00 LT) during certain times of the year.
