Introdução: O vírus HPV está associado a 90% dos casos de câncer de colo de útero e apesar de existir 200 tipos diferentes deste vírus, apenas dois possuem o maior índice de incidência, são eles: HPV 16 e o 18. Objetivo: O presente trabalho teve como intuito identificar os principais fatores genéticos virais que estão relacionados ao surgimento do câncer de colo uterino. O estudo é caracterizado como uma revisão sistemática e analisa os mecanismos de ação de cada fator genético e as principais consequências a nível celular que são essenciais para o desenvolvimento da neoplasia, destacando também os sinais e sintomas resultantes. Metodologia: Para a construção do trabalho, foram utilizados artigos publicados entre os anos de 2011 e 2021, encontrados em diferentes bases de dados como PubMed e Scielo, além de teses e dissertações de respaldo nacional. Resultados: Os vírus HPV 16 e 18 estão presentes em 70% dos casos deste tipo de câncer, estimativas sugerem que por ano têm-se 570 mil novos casos em todo o mundo. Dos seis genes virais precoces (E1 ,E2, E4, E5, E6 e E7) apenas três (E5, E6 e E7) estão diretamente relacionados ao desenvolvimento da doença. Conclusão: Os denominados oncogenes ou oncoproteínas E5, E6 e E7 bloqueiam os principais mecanismos celulares que têm como função suprimir o surgimento da neoplasia. Com base nos dados, observa-se que há grande importância e necessidade da discussão dos fatores genéticos, possivelmente visando nortear futuras pesquisas relacionadas às abordagens terapêuticas.
