Pittosporum eriocarpum Royle, a medicinally important taxon, is endemic to Uttarakhand region of Himalaya. It has become endangered due to over-collection and the loss of habitats. As raising plants through seeds in this plant is problematic, a reliable protocol for micropropagation using nodal explants has been developed. High shoot regeneration (95%) occurred in MS medium augmented with BA 0.4mg/l in combination IBA 0.6mg/l. In vitro regenerated shoots were rooted in MS medium supplemented with three auxins, of which 0.6 mg/l indole butyric acid proved to be the best for rooting (90%) with maximum number of roots per shoot. Thereafter, rooted plants were hardened and nearly 73% of rooted shoots were successfully acclimatized and established in the field. Start codon targeted (SCoT), inter simple sequence repeats (ISSR) and random amplified polymorphic DNA (RAPD) markers were used to validate the genetic homogeneity amongst nine in vitro raised plantlets with mother plant. DNA fingerprints of in vitro regenerated plantlets displayed monomorphic bands similar to mother plant, indicating homogeneity among the micropropagated plants with donor mother plant. The similarity values were calculated based on SCoT, ISSR and RAPD profiles which ranged from 0.89 to 1.00, 0.91 to 1.00 and 0.95 to 1.00 respectively. The dendrograms generated through Unweighted Pair Group Method with arithmetic mean (UPGMA) analysis revealed 97% similarity amongst micropropagated plants with donor mother plant, thus confirming genetic homogeneity of micropropagated clones. This is the first report on micropropagation and genetic homogeneity assessment of P. eriocarpum. The protocol would be useful for the conservation and large scale production of P. eriocarpum to meet the demand for medicinal formulations and also for the re-introduction of in vitro grown plants in the suitable natural habitats to restore the populations.

Zehneria is a genus of small creepers and climbers from the Old–World Tropics and Subtropics. In its broadest circumscription, it comprises up to 94 species and its centre of diversity is Southeast Asia. Zehneria has been split into several smaller genera but this remained controversial mainly due to morphological plasticity of the species groups and lack of comprehensive molecular data. Here, we use one nuclear DNA (ITS1–5.8S–ITS2) and three plastid DNA regions (trnL–trnF, rpl20–rps12, matK) sequenced for about half of the species in Zehneria and its segregate genera Anangia, Neoachmandra, and Pilogyne to test the monophyly of the genus and analyse the evolutionary history of the group. We find that Zehneria s.l. is monophyletic and splitting is not supported by molecular, morphological or biogeographical evidence. Within Zehneria, we recover two strongly supported clades, one consisting mainly of species placed in Pilogyne, the other including Anangia, Neoachmandra and the type of Zehneria. Both clades comprise African, Asian, and Australian/Pacific Island species, which is evidence for high dispersal capacity in this young bird–dispersed group. For India, we accept the five species Z. bodinieri, Z. hookeriana, Z. maysorensis, Z. odorata, and Z. thwaitesii. For Myanmar, we increase the number of known species from four to eight, including three still unnamed species. The diversity hotspots for the genus, however, remain Indonesia and Thailand with together 25 species.

In this study, we explored the evolutionary history and taxonomic treatment of the Caragana opulens complex taking information from morphological and molecular data. The complex consists of three species, C. opulens, C. licentiana and C. kansuensis. Moreover, the morphological characters currently used to differentiate the species present in the complex have been found insignificant and inconsistent and do not help diagnose the species. For the present study, we investigated its range and sampled 139 accessions from the different populations of the genus Caragana and 17 accessions of the complex. DNA sequence data from one nrDNA ITS and one cpDNA trnH-psbA loci were sequenced and analyzed using Maximum Likelihood and Bayesian methods. The resulting phylogenies were congruent in topologies. Based on morphological and molecular data, it is concluded that all three species of the complex are one of the same with significant morphological variations. Hence C. opulens is accepted as the correct name along with C. licentiana and C. kansuensis as synonyms.

Ex-situ conservation places such as botanical gardens require sufficient soil quality to support introduced species from various phytogeographical zones. The soil quality of the Botanic Garden of Indian Republic (BGIR), Noida, Uttar Pradesh, was evaluated to quantify soil nutrients. The dependency of one nutrient on the other nutrients was investigated using Pearson correlation and Multilinear regression analysis (MLRA). At the 0.05 level of significance, the nutrients Log10S and Log10EC (r = 0.97), N and OC (r = 0.98), Mn and OC (r = 0.97), Mn and N (r = 0.92), Ca and pH (r =  − 0.91), Cu and Fe (r = 0.94) were found to be associated. Correspondence Analysis (C.A.) has been performed to find the association of soil elements with the soil type of study site. The spatial indices like NDVI (Normalized Difference Vegetation Index), EVI2 (Enhanced Vegetation Index), ARVI (Atmospherically Resistant Vegetation Index), NPCRI (Normalized Pigment Chlorophyll Index), RDVI (Renormalized Difference Vegetation Index) have shown significant correlation with the Log10S, Mg, Log10Zn, B and Fe respectively (with respective Pearson correlation coefficient r = 0.88, r =  − 0.90, r =  − 0.93, r = 0.91, r = 0.92 at P < 0.05). ARVI, along with other indices SCI (Soil Composition Index), NDMI (Normalized Difference Moisture Index), and MSAVI (Modified Soil Adjusted Vegetation Index), are also the predictor variables for Log10Zn (r =  − 0.89, r =  − 0.88 r = 0.92 at P < 0.05 respectively). MAVI2 (Moisture Adjusted Vegetation Index) positively correlates with OC, Mn content (r = 0.91, r = 0.93 respectively). MSAVI is negatively interrelated with Ca (r =  − 0.89), SCI is negatively interrelated with Log10 K (r =  − 0.98), BSI (Bare Soil Index) is positively associated with pH (r = 0.91), and negatively with Ca (r =  − 0.93). At the same time, other indices like SAVI (Soil Adjusted Vegetation Index), SATVI (Soil Adjusted Total Vegetation Index), NDWI (Normalized Difference Water Index), and DVI (Difference Vegetation Index) have failed to explain the presence of soil nutrients based on spectral reflectance. This study is important for understanding the changing nutrient status of soil at the conservation site for successfully establishing plants from different phytogeographical zones.
