Koronavirüsler ilk olarak 1930'larda keşfedilmiştir. SARS ve MERS salgınlarından sonra ortaya çıkan COVID-19 pandemisi kısa sürede çok fazla enfeksiyon ve ölüme neden oldu. Koronavirüsler en büyük RNA genomuna sahip virüslerdir. İçerdiği glikoproteinlerden kaynaklı mikroskop altında taç görünümüne sahiptir. COVID-19 enfeksiyonunun spesifik semptomları olmamasına rağmen ateş, öksürük, balgam, miyalji ve baş ağrısı en belirgin semptomlarıdır. Teşhisinde en yaygın kullanılan yöntem PCR testidir. Henüz standart bir tedavisi bulunmayan COVID-19 enfeksiyonunda denenen tedavilerden biri de kök hücre tabanlı tedavilerdir.Bu derlemede COVID-19'un genel epidemiyolojisi, genomik yapısı ve COVID-19 için kök hücre tedavilerinin önemi irdelenecektir.
Objective: Mesenchymal stem cells (MSCs) are also promising in immunosuppressed patients after organ and tissue transplantation, in addition to their current wide range of uses and research areas. Sunitinib is a receptor tyrosine kinase with immunosuppressive properties and its cytotoxic activity in different types of cells is known. Our study aimed to elucidate the effect of oxytocin on sunitinib-treated MSCs. Methods: For this purpose, commercially available rat adipose tissue-derived MSC (ADMSCs) was used. The individual or combinational effect of the active substances on viability was evaluated with WST-1, the effect on apoptosis Annexin V, the effect on oxidative stress markers MDA, CAT, GPX, and SOD ELISA tests. Results: The IC50 value of sunitinib was determined as 44.57 μM at the 48th hour, and it was determined that oxytocin had no cytotoxic effect in doses up to 100 μM. Treatment of the two agents in combination increased the cytotoxic effect of sunitinib. Oxytocin attenuated the effect of sunitinib on apoptosis and lipid peroxidation. Conclusion: It is important to investigate the efficacy of these two substances individually and in combination with ADMSCs with further experiments to evaluate the potential use of oxytocin in organ and tissue transplantations.
ÖZET olarak adlandırılan SARS-CoV-2 (Şiddetli akut solunum yolu sendromu koronavirüs-2) virüsünün neden olduğu enfeksiyon başlangıçta Aralık 2019'da Çin'de tespit edilmiştir ve daha sonra dünyaya hızla yayılmıştır. 13 Ocak 2020 tarihinde Tayland Halk Sağlığı Bakanlığı Çin'in Wuhan şehrinde yaşayan 8 Ocak 2020 tarihinde Tayland'a giden 61 yaşında Çinli bir kadında ilk importe olguyu bildirmiştir. Daha sonra 11 Mart'ta Dünya Sağlık Örgütü (DSÖ) bu salgını küresel bir pandemi ilan etmiştir. Hastalığın yayılmasını önlemek ve pandemiyi kontrol etmek için ilaçların etkinliği araştırılmakta olup çok sayıda COVID-19 aşı adayı ve ilaç adayı geliştirilmektedir. ABD-Gıda ve İlaç İdaresi (FDA), Aralık 2020'de sırasıyla Pfizer ve Moderna tarafından geliştirilen iki mRNA aşısı için acil kullanım onayı yayınlamıştır. Şu anda geliştirilmekte olan diğer COVID-19 aşıları mRNA, DNA, viral vektör, subünite, inaktive edilmiş ve canlı zayıflatılmış aşıları kapsayan çeşitli platformlara dayanmaktadır. Bu derlemede COVID-19 tanı ve/veya tedavisinde kullanılan testler, aşılar ve ilaçların rolünü incelenmektedir.
