Eine Reihe von quadratisch‐planaren Dithiocarbamato‐Komplexen des Typs (R′CO2R″)2 (M = Ni, Pd, Pt; R″ = H, Et) mit verschiedenen α‐Aminosäuren als Aminkomponenten wird beschrieben und 1H‐NMR‐ und IR‐spektroskopisch charakterisiert.
cis-Dichlorodipeptide esterplatinum complexesCl2Pt(MetGlyOEt),Cl2Pt(EthionylGlyOEt), Cl2Pt(GlyGlyOEt)2 and Cl2Pt(GlySerOEt)2 are prepared from the α-amino acid complexes by peptide synthesis using platinum as an amino protecting group. cis-Cl2Pt(GlyGlyOEt)2 and cis-Cl2Pt(GlySerOEt)2 have been prepared also directly from K2PtCl4 and the dipeptidesters. cis-Cl2Pt(GlyGlyOEt)2 (2 a) and cis-Cl2Pt(NH3)2 (5) lead to a prefered inhibition of the DNA-synthesis of sarcoma 180, Yoshida-sarcoma and Walker-256-carcinosarcoma in vitro; RNA- and protein biosynthesis are influenced to a much lower degree. 2a and 5 cause filamentous growth in Escherichia coli B. The DNA polymerase deficient strain of E. coli, p 3478 pol A-, is more inhibited by 2 a and 5 than the non deficient strain W 3110 pol A+. Tumor growth of di-2-chloro-ethylmethylamine (HN2) resistant sarcoma 180 and of Yoshida sarcoma is weakly inhibited, whereas Walker-256-carcinosarcoma is markedly inhibited; however 2a and 5 show similar inhibition of the same tumor.

In vitro, a marked effect on the DNA synthesis of mammary tumor cells and an inhibition of the 'H-E, receptor interaction can be. demonstrated. A binding to DNA and an inhibition of the proliferation stimulating effect of endogenous estrogens by blocking the hormone receptors are discussed as modes of action.

Reactions of the a-Methylene Group in (Glycinato)platinum(II) Chelate ComplexesThe reaction of the glycinato complex (n-Bu,P)Pt(Cl)(NH2CHzC0,) (1) with aldehydes or alkylating agents (dimethyl sulfate, benzyl chloride) affords the chelate compounds 3 -6 with substituted a-amino acids. Die a-Methylen-Gruppe von Glycinat wird durch Chelatbindung an ein Metall-Ion aktiviert. Akabori et al. beschrieben erstmals Aldolkondensationen an (Glycinato) kupfer(I1)-Komplexen; z. B. fuhrt die Umsetzung mit Acetaldehyd zu Threonin. Weitere Reaktionen an der CHz-Gruppe von koordiniertem Glycinat oder Glycin-Schiffbasen sind in Lit. zusammengefal3t. Durch Reaktion an optisch aktiven (Glycinato)kobalt(III) Komplexen gelang die asymmetrische Synthese von Threonin ' 1. In Fortfuhrung unserer Arbeiten an Platin(I1kkoordinierten Aminosauren4) wird irn folgenden iiber Reaktionen an der a-Methylengruppe des Glycinat-Chelatliganden in Platinkomplexen berichtet. Komplexe wie z. B. Pt(NH2CH2C02)2 ' ) oder (CzH4)Pt(CI)(NH2CH2CO~)6' erweisen sich wegen ihrer geringen Loslichkeit bzw. Stabilitat im alkalischen Medium fur solche Reaktionen als nicht geeignet. Brauchbdr ist dagegen der Tributylphosphin-haltige Komplex 1, der aus der CI-verbruckten Verbindung ( n -B~~P )~P t~c l~ ' ) mit Glycinat unter Spaltung der C1-Brucken und Substitution eines Chlorids zuganglich ist .
