The human papillomavirus (HPV) vaccine is one of the ways to prevent HPV infection that can lead to cervical cancer, which is the second most common cause of cancer-related death among women. It counts for a high mortality and prevalence rate due to the lack of proper awareness about its risks, transmissions, and especially knowledge about prevention of these infections. This research aims to investigate the level of knowledge about the HPV vaccine among female students at SMA Negeri 3 Jambi City from January to March 2022. This research was conducted by using the descriptive method with a cross-sectional approach. Sampling used a consecutive sampling technique with a total sample of 288 respondents. The sample was collected by giving questionnaires and subsequently analysed with Statistical Product and Service Version 26. The research findings showed that the majority of respondents (45.13%) had an inadequate level of knowledge about the HPV vaccine. Whilst 28.1% of respondents had a good level of knowledge and 26.7% had enough knowledge about the HPV vaccine. This research concluded that the level of knowledge among female students at SMA Negeri 3 Jambi City about the HPV vaccine was inadequate. Penyebab tingginya kejadian dan kematian akibat infeksi HPV adalah kurangnya pengetahuan akan bahaya, cara penyebaran, dan khususnya pengetahuan akan pencegahan terhadap infeksi tersebut. Penelitian ini bertujuan mengetahui pengetahuan siswi di SMA Negeri 3 Kota Jambi mengenai vaksin HPV pada bulan Januari hingga Maret 2022. Penelitian ini menggunakan metode deskriptif dengan pendekatan cross-sectional. Pengambilan sampel menggunakan teknik consecutive sampling dengan jumlah sampel sebanyak 288 responden Pengumpulan data dilakukan dengan menggunakan kuesioner dan kemudian dianalisis dengan program statistik SPSS (Statistical Product and Service Solutions) versi 26. Hasil penelitian menunjukkan bahwa tingkat pengetahuan responden mengenai vaksin HPV mayoritas berada dalam kategori kurang, yaitu sebesar 45,13%. Sementara kategori baik sebanyak 28,1%, dan kategori cukup sebanyak 26,7%. Kesimpulan dari penelitian didapatkan siswi SMA Negeri 3 Kota Jambi mempunyai pengetahuan kurang mengenai vaksin HPV.
