Aims: This study aimed to evaluate the toxicity and humoral and cellular immune response of three heterologous vaccines against Leishmania infantum, yet containing synthetic peptides from Leishmania major in the experimental model in hamsters. Methods and Results: Through bioinformatics analyses, two Leishmania major Gp63 peptides were predicted and selected for vaccine formulations. Hamsters were divided into four groups, with each group receiving doses of three vaccine formulations containing HLA-DR1 or HLA-A2 peptides plus Montanide TM or both associated with the adjuvant. The animals received three vaccine doses and were evaluated for toxicity after each dose, in addition to being analysed for the production of antibodies and lymphoproliferation on day 211 after the last vaccine dose. Peptides predicted in association with oily adjuvant induced a humoral response and strong lymphoproliferation to Leishmania infantum antigen-specific stimulation.
The Brazilian Ministry of Health determined in 2012 that the official protocol for diagnosis of Canine Visceral Leishmaniasis (CVL) would be the Dual-Path Platform (DPP) for screening, followed by enzyme-linked immunosorbent assay (ELISA) for confirmation. This study evaluated serum samples from 426 dogs from a region in northern Brazil. All samples were tested according to the Official Protocol and the sequence inverting (ELISA followed DPP). Regardless of the pr otocol adopted, prevalence (14.7%) has not changed. The approach using ELISA followed by DPP state that, the number of positive animals in screening was higher compared to the official protocol. Screen the ELISA test could be more appropriate.
Introdução:A Leishmaniose Visceral (LV), transmitida por Lutizomya longipalpis tem se alastrado no mundo. É consenso na comunidade científica a necessidade de uma vacina capaz de quebrar o ciclo de transmissão da Leishmania infantum no reservatório canino, visando a redução de incidências de casos humanos. Objetivo: Avaliar a resposta imune humoral de Hamsters (Mesocricetus auratus) através de um novo candidato vacinal heterólogo contra leishmaniose visceral Canina (LVC), constituído de dois peptídeos sintéticos da protease gp63 da Leishmania major, com elevada predição para MHC-I e II. Material e Método: O desenho dos peptídeos foi feito por meio da predição de epítopos da gp63 da Leishmania major restritos ao MHC de classes I e II, utilizando o software SYFPEITHI. Os mesmos foram sintetizados utilizando a metodologia da fase sólida conforme protocolo padrão. Para a purificação foi utilizado a cromatografia líquida sob condições de baixa pressão em coluna de fase reversa e gradiente de acetonitrila (5 a 95%). Os peptídeos tiveram sua identidade química confirmada por espectrometria de massas. O estudo foi aprovado pelo Comitê de Ética no Uso de Animais Nº 288/2013. Nove hamsters (Mesocricetus auratus) foram divididos em três grupos experimentais, contendo três animais. O grupo controle recebeu 100 μL de solução salina estéril 0,85%; o grupo adjuvante: 30 µL do adjuvante Montanide ISA-61VG em 70 µL de solução salina 0,85% e o grupo vacina: 30 µL do peptídeo MHC-I + 30 µL do peptídeo MHC-II, em 30 µL de Montanide, diluídos em 10 µL de solução salina 0,85%. A resposta imune humoral foi avaliada por Enzyme-Linked Immunosorbent Assay (ELISA), com diluição dos soros em 1:40, seguido recomendação do Ministério da Saúde, e com leitura em espectrofotômetro em 490nm. As análises estatísticas foram realizadas por ANOVA e consideradas significativas quando p<0,05. Resultados e Discussão: Os níveis de anticorpos igG, anti-Leishmania, foram avaliados no soro de hasmters, através do teste de ELISA. Com os resultados obtidos no ensaio, foi observado similaridade entre as médias obtidas do título de anticorpos Ig-G anti-Leishmania no grupo controle e no grupo apenas com o adjuvante, com valores de 0,028 e 0,035 respectivamente, o que pode ser justificado pela falta de estímulo devido à ausência do peptídeo na formulação vacinal. Por outro lado, no grupo dos animais inoculados com o peptídeo + adjuvante o valor da média obtida na titulação de anticorpos foi de 0,289, demonstrando que o grupo vacina estava estimulando a proliferação de plasmócitos, produtores de anticorpos. Entretanto, em leishmaniose, a imunidade celular é mais abrangente, contudo, anticorpos podem ter ação de aglutinação do parasito, além de sinalizar a entrada da Leishmania na célula, estimulando a formação do complexo 1 Engenheiro(a
