Bakteri, maya, küf ve mavi yeşil algler tarafından salgılanan EPS (ekzopolisakkarit veya ekstraselüler polimerik madde), proteinler, eDNA (extracellular DNA, hücre dışı DNA), çeşitli enzimler ve mikroorganizmanın kendisini içeren kompleks yapıdaki matrikse "biyofilm" denir. Biyofilm mikroorganizmaların kendini başta zorlu veya toksik koşullardan korumakta, ortamlarda mikrobiyal rekabet gücünü artırmakta ve bazı hücresel çeşitli işlevlerde de kullanılmaktadır. Biyofilmler, ekolojik olarak koruyucu ve işlevseldir. Endüstride (gıda, tekstil, kağıt ve madencilik gibi), atıkların biyoremediasyonunda kullanılmak üzere farklı bakteri türlerinden oluşan biyofilmlerden yararlanılmaktadır. Ayrıca mikrobiyal enerji üretiminde ve biyogübre üretiminde kullanılan biyofilmler bulunmaktadır. İnsan bağırsağına tutunan bakterilerin oluşturduğu biyofilmler ise gıda kaynaklı patojen mikroorganizmalara karşı koruyucu olarak görev almaktadır. Ancak biyofilm, dişlerde plak oluşumu ve doku enfeksiyonlarının başlıca kaynağıdır. Ayrıca gıda endüstrisinde de çeşitli sorunlara yol açmaktadır. Özellikle çelik yüzeylerde, boruların iç yüzeylerinde, kullanılan yardımcı alet ve ekipmanlarda gelişerek gıdalarda kirlilik, patojenite ve bozulmaya neden olmaktadır. Bu durum ciddi ekonomik kayıplara da yol açmaktadır. Bu yüzden gıda endüstrisinde biyofilm oluşumunun başlangıç aşamasında engellenmesi ya da oluşan biyofilm yapısının ortadan kaldırılması gerekmektedir. Biyofilm yapılarının iyi bilinmesi bu yapıların engellenmesinde kullanılacak yöntemlerin uygulanmasında kolaylık sağlamaktadır. Bu derlemede, biyofilm tanımı, gıda ve diğer bazı alanlarda biyofilm oluşumu ve önlenmesi hakkında bilgi verilmiştir.
In this study, clotted cream was produced by adding plasmin (25 μL/L) and plasminogen (75 μg/L) to raw cow milk. The purpose of the study was to determine the effect of plasmin and plasminogen enzymes on some properties of clotted cream during 10‐day storage period. β‐Casein was hydrolyzed by plasmin and plasminogen in samples during storage. In sensory evaluation, appearance and structure scores of clotted cream samples containing plasmin and plasminogen were higher than the control sample during storage. Total aerobic mesophilic microorganism, proteolytic and lipolytic bacteria count in all samples were identified as 2.451–3.320 logcfu/g, 1.929–1.977 logcfu/g, 1.312–1.736 logcfu/g on the 10th day of storage, but psychrophilic bacteria count were determined below 1 logcfu/g during storage. Statistically the difference between samples for plasmin activity, α‐casein, β‐casein, hardness, and spreadability was found to be significant (P<0.05). Plasmin and plasminogen may affect positively on texture of clotted cream.
Practical applications
Clotted cream is a breakfast dairy product consumed in many countries. The effect of plasmin, the natural enzymes of milk, on the quality of the cream is important. This information would be useful, especially to individuals who produce and consume clotted cream.
Bu çalışmada farklı il (Antalya, Konya, Isparta) ve ilçelerden (Kulu, Şarkikaraağaç, Atabey) toplanan farklı habitatlara ait 28 toprak örneğinden 19 miksobakteri suşu izole edilmiştir. İzole edilen 19 miksobakteri arasında Myxococcus sp KK2’nin en yüksek α-amilaz aktivitesine sahip olduğu belirlenmiştir. Protein çöktürülmesi ile ilk uygulamada %40, 65 ve 80 amonyum sülfat konsantrasyonları, ikinci uygulamada ise %20, 40 ve 65 konsantrasyonları kullanılarak enzim saflığı artırılmıştır. Diyaliz ve jel filtrasyon yöntemleri kullanılarak ilk uygulamada elde edilen enzimin aktivitesi 883.27 U’ya ulaşırken enzim saflığı 28.35 kat artmıştır. İkinci uygulama ile ise enzim aktivitesi 1754.99 U olarak saptanmış ve enzim saflığı 810.24 kat artmıştır.
