Research institutions have emphasized alternative disease control agents because they represent a great option with beneficial effects on human and animal health and ecosystem balance. Despite the studies already done on botanical fungicides, their effective use requires knowledge about the applicability of natural products to different production systems. The main objective of this study is to evaluate the efficacy of essential oils of Ocimum pupuraceus L., Cymbopogon nardus (L.) Rendle, Cymbopogon citratus (DC.) Stapf, and Lippia sidoides Cham. on the inhibition of mycelial growth and germination of conidia of B. maydis. The other objectives of this study were to perform gas chromatographic and phytotoxicity analyses, and test the control of bipolaris leaf spot using essential oils of L. sidoides applied as a preventive and curative agent. Among the treatments studied, the essential oil of L. sidoides is effective in inhibiting mycelial growth and conidial germination at the concentrations of 5 and 1%, respectively. The main constituent of the oil is thymol (92.68%). The concentration range 0.75-3% of L. sidoides essential oil is phytotoxic to maize plants. Lower values of the area under the progress curve of bipolaris leaf spot are observed at concentrations 0.1-0.5%, when the essential oil was applied as a preventive agent prior to the colonization of plant tissues by the pathogen. The application of the oil as a curative to plants with the disease also shows efficacy at the concentration 0.1%, reducing the severity by more than 54%. These results demonstrate the potential effects of L. sidoides essential oil on preventive and curative control of bipolaris leaf spot in maize and mycelial growth.
MORPHOLOGICAL AND MOLECULAR IDENTIFICATION OF Curvularia sp. CAUSAL AGENT OF CORN LEAF STAINABSTRACT -Fungi of the genus Curvularia are widely distributed around the world and may be associated with plant species, in the saprophytic, endophytic or parasitic form. Species of this genus are responsible for several diseases in cultivated plants, causing mainly foliar stains. The correct identification of the species is important for the diseases knowledge and management, being these identifications based mainly on morphological characteristics as aspects of the mycelia, conidia forms and size, hyphae morphology. However, molecular techniques such as the amplification of specific genes, as well as digestion pattern analyzes of ITS regions through restriction enzymes, have contributed to the identification and genetic differentiation between species. In the present study, the morphological and molecular characterizations of Curvularia isolates from infected corn plants were realized. The isolates were grouped according to the morphological and molecular identification, all of which presented the same DNA amplification profile.Morphological and molecular techniques allowed the characterization of a single species, Curvularia lunata.
High temperatures can negatively affect lettuce production by promoting early bolting, which leads to increased levels of latex accumulation in the leaves, causing them to become bitter. The lack of adaptation of this culture to such conditions made it an object of study for plant breeding programs, resulting in well succeeded studies. However, little is known about the genes that regulate lettuce flowering. A better understanding of the complex genic interactions involved in the process of lettuce floral initiation is of great importance, since it can enable the development of late-bolting cultivars through plant genetic transformation. MADS-box transcriptional factors are key flowering regulators and have been extensively studied during the flowering process in several species. Thus, this study aimed to identify and characterize the Lettuce MADS-box gene family through the use of bioinformatics tools. The computational analysis consisted in gene prediction, alignment, and phylogenetic analysis. 91 sequences of putative MADS-box genes were identified and characterized by a phylogenetic study of 20 MADS-box genes. Future studies comprising mutants for these genes in plant model species and in lettuce will enable a better understanding of the functions performed by these genes during lettuce flowering, as well as, a better comprehension of this process.
